Individual phospholipid vesicles, 1 to 5 micrometers in diameter, containing a single reagent or a complete reaction system, were immobilized with an infrared laser optical trap or by adhesion to modified borosilicate glass surfaces. Chemical transformations were initiated either by electroporation or by electrofusion, in each case through application of a short (10-microsecond), intense (20 to 50 kilovolts per centimeter) electric pulse delivered across ultramicroelectrodes. Product formation was monitored by far-field laser fluorescence microscopy. The ultrasmall characteristic of this reaction volume led to rapid diffusional mixing that permits the study of fast chemical kinetics. This technique is also well suited for the study of reaction dynamics of biological molecules within lipid-enclosed nanoenvironments that mimic cell membranes.  相似文献

The chromosomal localization of the genes encoding the four subunits of muscle nicotinic receptor was determined by analyzing restriction fragment length polymorphisms between two mouse species Mus musculus domesticus (DBA/2) and Mus spretus (SPE). Analysis of the progeny of the interspecies mouse backcross (DBA/2 X SPE) X DBA/2 showed that the alpha-subunit gene cosegregates with the alpha-cardiac actin gene on chromosome 17, that the beta-subunit gene is located on chromosome 11, and that the gamma- and delta-subunit genes cosegregate and are located on chromosome 1.  相似文献

Boly et al. (Reports, 13 May 2011, p. 858) investigated cortical connectivity patterns in patients suffering from a disorder of consciousness, using electroencephalography in an auditory oddball paradigm. We point to several inconsistencies in their data, including a failure to replicate the classical mismatch negativity. The study of past forest change provides a necessary historical context for evaluating the outcome of human-induced climate change and biological invasions. Retrospective analyses based on fossil and genetic data greatly advance our understanding of tree colonization, adaptation, and extinction in response to past climatic change. For instance, these analyses reveal cryptic refugia near or north of continental ice sheets, leading to reevaluation of postglacial tree migration rates. Species extinctions appear to have occurred primarily during periods of high climatic variability. Transoceanic dispersal and colonization in the tropics were widespread at geological time scales, inconsistent with the idea that tropical forests are particularly resistant to biological invasions.  相似文献

This paper demonstrates that teamwork in science increasingly spans university boundaries, a dramatic shift in knowledge production that generalizes across virtually all fields of science, engineering, and social science. Moreover, elite universities play a dominant role in this shift. By examining 4.2 million papers published over three decades, we found that multi-university collaborations (i) are the fastest growing type of authorship structure, (ii) produce the highest-impact papers when they include a top-tier university, and (iii) are increasingly stratified by in-group university rank. Despite the rising frequency of research that crosses university boundaries, the intensification of social stratification in multi-university collaborations suggests a concentration of the production of scientific knowledge in fewer rather than more centers of high-impact science.  相似文献

Contribution of the Patagonia Icefields of South America to sea level rise   总被引:2,自引:0,他引:2  
Digital elevation models of the Northern and Southern Patagonia Icefields of South America generated from the 2000 Shuttle Radar Topography Mission were compared with earlier cartography to estimate the volume change of the largest 63 glaciers. During the period 1968/1975-2000, these glaciers lost ice at a rate equivalent to a sea level rise of 0.042 +/- 0.002 millimeters per year. In the more recent years 1995-2000, average ice thinning rates have more than doubled to an equivalent sea level rise of 0.105 +/- 0.011 millimeters per year. The glaciers are thinning more quickly than can be explained by warmer air temperatures and decreased precipitation, and their contribution to sea level per unit area is larger than that of Alaska glaciers.  相似文献

Phospholipase A(2)(PLA(2)) enzymes are considered the primary source of arachidonic acid for cyclooxygenase (COX)-mediated biosynthesis of prostaglandins. Here, we show that a distinct pathway exists in brain, where monoacylglycerol lipase (MAGL) hydrolyzes the endocannabinoid 2-arachidonoylglycerol to generate a major arachidonate precursor pool for neuroinflammatory prostaglandins. MAGL-disrupted animals show neuroprotection in a parkinsonian mouse model. These animals are spared the hemorrhaging caused by COX inhibitors in the gut, where prostaglandins are instead regulated by cytosolic PLA(2). These findings identify MAGL as a distinct metabolic node that couples endocannabinoid to prostaglandin signaling networks in the nervous system and suggest that inhibition of this enzyme may be a new and potentially safer way to suppress the proinflammatory cascades that underlie neurodegenerative disorders.  相似文献
