ABSTRACT: At birth, the immune system is still in development making neonates more susceptible to infections. The recognition of microbial ligands is a key step in the initiation of immune responses. It can be mimicked to stimulate the immune system by the use of synthetic ligands recognising pattern recognition receptors. In human and mouse, it has been found that neonatal cytokine responses to toll-like receptor (TLR) ligands differ in many ways from those of adults but the relevant studies have been limited to cord blood and spleen cells. In this study, we compared the responses in neonate and adult sheep to CpG oligodeoxynucleotides (ODN), a TLR9 ligand, in both a mucosal and a systemic organ. We observed that in response to CpG-ODN more IL-12 was produced by neonatal than adult sheep cells from mesenteric lymph nodes (MLN) and spleen. This higher IL-12 response was limited to the first 20 days after birth for MLN cells but persisted for a longer period for spleen cells. The major IL-12-producing cells were identified as CD14+CD11b+. These cells were poor producers of IL-12 in response to direct stimulation with CpG-ODN and required the cooperation of other MLN cells. The difference in response to CpG-ODN between neonates and adults can be attributed to both a higher proportion of CD14+CD11b+ cells in neonate lambs and their higher capacity to produce IL-15. Objective: To report frequency and type of complications, and outcome in dogs with severe neurologic signs secondary to internal, suspected obstructive hydrocephalus treated by ventriculoperitoneal (VP) shunting. Study Design: Case series. Animals: Dogs (n=14). Methods: Medical records (2001–2006) was reviewed for dogs that had VP shunting. Inclusion criteria were complete medical record, progressive forebrain signs unresponsive to medical treatment, normal metabolic profile, negative antibody titers and/or cerebrospinal PCR for Toxoplasma gondii, Neospora caninum, and canine distemper virus, magnetic resonance images of the brain, confirmed diagnosis of VP shunting, and follow‐up information. Results: Hydrocephalus was idiopathic in 5 dogs and acquired (interventricular tumors, intraventricular hemorrhage, inflammatory disease) in 9 dogs. Four dogs developed complications 1 week to 18 months postoperatively, including ventricular catheter migration, infection, shunt under‐drainage, kinking of the peritoneal catheter, valve fracture, and abdominal skin necrosis. Three of these dogs had 1 or more successful revision surgeries and 1 dog was successfully treated with antibiotics. All, but 1 dog, were discharged within 1 week of surgery, and had substantial neurologic improvement. Median survival time for all dogs was 320 days (1–2340 days), for dogs with idiopathic hydrocephalus, 274 (60–420) days and for dogs with secondary hydrocephalus, 365 (1–2340) days. Conclusions: VP shunting was successful in relieving neurologic signs in most dogs and postoperative complications occurred in 29%, but were resolved medically or surgically. 相似文献
Objective: To evaluate the clinical outcome of minimally invasive plate osteosynthesis (MIPO) for nonarticular tibial fractures stabilized using bone plates with or without an intramedullary rod (IMR). Study Design: Prospective study. Animals: Dogs (n=28) and 8 cats. Methods: After closed reduction, fracture fixation was achieved using an epiperiosteal plate inserted percutaneously through epiphyseal small incisions. In some fractures, an IMR was inserted via medial parapatellar arthrotomy. Radiographs were recommended every 3 weeks until clinical union. Postoperative tibial length and alignment were compared with contralateral measurements (P<.05). Time to clinical union and complications were recorded. Results: An IMR was used in 30.5% of the cases. Repaired tibiae were 1% shorter than contralateral tibiae (P<.05). Frontal and sagittal alignment were similar between repaired and contralateral tibiae (P>.05). Six dogs were lost for follow‐up; owners indicated normal function. In 30 cases for which bone healing was documented, mean±SD healing time was 45±20.8 days; however, when considering the 23 cases, which completed preestablished scheduled follow‐ups, healing time was 36±11.6 days. Minor complications occurred in 4 cases (11%). One major complication (3%) consisting of a plate fracture was successfully revised using MIPO with a larger plate. Conclusions: Consistent restoration of alignment was accomplished using MIPO techniques. Furthermore, MIPO appeared to yield faster healing times and lower complication rates than those reported with conventional plate osteosynthesis. 相似文献
Pectobacterium brasiliense (Pbr) infects a wide range of crops worldwide, causing potato blackleg and soft rot and vegetable soft rots. This study aimed to characterize the genetic diversity and virulence variability among 68 Pbr strains isolated from either symptomless potato progeny tubers, diseased potato plants, ware potatoes wash water, or vegetables grown in Israel, as well as strains isolated from symptomless seed tubers grown in Europe, or diseased potato plants grown in France. The collection was typed using PCR and TaqMan real-time PCR analyses, dnaX sequence analysis, pulsed-field gel electrophoresis (PFGE), and pectolytic activity. dnaX phylogeny grouped almost all strains in a common genetic clade related to Pbr, which was distinct from the other Pectobacterium species. PFGE analysis identified two main clusters, including one major group of 47 strains with 95%–100% similarity. Maceration assays on two potato cultivars showed significant differences between strains but with no correlations with the source of the strains nor the status of the host (with/without symptoms). Molecular (dnaX sequences and PFGE profiles) and phenotypic analyses (tuber maceration tests) showed that the tested Pbr strains are not a homogeneous group. Analysis of the tested Pbr strains isolated from potato and vegetables grown in fields with a history of potato cultivation suggests that seed tubers imported from Europe may be the main source for Pbr in Israel. To the best of our knowledge, this is the first study that describes biodiversity and population structure of P. brasiliense isolated from potato and vegetables under hot climate conditions. 相似文献

Arbuscular mycorrhizal fungi (AMF) establish a mutualistic symbiosis with several plants and play a key role in improving plant growth, tolerance to abiotic and biotic stresses as well as the soil structure. This work aimed at elucidating the AMF temperature stress modulating impact on four pearl millet lines plant growth and soil aggregation. Experimental trials were carried out in both greenhouse and growth chamber to determine the response of the four millet lines to inoculation with two AMF strains (Rhizophagus aggregatus and Funneliformis mosseae) under heat and non-stress conditions. We first investigated the mycorrhizal colonization (MC) and the mycorrhizal growth response (MGR) of millet lines in relation with their soil aggregation potential (root adhering soil/root biomass, MAS/RB) in the greenhouse. Secondly, the four millet lines were grown in two separated growth chambers and subjected to a day/night temperature of 32/28?°C as the control treatment and 37/32?°C as the temperature stress treatment. Plant growth, mycorrhization rate and several physiological, mycorrhizal and soil parameters were measured. Results showed that the mycorrhization rates of millet lines were low and not significantly different. Funneliformis mosseae (31.39%) showed higher root colonization than Rhizophagus aggregatus (22.79%) and control (9.79%). The temperature stress reduced the mycorrhizal colonization rate, shoot and root biomass, and the soil aggregation for all tested lines. L220 and L132 showed more MC rate and MGR than the other lines under control and high-temperature treatment. The MGR was significantly better under temperature stress conditions than in the control. Under the temperature stress conditions, inoculation with R. aggregatus and F. mosseae increased chlorophyll concentration, root dry weight and shoot dry weight as compared to non-inoculated plants. AMF inoculation, particularly with F. mosseae had a positive influence on the tolerance of millet lines to temperature stress. This study demonstrates that AMF play an important role in the response of these four millet lines to temperature stress. AMF is therefore an important component in the adaptation of crops to climatic variations in Sub-Saharan Africa.
Aged plants are more difficult to infect than young plantlets. This modification of susceptibility is described as mature plant resistance (MPR). For potato virus Y (PVY), MPR is known to lead to low infection rates of plants inoculated at the postflowering stage and a decrease in the number of infected daughter tubers. However, the impact of inoculation date on the capacity of PVY to accumulate in daughter tubers has not been studied so far. Field and greenhouse experiments were carried out to better understand PVY epidemiology and to help potato growers to evaluate consequences of early/late infections on the quality of their crops. In field trials, potato plants (cv. Bintje) were covered by insectproof nets from planting to harvest except for a 14-day period to expose plants to natural PVY infections. Under controlled conditions, potato plants were mechanically inoculated with PVY at different dates from preflowering stages (early inoculations) to postflowering stage (late inoculations). At harvest, daughter tubers were individually collected and analysed to define proportions and viral load of infected tubers according to the time between virus inoculation and harvest. Our results showed that although the age of plants at the time of inoculation can modify their susceptibility to PVY infection, in return, early and late PVY inoculations lead to similar rates of infected tubers at the plant scale and equivalent viral accumulation in infected tubers. All together, these data revealed that both early/late infections are high risks for the sanitary quality of potato tubers. 相似文献
Studies on stem profile of teak (Tectona grandis) hardly exist. This can be possibly attributed to the occurrence of forks that is prevalent on teak trees. Stem profile model was therefore developed for teak in West Africa, which took into account the occurrence of forks. Trees were destructively sampled from Moist Evergreen Forest (MEF), Moist Semi-deciduous Forest (MSDF), Dry Semi-deciduous Forest (DSDF) and Savannah ecological zones in both Ghana and Côte d’Ivoire for the study. A single set of parameters could be used explicitly to predict stem profile of any teak tree, irrespective of country, eco-climatic zone, edaphic variables, site index, tree age, stand density and tree competition. The model efficiency and root mean square error (in relative diameter) were calculated to be 0.97 and 0.053, respectively. The relative position of the forks along the stem bole, with respect to total tree height, was highly variable and did not follow any particular trend, except higher value for the first fork in the Moist Evergreen Forest zone. However, average tree height to the first fork decreased along the ecological gradient from MEF, MSDF and DSDF to Savannah. The extent of diameter reduction due to the forks could be predicted. Relative tree form for zero-forked, one-forked and two-forked trees was calculated to be 0.39, 0.36 and 0.33, respectively. Relative loss of stem volume due to one fork and two forks was estimated to be 6.5 and 13.9%, respectively. The measured stem volume when related to the corresponding predicted value yielded coefficient and intercept not significantly different from unity and zero, respectively, with an r2 value of 0.97. 相似文献

Two batches of 111 and 102 French boards were graded, respectively, of Spruce (Picea excelsa) and Douglas-fir (Pseudotsuga menziesii). For the visual grading the most discriminant criterion was noted: knots, cracks, wane, etc.

Members of the bacterial genus Azospirillum are root-associated bacteria that increase yield in cereals by promoting growth and alleviating drought stress. How plants integrate the many bacterium-derived growth-promoting stimuli with other environmental factors to generate a coordinated response remains unresolved. Using a commercial Azospirillum strain, A. lipoferum CRT1 and two host maize cultivars, it was observed that bacterization reduced the drought-induced increase in lateral root growth and enhanced the flood-induced increase in lateral root growth in the more drought- and flood-sensitive cultivar. In the other one, A. lipoferum CRT1 only elicited a moderate root growth response under low soil water potential. The photosynthetic potential and activity were increased in the earlier cultivar and decreased in the later one, irrespective of the soil water content. No impact of the bacterium was seen on the growth of the leaves of both cultivars under both stresses until the third leaf stage, therefore suggesting that it is a consequence of multiple primary adaptations to biotic and abiotic stresses. It is suggested that host–bacteria recognition leads to a stress-specific modulation of the root response and a differential stress-independent effect on photosynthesis. This is the first report of the impact of Azospirillum under flood conditions. 相似文献
