A study was carried out on the transmission parameters of the European stone fruit yellows phytoplasma by the vector Cacopsylla pruni. In the greenhouse, using groups of psyllids, the minimum acquisition period was 2–4 days, the minimum latent period 2–3 weeks and the minimum inoculation period 1–2 days. The vectors retained infectivity until their death. Under natural conditions retention of infectivity in C. pruni lasts through the winter and the following spring, when the overwintering insects reach the stone fruit trees, they are already infected and infective. The research shows that the vector C. pruni transmits the European stone fruit yellows phytoplasma in a persistent manner. 相似文献

The ability of microbes to counter the scientific and therapeutic advancements achieved during the second half of the twentieth century to provide effective disease treatments is currently a significant challenge for researchers in biology and medicine. The discovery of antibiotics, and the subsequent development of synthetic antimicrobial compounds, altered our therapeutic approach towards infectious diseases, and improved the quality and length of life for humans and other organisms. The current alarming rise in cases of antibiotic-resistance has forced biomedical researchers to explore new ways to recognize and/or produce new antimicrobials or to find other approaches for existing therapeutics. Aquatic organisms are known to be a source of compounds having the potential to play a role in fighting the battle against pathogenic microbes. In this connection, cnidarians occupy a pre-eminent role. Over the past few decades several studies have explored the antimicrobial/antibiotic properties of cnidarian extracts with the aim of isolating compounds possessing useful therapeutic features. An 8-year-old intact male mongrel dog with alopecia and weight loss was referred to the Veterinary Faculty of Naples. The dog had pale mucous membranes, enlarged prescapular lymph nodes, and splenomegaly. Laboratory abnormalities included anemia, thrombocytopenia, and hyperglobulinemia. Bone marrow aspirate smears contained numerous Leishmania amastigotes and an immunofluorescent antibody titer was strongly positive (1:1280) for leishmaniasis. The dog was treated with a combination of meglumine antimoniate and allopurinol for 60 days and showed clinical improvement. Two months after the end of treatment the dog was again referred because of relapse of leishmaniasis and the presence of a firm subcutaneous mass on the medial right thigh. Based on cytologic examination of fine needle aspirates of the mass, a diagnosis of large-cell lymphoma was made. Flow cytometry of tumor cells revealed gammadelta-T-cell lymphoma with a CD5+, CD3+, TCRgammadelta+, CD4-, CD8-, CD45RA+ immunophenotype. Using nested PCR, amastigotes were not detected in the neoplastic tissue. An association between leishmaniasis and hematopoietic tumors has been described rarely. gammadelta-T cells may be involved in the host response to this parasite, and prolonged antigenic stimulation and chronic immunosuppression (typical of leishmaniasis) play a crucial role in the etiopathogenesis of T-cell lymphoma. 相似文献

In the last decades, a large body of literature has grown to evaluate the impact of forest management on epiphytic lichens in boreal coniferous forests. However, information is still lacking on coniferous forests of the Alps. This study compares lichen diversity between spruce forest stands of four successional stages: (1) young, (2) intermediate, (3) mature forests managed for timber production with a rotation cycle of 120–180 years, and (4) old-growth protected forests. The emphasis was placed on the occurrence of nationally rare and calicioid species (lichens and fungi traditionally referred to as Caliciales, known to be indicative of forest age and continuity). For each forest successional stage, four plots were selected. In each plot, 7 spruce individuals were surveyed for epiphytic lichens according to a standardised sampling method. Species richness increased from young to mature stands, while no difference was detected between mature and old-growth stands. This pattern was also confirmed for rare and calicioid species which are, however, more frequent in old-growth stands. Differences in species composition were also found between the different forest successional stages. Mature and old-growth plots slightly overlap, indicating that to some extent comparable lichen assemblages could be found in these stands. A nested pattern of species assemblages was found, old-growth stands hosting most of the species which were also found in stands belonging to the previous forest successional stages. Our results support the hypothesis that the management regime applied to spruce forests of the Italian Alps renders mature stands managed for timber production somewhat similar to old-growth stands as lichen habitat. However, we found a higher complexity in old-growth forests, and many species of conservation concern clearly preferred old-growth stands. In this perspective, a further prolongation of the normal cycle it is likely to be a most favourable conservation-oriented management to be recommended at least within protected areas and Natura 2000 sites, where conservation purposes should receive a high priority. 相似文献

Despite the dangerousness of the dry wood-boring beetle Trichoferus holosericeus (Rossi) (=Hesperophanes cinereus (Villiers)) (Coleoptera: Cerambycidae), little is known about its biological and nutritional requirements. The aim of this
research was therefore to investigate into the dietary preferences of this insect, in order to determine if carbohydrates
can be considered as its main nutritional source, to clarify whether this cerambycid is specialised on hardwoods and finally
to select a suitable diet for laboratory cultures. Different substrates were tested and the decay grade, as well as the survival
of larvae, was recorded after 3 and 6 months. Chemical analyses were also carried out on excrements, residues of wood after
larvae excavation and sound wood. Results indicated that T. holosericeus fed mainly on glucose and peptone added to an artificial diet, while on natural wood it preferred structural carbohydrates
constituting the hemicelluloses and the non-crystalline cellulose. Furthermore, Scots pine sapwood had no toxic effect to
the beetle. In terms of number and dimensions of emerged larvae, the artificial diet resulted to be the most suitable substrate

Tomato is challenged by several pathogens which cause loss of production. One such pathogen is the oomycete Phytophthora infestans which is able to attack all the aerial parts of the plant. Although a wide range of resistance sources are available, genetic
control of this disease is not yet successful. Pyramiding R-genes through genetic transformation could be a straightforward
way to produce tomato and potato lines carrying durable resistance to P. infestans. In this work the R1 potato gene was transferred into tomato lines. The tomato transgenic lines were analyzed by using q-RT-PCR
and progeny segregation to determine the gene copy number. To test the hypothesis that R1 represents a specifically regulated R-gene, transgenic tomato plants were inoculated with P. infestans isolate 88133 and IPO. All the plants containing the R1 gene were resistant to the late blight isolate IPO-0 and susceptible to isolate 88133. These results provide evidence for
specific activation of the R1 gene during pathogen challenge. Furthermore, evidence for enhancement of PR-1 gene expression during P. infestans resistance response was obtained. 相似文献

BACKGROUND: Fenoxaprop‐P‐ethyl is a herbicide used on cereals and in particular on rice, the degradation of which leads to several relevant metabolites. The herbicide is used together with an agronomic safener such as isoxadifen‐ethyl, which also generates some metabolites. The present work was aimed at developing and validating an analytical method for the determination of the above parent compounds and their main metabolites in the edible fractions of rice. Parent compounds were extracted in acetonitrile and determined by gas chromatography with a mass spectrometer detector, while metabolites were extracted in acetonitrile and analysed by liquid chromatography tandem mass spectrometry. RESULTS: The method was validated through recovery tests in rice straw, grain and plant: accuracy was in the range 76–86% and 90–103% for parent compounds and metabolites respectively. Precision, as relative standard deviation, was in the range 3–11% and 6–17% for parent compounds and metabolites respectively. The limit of detection was 0.01 mg kg?1 for each analyte, while the limit of quantification was set at 0.05 mg kg?1. A defining factor for the commercial value of durum wheat pasta is its amber colour, which depends on the semolina yellow pigment concentration and on the oxidative enzymatic activity. Among carotenoids controlling yellow colour, the presence of β-carotene is also important as precursors of vitamin A. The aim of the present study was to detect quantitative trait loci (QTL) for yellow pigment concentration, yellow index and individual carotenoid compounds (lutein, zeaxanthin, β-cryptoxanthin, α-carotene and β-carotene) in a durum segregant population. Total carotenoid concentration amounted to 37% of the yellow pigments, indicating unknown colour-producing compounds in the durum extracts. Lutein was the most abundant carotenoid, followed by zeaxanthin, α-carotene and β-carotene, while β-cryptoxanthin was a minor component. Phytoene synthase marker Psy-A1, 150 SSR and EST-SSR markers, and 345 DArT® markers, were used to construct the linkage map for subsequent QTL analysis. Clusters of QTL for total and/or one or more carotenoid compounds were detected on the same chromosome regions (2A, 3B, 5A and 7A) where QTL for yellow pigment concentration and yellow index were identified. The molecular markers associated to major QTL would be useful for marker-assisted selection programs to facilitate high carotenoid concentration with high nutritional carotenoid compounds in wheat grain. 相似文献
