中国巴马小型猪内源性反转录病毒的检测

目的 :对我国特有巴马小型猪内源性反转录病毒 ( porcine endogenous retrovirus,PERV)的存在与 m RNA的表达情况进行检测 ,了解巴马小型猪内源性反转录病毒的携带情况。方法 :根据以往建立的 PCR、RT-PCR检测方法 ,对来自于巴马小型猪外周血淋巴细胞的 DNA和RNA样品进行 PERV核心蛋白基因 ( gag)、多聚酶基因 ( pol)及囊膜基因 ( env)的存在与表达进行检测 ;同时 ,根据目前通用的 env基因分型方法检测 PERV env-A、env-B、env-C的存在与表达。结果 :在 1 2个被检的 DNA样品中均检出了PERV特异性 DNA的存在 ;同样 ,在 1 2个被检的 RNA样品中均有 PERV特异性 RNA的表达 ,且所表达的 PERV均为 A型和 B型 ;其中有9个 DNA样品检测出 PERV-C型的存在 ,所有样品中均未检出 C型 PERV的表达。结论 :检测结果表明 1 2个被检巴马小型猪基因组中存在着内源性反转录病毒序列 ,且能以 m RNA的形式表达 ,这一结果为我国特有小型猪的开发、利用及其病毒安全性评价奠定了基础。     

Verification by polymerase chain reaction of vertical transmission of Theileria sergenti in cows

To evaluate the transplacental transfer of Theileria sergenti infection in cattle, we used DNA probes to detect T. sergenti in 6 pregnant cows and their calves. All the animals were monitored by parasitologic, serologic, and polymerase chain reaction (PCR) assays for a predicted 875-base-pair (bp) DNA product and a 684-bp amplicon detected by nested PCR in the blood and spleens of aborted fetuses. An open reading frame (ORF) starting at nucleotide 170 and terminating at position 1021 was shown to code for a polypeptide of 283 amino acid residues. All 6 dams and 5 calves were positive for T. sergenti in all tests. One calf was positive only with nested PCR. We conclude that transplacental transmission of T. sergenti is a significant problem. The relevance of the data in the programmed introduction of new (especially pregnant) animals into established clean herds needs serious consideration with regard to control of theileriosis and other tickborne diseases.     

The use of a Giardia vaccine as an immunotherapeutic agent in dogs.

Dogs (n = 13), which had failed to be cured of giardiosis following chemotherapeutic measures, were treated with a Giardia vaccine (2-3 injections). Clinical signs resolved between 16 and 42 days postvaccination and cessation of fecal cyst shedding was between 21 and 70 days. Vaccination is a potential method of treating giardiosis in dogs.

多层喷射沉积制备耐热铝合金管坯热流分析(Ⅱ) --沉积过程热流分析

通过建立沉积阶段热流分析模型，对多层喷射沉积制备耐热铝合金管坯过程中喷射流沉积后的凝固和冷却进行了分析．喷射流沉积后与基体／沉积坯的传导热交换占热输出率的90％以上，在沉积过程中，沉积面的温度变化呈周期性的升降．计算结果表明，多层喷射沉积采用移动坩埚式扫描方式，延长了喷射流沉积时间间隔，喷射流沉积后可以获得10^3K／s以上的冷却速度。     

The RNA interference

The phenomenon of RNA interference (RNAi) is highly conserved mechanism in the organism evolution. As a immune system, RNAi is a ubiquitous mechanism against invading microorganism in plant and animal cells. Recently, it has been found that RNAi is the process by which double-strand RNA(dsRNA) directs sequence-specific degradation of messenger RNA and the mediations of sequence specific messenger RNA degradation are 21-and 23-nucleotide small interfering RNAs that generate by ribonuclease from endogenous longer dsRNA or by transfectious technics from heterologous dsRNA. Over the past few years, the way in which cells respond to dsRNA by silencing homologous genes has revealed a new regulating paradigm in biology.

从上海光明乳业股份有限公司车间发酵罐中分离出4株酸奶噬菌体。1号噬菌体头部呈六角形，平均直径为95.1nm，头部剖面平均面积为8184．8nm^2，尾部的平均直径为18．3nm，平均长度为403．4nm；2号噬菌体头部呈六角形，平均直径为85．4nm，头部剖面平均面积为8046．2nm^2，尾部的平均直径为23．3nm，平均长度为549．4nm；3号噬菌体头部呈六角形，平均直径为95．5nm，头部剖面平均面积为9792．3nm^2，尾部的平均直径为19．8nm，平均长度为450．5nm；4号噬菌体头部呈六角形，平均直径为119．3nm．头部剖面平均面积为12586．1nm^2，尾部的平均直径为23．6nm，平均长度为520．5nm。
