This study was performed to evaluate reproductive performance after non‐surgical embryo transfer (Ns‐ET) of 10–15 porcine expanded blastocysts (ExBs) that had been vitrified and warmed (V/W) using the micro volume air cooling (MVAC) method. The effect of asynchrony between the donor and recipient estrous cycle was investigated. Ns‐ET was conducted in recipients whose estrous cycle was asynchronous to that of donors by a delay of 2, 1, or 0 days. In the 2‐day and 1‐day groups, the similar farrowing rates (27.3% and 25.0%) and survival rates to term (13.9% and 15.7%) were obtained after Ns‐ET of V/W ExBs. None of the recipients in 0‐day group farrowed. Artificial insemination (AI) prior to Ns‐ET was then evaluated. Ten–15 V/W ExBs were transferred non‐surgically to 12 recipients whose estrous cycles were asynchronous to that of donors by a 2‐day delay. All of the recipients produced piglets, and all (100.0%) delivered piglets were derived from the transferred V/W ExBs. The survival rate of V/W ExBs to term was 25.2%. These results demonstrate that Ns‐ET of V/W ExBs using MVAC can facilitate piglet production, even if 10–15 embryos are transferred. Moreover, piglets were obtained stably when AI was performed prior to Ns‐ET.  相似文献

Embryo transfer (ET) of 20 porcine expanded blastocysts (ExBs) vitrified and warmed (VW) by open pulled straw (OPS) to a recipient allows stable piglet production. The efficiency of artificial insemination (AI) prior to ET of 10 VW ExBs for piglet production was investigated. For one trial, 10–15 VW ExBs from single donor were assigned, 10 were used for ET and the remains were assessed for their in vitro viability. In the non‐AI/ET group, 10 were transferred to each of five recipients. As AI/ET group, 10 were transferred to each of five recipients after AI. In AI/non‐ET group, only AI was performed to seven gilts. In the non‐AI/ET group, the pregnancy rate was 40%, but none of them farrowed. In the AI/ET group, all recipients produced piglets. Four (80.0%) delivered piglets from transferred VW ExBs. The survival rate of VW ExBs to term was 20.0% (10/50). In the AI/non‐ET group, six of the seven gilts farrowed. There was no difference in in vitro viability between the non‐AI/ET and AI/ ET groups (62.5% and 68.3%, respectively). AI prior to ET can be an appropriate way to maintain pregnancy and assist the development of a low number of VW ExBs to term.  相似文献

An aqueous ethanol extract of Bergenia crassifolia rhizomes strongly inhibited human pancreatic lipase activity and increased scavenging of DPPH free radicals in vitro. Chromatographic separation of this extract led to isolation of the hydrolysable tannins (+)-catechin 3,5-di-O-gallate () and (+)-catechin 3-O-gallate (). This is the first report of the isolation of compound 1 from plant material. This compound strongly inhibited human pancreatic lipase (with an IC50 value of 0.42 μg/ml) and exhibited a remarkable free radical-scavenging ability (with an SC50 value of 1.04 μg/ml). The chemical structures of 1 and 2 were elucidated using MS, NMR and chemical approaches.  相似文献

The objective of this study was to clarify the effect of ovarian status and follicular size on morphological normality and maturational ability of cat oocytes. Ovarian status was classified into inactive, follicular, luteal and prepubertal, and follicles were classified into three groups according to their diameter (400-800, 800-1200 and 1200-2000 μm). In each ovarian status, the number of follicles decreased but the percentage of morphologically normal oocytes increased with the growth of follicles (p<0.05). Only a single follicle that was 1200-2000 μm in diameter was observed in two of the five prepubertal cats. In follicles that were 800-1200 μm in diameter, the percentage of normal oocytes and maturation rate were higher in prepubertal cats than in mature cats (p<0.05). Oocyte diameter tended to increase with the growth of follicles. After oocytes were cultured individually in droplets of maturation medium, the oocyte maturation rate increased with the growth of follicles in each ovarian status (p<0.05). In conclusion, oocytes collected from larger follicles possess higher maturational ability in vitro in sexually mature cats. In prepubertal cats, a higher maturation rate can be obtained from oocytes derived from small follicles compared with in mature cats.  相似文献

It has been considered by investigators, no matter whether they stand by the lignin-protein complex theory or not, that lignin plays an important role in the humus formation of plant residues. When plant residues are added to the soil, though almost all the plant constituents are decomposed, only lignin accumulates in the soil as a component of humus, as it is resistant to the microbial decomposition. However, lignin does not remain unchanged in this case and is thought to be modified, either through chemical agencies or as a result of specific microbial action. Though it is not yet fully clarified what changes it undergoes in the humification process, at least it is indicated that the methoxyl, as a characteristic radical of lignin, is attacked during the process.  相似文献

Uncertainties in the rate of biomass variation with forest ageing in tropical secondary forests, particularly in belowground components, limit the accuracy of carbon pool estimates in tropical regions. We monitored changes in above- and belowground biomass, leaf area index (LAI), and biomass allocation to the leaf component to determine the variation in carbon accumulation rate with forest age after shifting cultivation in Sarawak, Malaysia. Nine plots in a 4-year-old forest and fourteen plots in a 10-year-old forest were monitored for 5 and 7 years, respectively. Forest and plant part biomass were calculated from an allometric equation obtained from the same forest stands. Both above- and belowground biomass increased rapidly during the initial decade after abandonment. In contrast, a much slower rate of biomass accumulation was observed after the initial decade. LAI also increased by approximately double from the 4-year-old to 10-year-old forest, and then gently increased to the 17-year-old forest. We also found that allocation variation in leaf biomass and nitrogen was closely related to the rate of biomass accumulation as a forest aged. During the first decade after abandonment, a high biomass and nitrogen allocation to the leaf component may have allowed for a high rate of biomass accumulation. However, reduction in those allocations to leaf component after the initial decade may have helped to suppress the biomass accumulation rate in older secondary forests. Roots accounted for 14.0–16.1% of total biomass in the 4–17-year-old abandoned secondary forests. We also verified the model predicted values for belowground biomass by Cairns et al. (1997) and Mokany et al. (2006), although both models overestimated the values throughout our data sets by 45–50% in the 10-year-old forest. The low root:shoot ratio in the secondary forests may have caused this overestimation. Therefore, our results suggest that we should modify the models to estimate belowground biomass considering the low root:shoot ratio in tropical secondary forests.  相似文献

Variations in leaf photosynthetic, morphological and biochemical properties with increasing plant height from seedlings to emergent trees were investigated in five dipterocarp species in a Malaysian tropical rain forest. Canopy openness increased significantly with tree height. Photosynthetic properties, such as photosynthetic capacity at light saturation, light compensation point, maximum rate of carboxylation and maximum rate of photosynthetic electron transport, all increased significantly with tree height. Leaf morphological and biochemical traits, such as leaf mass per area, palisade layer thickness, nitrogen concentration per unit area, chlorophyll concentration per unit dry mass and chlorophyll to nitrogen ratio, also changed significantly with tree height. Leaf properties had simple and significant relationships with tree height, with few intra- and interspecies differences. Our results therefore suggest that the photosynthetic capacity of dipterocarp trees depends on tree height, and that the trees adapt to the light environment by adjusting their leaf morphological and biochemical properties. These results should aid in developing models that can accurately estimate carbon dioxide flux and biomass production in tropical rain forests.  相似文献

ABSTRACT: Early growth, yolk and oil globule absorption, early morphological development and initial feeding were studied in the Black Sea turbot Psetta maxima . Based on energy transition and morphological development, the nutritional transition process from endogenous to exogenous sources was divided into six phases: (i) primordial phase (from hatching to ≈ 30 h after hatching, HAH); (ii) organogenesis phase (to ≈ 90 HAH); (iii) onset of feeding (to ≈ 110 HAH); (iv) early feeding (to ≈ 190 HAH); (v) intensified feeding (to ≈ 230 HAH); and (vi) completion of oil globule absorption (to beyond 345 HAH). Based on comparisons with early life stage features of other marine species, the turbot larvae were shown to possess the following characteristics: (i) oil globule remaining for a long period, resulting in an extended mixed feeding period; and (ii) feeding rate extremely high in larvae immediately following final absorption of the oil globule. These features in early turbot larvae were regarded as advantageous for survival in a rearing condition, although the coincidence of initial feeding with completion of yolk absorption signaled a period of leveled-off growth rate.  相似文献

An aqueous ethanol extract of Pyrola rotundifolia L. induced AMP-activated protein kinase (AMPK) phosphorylation in C2C12 myotubes. The bioassay-guided fractionation of the extract led to the isolation a 2-methyl-7-hydroxymethyl-1,4-naphthoquinone, or a 7′-hydroxy-chimaphilin, which showed concentration-dependent AMPK phosphorylation activity at 2.5–20 μg/ml. At a concentration of 10 μg/ml (50 μM), an approximately four-fold increase in the AMPKα(Thr172) phosphorylation level was observed. The stimulatory effect of naphthoquinone on AMPK activity was comparable to that of known compounds found in natural sources that activate the AMPK signaling pathway.  相似文献
