An immunohistochemical study of endocrine cells in the abomasum of vagotomized calf

The effect of thoraco-vagotomy on the distribution and frequency of chromogranin-, serotonin-, somatostatin- and gastrin-immunoreactive cells in the abomasum of the calf were investigated by immunohistochemistry. Calves were vagotomized at 1 week old and sampled 2 and 4 weeks later. The endocrine cells generally decreased in number in vagotomized calves as compared to non-operated control calves. However, the detailed responses of endocrine cells to vagotomy varied depending on the endocrine cell type, region of gastric mucosa, and period after vagotomy. The present result suggests that the vagus nerve has an influence on the intrinsic regulatory system by endocrine cell control in the ruminant abomasum.     

微量元素硒对成年皖西白鹅输卵管膨大部粘蛋白的影响

应用组织化学PA S-AB染色和定量图像分析法,研究了不同水平微量元素硒对成年皖西白鹅输卵管膨大部粘蛋白的影响,结果表明:日粮中添加0.2 m g/kg微量元素硒鹅输卵管膨大部粘蛋白与对照组(不加硒)差异显著(P<0.05);日粮中添加0.60 m g/kg与对照组差异极显著(P<0.01);当日粮中添加硒到1.1 m g/kg时,与对照组差异不显著(P>0.05);说明随着硒添加量的加大,皖西白鹅会发生慢性中毒。因此,微量元素硒对成年皖西白鹅输卵管膨大部粘蛋白的影响有显著差异。     

4味中药及其与抗菌药的复方制剂的MIC测定

用改进的试管两倍稀释法测定了黄连、鱼腥草、大青叶和苦参4味中药及其相互配伍（1：1）对标准大肠杆菌的最小抑菌浓度和4味中药与抗菌药配伍（1000：1）对临床分离鸡大肠杆菌的最小抑菌浓度。结果表明，4味中药及其相互配伍对大肠杆菌标准株有一定的抑制作用，其MIC值介于3．91—62．50mg／mL之间，其中大青叶和鱼腥草以1：l配伍对大肠杆菌抑菌效果最好，MIC为3．91mg／mL；对临床分离株，单方药中黄连抑菌效果最好，MIC为31．25mg／mL，24种复方药中，黄连和盐酸多西环素或加替沙星以1000：1配伍的抑菌效果最好，其MIC为3．91mg／mL。     

Prevention of isinglass on the chronic atrophic gastritis in rats and its mechanism

AIM: To evaluate the effect of isinglass on chronic atrophic gastritis(CAG) in rats and its mechanism. METHODS: An animal model of CAG in accordance with the previous experience of combined administration of 60% ethanol, 20 mmol/L sodium deoxycholate and 0.1% ammonia water was established in SD rats. Isinglass was used as preventive therapy while we were establishing CAG rat model. Finally all the rats were executed and pathologic changes of the gastric mucosa were studied by gross appearance and microscopy and serum epidermal growth factor (EFG) and growth hormone(GH) contents were tested. RESULTS: In each isinglass prevention group, inflammation grade of gastric antrum was less than that in model group (P<0.01) while the mean ratio of the thickness of gastric mucosal gland and muscularis mucosa (L1/L2), the number of gastric glands in 1 mm lengths of mucosal layer in longitudinal sections were much better than those in model group (P<0.01).They were very close to normal control group (P>0.05). The expression of proliferating cell nuclear antigen (PCNA) in gastric mucosa and serum EFG level were higher than those in model group (P<0.01, P<0.05), but serum GH content showed no different between isinglass prevention group and model group. CONCLUSION: Isinglass preventes the gastric mucosal atrophy in the CAG model. Its mechanism may be related to the effects of decreasing the gastric mucosal damage， promoting the cell proliferation and increasing of internal EFG secretion.     

Effect of rhEGF activating extracellular signal-regulated kinase on the expression of Bcl-2 and p53 protein in human amniotic cells

AIM: To explore the effect of recombinant human epidermal growth factor (rhEGF) on the ERK1/2 pathway and its safety in use when the proliferation of cultured FL cells (human amnion epithelial cells) were promoted by rhEGF.METHODS: FL cells were stimulated by rhEGF at various concentrations.The proliferation rate of FL cells was evaluated by MTT assay.Western blotting was used to detect phosphorylation of ERK1/2 (p-ERK1/2) and the change of Bcl-2 and P53 protein.RESULTS: The proliferation rate FL cells reached the maximum when the concentration of rhEGF was 10 μg/L (42.4％，P<0.01).Western blotting showed that the activity of p-ERK1/2 increased significantly when rhEGF ranged in 10 μg/L to 60 μg/L.Level of Bcl-2 was unchanged in each groups.Level of P53 decreased significantly when the concentration of rhEGF was 60 μg/L.CONCLUSION: It is safe when rhEGF presents in optimization concentration of 10 μg/L.The capability of apoptosis may be depressed when rhEGF is in larger dose.     

Immunohistochemical Colocalization of Serotonin, Substance P and Met-enkephalin-Arg6-Gly7-Leu8 in the Endocrine Cells of the Ruminant Duodenum

The duodenum of various ruminants was examined by immunohistochetnical staining for serotonin, substance P and Metenkephalin-Arg⁶-Gly⁷-Leu⁸ (MENK8). Serotonin- and substance P-immunoreactive endocrine cells were detected in samples from cow, calf, sheep, goat, and barbary sheep. MENK8-immunoreactive endocrine cells were detected exclusively in samples from cow and calf. Substance P and MENK8 immunoreactivity was found in serotonin-immunoreactive endocrine cells of cattle. Substance P- and MENK8-immunoreactive nervous elements were detected in all ruminants examined. The present results suggest that the expression and/or the mechanism that controls the expression of each peptide might differ between endocrine cells and nerve cells and might also depend on animal species.

HPLC法测定蒺参胶囊中人参皂苷Re的含量

目的建立蒺参胶囊中人参皂苷Re的含量测定方法。方法采用高效液相色谱法测定。结果人参皂苷Re在1.65～6.05μg之间与峰面积呈良好的线性关系(r=0.999 7),平均回收率为95.98%,RSD为1.35%(n=5)。结论方法简便、可靠、准确,可用于蒺参胶囊的质量控制。
