Tillage and weed control are critical components of cropping systems that need to be combined such that crops benefit from reduced competition. However, weeds may also contribute to the biological diversity within the agro‐environment. This greenhouse study investigated whether common weeds of arable cropping systems were suitable host plants for arbuscular mycorrhizal fungi (AMF), allowing the development of extraradical mycelium (ERM) that can contribute to the early colonization of a following wheat crop, especially in the absence of soil disturbance. Weeds were allowed to grow for up to 2 months before being controlled by soil disturbance or herbicide application (glyphosate or paraquat). Pregerminated wheat seeds were then planted. Chemical control of the weeds prior to sowing enhanced the early arbuscular mycorrhiza (AM) colonization rate of wheat roots, whereas mechanical disturbance was less acceptable as a method of weed control for rapid AM colonization. The type of herbicide (contact or systemic) had no impact on colonization of the wheat crop. Enhanced AM colonization promoted early P acquisition and growth of the crop. Appropriate management of weeds emerging between two consecutive cropping seasons coupled with no‐till soil management could ensure a quick and efficient AM colonization of the following wheat plants. 相似文献

Anaplasmosis, caused by Anaplasma marginale, results in significant economic losses of cattle in tropical and subtropical regions worldwide. Six major surface proteins (MSPs) were well characterized and designated as MSP1, MSP2, MSP3, MSP4, and MSP5. The objective of this study was to evaluate the humoral immune response of BALB/c mice against the recombinant MSPs, incorporated into immunostimulating complex (ISCOM). The recombinant proteins purified by Ni-NTA columns were incorporated into ISCOM and ISCOMATRIX by the lipid film hydration method. BALB/c mice immunized with ISCOM/rMSPs and ISCOMATRIX/rMSPs vaccines produced whole IgG, IgG1, and IgG2a, in contrast to the negative groups (PBS and ISCOMATRIX adjuvant). All groups that received antigen responded specifically against the rMSPs by Western blotting, showing the rMSP1a (60-105kDa), rMSP1b (100kDa), rMSP4 (47kDa), and rMSP5 (29kDa). Additional studies will have to be performed in cattle to evaluate the humoral and cellular mechanisms of this subunit vaccine and their possible use as protective vaccines against homologous and heterologous strains of A. marginale. 相似文献

The analytic-synthetic scheme makes use of haploidization and 2n gametes-mediated sexual polyploidization to incorporate genetic diversity from wild related species in potato breeding. Whereas 2n pollen can be directly screened by microscopy, 2n eggs are routinely screened indirectly by interploid crosses, a time-consuming and inaccurate technique. In five haploid tbr × wild diploid species families with variability for agronomic characters, 2n eggs were screened indirectly to incorporate this germplasm into a useful form and directly by microscopy to establish the feasibility of this approach. For the indirect screening, at least 15 flowers of each of 49 genotypes/family were crossed with a commercial cultivar. Eighty-five fruits were produced; only 15 of them had seeds (1–25). Chromosome numbers were determined in a random sample of five seeds/fruit, resulting diploid in two genotypic combinations and tetraploid in another two. For the direct screening, eight combinations of staining and destaining (times and concentrations) were evaluated in detached ovules to adjust a clarification technique. The best combination was one day of staining and one day of destaining in 0.5 % acetic acid. In an indirectly detected diplogynoid, the average diameters of the central cell nuclei (CCN) and nucleoli (CCNu) were, respectively, 9.3 μ and 3.4 μ. Gametophytes with diametres 1.26 times the average were considered 2n. By crosses, eleven seeds were obtained in one fruit of this clone whereas the 2n egg frequency determined by microscopy was 7%. Summary Artificial selection in autogamous species is investigated applying standard algebraic techniques to conventional self-pollinating breeding systems as well as to recurrent cross-pollinating ones for both one and two-locus models. More than two loci are considered by means of computer simulations. Allelic and non-allelic genotypic interactions as well as genes with a large effect are considered whenever possible. The overall picture shows a clear superiority of recurrent selection systems over self-pollinating breeding systems when negative linkage disequilibrium is either present or generated as is expected in actual breeding populations. When epistatic interaction enhances the selective advantage of new recombinants the superiority of recurrent selection is even greater. 相似文献

RAPD markers were used for the study of 19Albanian olive cultivars and two wild olives (oleasters). A total of 76polymorphic
bands (4.8 polymorphic markers per primer) out of 107 reproducible were obtained using 16 primers. The number of bands per
primer ranged from 4 to 10,whereas the number of polymorphic bands ranged from 1 to 9, corresponding to 71%of the total amplification
products. All the accessions could be identified by the combination of four primers: OPA-19;OPA-02; OPK-16 and OPP-19. The
dendrogram,based on Jaccard's index, included three major groups according to their origin: 1)most of the cultivars from the
area of Berat (South of Albania) 2) cultivars from the Centre and Centre-North of Albania and3) cultivars from the Centre
and North-West of Albania along with the oleaster from Elbasan. In order to evaluate the origin of Albanian cultivars they
were compared to those diffused in other countries like Greece, Italy and Turkey, due to geographical and historical affinity
among these countries, by using a one way AMOVA. Although most of the genetic diversity was attributable to differences among
cultivars within each country (91.47%) significantφ-values among countries(φst = 0.085; p < 0.001)suggested the existence of RAPD phenotypic differentiation. Significant φ-values in all pairs formed by Albania with
the other countries were observed. These results are consistent with the autochthonous origin of Albanian cultivars.

Summary We conducted a joint Ecuador/Colombia/United States wild potato (Solanum sect. Petota) germplasm collecting expedition in Ecuador from April 13–July 1, 1991. The goals of the expedition were to collect germplasm and study the species boundaries of all of the 25 Ecuadorian taxa accepted by current taxonomists. We made 126 collections of 24 of these 25 taxa, 113 as germplasm samples, 13 only as herbarium collections. We synonymize six of these 25 names (S. baezense Ochoa, S. cyanophyllum Correll, S. pichinchense Bitter & Sodiro, S. serratoris Ochoa, S. suffrutescens Correll as synonyms of S. andreanum Baker; S. correllii Ochoa as a synonym of S. regularifolium Correll). Four other names (S. chomatophilum f. angustifoliolum Correll, S. moscopanum Hawkes, S. solisii Hawkes, S. tundalomense Ochoa) could not be consistently distinguished from S. colombianum Dunal in the field. Genetic evaluation aims to identify genotypes with high empirical breeding values (EBVs) for selection as parents. In this study, 2157 potato genotypes were evaluated for tuber yield using 8 years of early‐stage trial data collected from a potato breeding programme. Using linear mixed models, spatial parameters to target greater control of localised spatial heterogeneity within trials were estimated and variance models to account for across‐trial genetic heterogeneity were tested. When spatial components improved model fit, correlations of errors were mostly small and negative for marketable tuber yield (MTY) and total tuber yield (TTY), suggesting the presence of interplot competition in some years. For the analysis of multi‐environment trials, a variance model with a simple correlation structure (with heterogeneous variances) was the most favourable variance structure fitted for TTY and PTY (per cent marketable yield). There was very little difference in model fit when comparing a factor analytic structure of order 2 (FA2) with either FA1 or simple correlation structures for MTY, indicating that simple variance models may be preferable for early‐stage genetic evaluation of potato yield. 相似文献

Ectomycorrhiza (EM) community structure was analyzed at one bare heap site (BHS), one heap site with organic cover (HS-OH) and one reference site (RS) in the former uranium mining area near Ronneburg (Thuringia, Germany). Twenty-three EM morphotypes were distinguished, and 14 of them were additionally characterized by polymerase chain reaction (PCR), restriction fragment length polymorphism (RFLP) and internal transcribed spacer (ITS) sequence analysis. Colonization of birch by the different morphotypes was quantified, and the EM diversity at the different sites was investigated. Compared to RS, total EM colonization was reduced by 6% (P=0.851) at HS-OH and by 58% (P<0.001) at BHS. Likewise, EM diversity was reduced by 16% (P=0.229) at HS-OH and 52% (P<0.001) at BHS. The Sørensen similarity between EM samples from RS was nearly independent from the sampling date, whereas at HS-OH and especially BHS, the Sørensen similarity decreased with increasing time between the samplings. All EM fungal species dominating at the two heap sites were also present at RS. Thus, fungi with high tolerance against uranium and other stress factors at the heap sites (e.g. heavy metals, nutrient limitation, drought) were selected among all EM fungi of the area.
