Quercus serrata Thunb. ex Murray is a widespread deciduous oak in China, the Korean Peninsula, and Japan, and a strong isoprene emitter. Establishing accurate inventories of this species and estimating net carbon budgets, including biogenic volatile organic compounds (BVOC), necessitates detailed evaluation of BVOC emission and oxidation characteristics. Emissions of isoprene, the most abundant BVOC, presumably contribute to atmospheric chemistry through the formation of photochemical oxidants and secondary organic aerosols. We built an isoprene flux monitoring system to simultaneously reveal characteristics of the flux and fate of isoprene at multiple locations in Q. serrata forests. We used proton transfer reaction mass spectrometry (PTR-MS) and an automated closed chamber to measure isoprene emissions from soil and leaves in a warm-temperate Q. serrata forest. We used a relaxed eddy accumulation system with PTR-MS to simultaneously measure the canopy flux. In continuous foliage chamber measurements, we observed daily variations of isoprene emissions and continuous nocturnal emissions from leaves. Nocturnal emissions exceeded 25 % of total daily leaf emissions and were relatively high at sunset and low at sunrise. These results suggest that nocturnal emissions from mature trees may not be negligible. When leaf emissions were high in the daytime, the canopy isoprene flux tended to plateau at an upper limit. Observations of isoprene concentrations and gradients suggest that the plateau was caused by acceleration of isoprene oxidation, and sequential formation of secondary organic aerosols may occur near the leaf just after emission. Elucidation of these linkages may require continuous field measurements with a simultaneous multi-flux monitoring system. 相似文献

The effect of addition of Acacia mangium bark powder on the thermosetting processes of two commercial phenol resins, PF-A and PF-B, was examined by bond strength
test, torsional braid analysis, and differential scanning calorimetry. When the bark powder was added to PF-A, the bond strength
of plywood pressed at 110°C increased and was comparable to that of plywood pressed at 120 and 130°C. However, when the bark
powder was added to PF-B, the bond strength of plywood pressed at 110°C was still lower than that of plywood pressed at 120
and 130°C. The relative rigidity and loss tangent of PF-A cured with the bark powder obtained by heating at 100°C were comparable

Heterotrophic respiration strongly influences carbon cycles at the ecosystem and global scales. We used an automated chamber system to measure the heterotrophic respiration of coarse woody debris (CWD) and leaf litter in a secondary broadleaved forest in southern Kyoto Prefecture. This system, which targeted only organic matter, could detect heterotrophic respiration responses to changes in environmental factors, especially rainfall events. The temporal trends and responses of respiration to environmental factors differed dramatically between CWD and leaf litter. CWD respiration showed a clear diurnal change corresponding to changes in CWD temperature and a clear decrease during rainfall events. Leaf litter respiration did not change clearly but increased at the beginning of rain events and returned to pre-rain rates when soil water content declined. The temporal patterns of the residuals between the observed respiration and baseline respiration, developed from the temperature?Cresponse curves under pre-rain conditions, differed between CWD and leaf litter respiration. The typical trend in CWD respiration response to rainfall events was a clear decrease and then gradual increase in the residuals after the event. The response of leaf litter respiration to wetting was an increase in the residuals during rainfall events and then a gradual decrease during drying. The difference in the responses of these respirations to wetting and drying processes are likely caused by differences in the physical characteristics of the CWD and the leaf litter layer. Measurements targeting only organic matter using an automated chamber system could detect the responses of heterotrophic respiration to environmental factors. 相似文献

Collection efficacy and in vitro embryo developmental ability of oocytes obtained from Duroc‐breed ovary donors at different stages of the estrous cycle (days 6, 12 and 16 after estrus) were performed. The numbers of collected oocytes did not differ significantly among the different estrous cycle groups (total 72–90 oocytes per gilt). However, the blastocyst rates of oocytes collected on days 12 and 16 (9.2% and 19.4%, respectively) were significantly higher than those on day 6 (1.1%). More oocytes were obtained on day 16 from small follicles (<2 mm in diameter; 85.3 oocytes per gilt) than from medium‐sized (≥2–<6 mm) and large (≥6 mm) follicles (17.5 and 12.8 oocytes, respectively). The blastocyst rates in both the medium‐sized and large follicle groups (20.0% and 19.2%, respectively) were significantly higher than that in the small follicle group (6.3%). The blastocyst cell numbers in both the medium‐sized and large follicle groups (39.4 and 43.3 cells, respectively) were significantly higher than that in the small follicle group (30.6 cells). The results suggest that oocyte collection from cycling Duroc pigs can be carried out efficiently from the late luteal to follicular stage. Those oocytes collected from medium‐sized and large follicles show better embryo development. 相似文献

We investigated the production of inhibin in boars from the infantile to pubertal periods by: (1) measurement of testicular and circulating levels of inhibin, (2) characterization of inhibin forms and (3) localization of inhibin subunits in the testis. Total inhibin levels in the testis increased until 8 weeks of age but then declined to much lower values at 15 weeks. Testicular inhibin A and inhibin B were high until 8 weeks. Circulating levels of total inhibin and inhibin A were also high until 8 weeks, then declined from 10 weeks; inhibin B was not detected, because of low sensitivity of the inhibin B assay. Analyses of inhibin A and inhibin B levels in the eluted fractions obtained from testes after immunoaffinity chromatography and SDS-PAGE showed the presence of a peak of approximately 45 kDa until 10 weeks of age. As the boars aged, the levels of inhibin A and inhibin B increased in the molecular weight region of 29–31 kDa. The fractions corresponding to 29 and 30 kDa suppressed FSH release from rat pituitary cells, but the 45 kDa fraction had no FSH-suppressing activity. Total amounts of inhibin A isolated from the SDS gels were similar to those of inhibin B until 10 weeks of age, but were three times higher than those of inhibin B between 15 and 25 weeks. Further fractionation by reverse phase high-performance liquid chromatography revealed that the 29–31 kDa immunoreactive material was composed of mature forms of inhibin A and inhibin B, in addition to a 26 kDa monomer. Immunohistochemistry indicated that positive immunostaining for the subunits was observed in Sertoli cells from the infantile to pubertal periods. Elongated spermatids also showed positive signals at age 25 weeks. These results clearly indicated that: (1) the boar testis has the ability to produce inhibin A and inhibin B during the infantile period but inhibin A is the predominant form towards puberty and (2) the molecular weight forms of inhibin and the sites of production of inhibin change with testicular development. 相似文献

The ultimate goal of this study was to establish an in vitro system to produce sperms. To pursue this goal, immature porcine testicular cells were cultured in stereostructural form and cultured testicular cord was investigated morphologically. At 4 weeks of age, the seminiferous tubules of the porcine testes consisted of undifferentiated germ cells (gonocytes and undifferentiated spermatogonia) and immature Sertoli cells. The interstitial tissue was largely occupied by Leydig cells. The testes were enzymatically digested, and the dispersed cells were encapsulated with alginate either immediately or after freeze-thawing. The resulting testicular cell cords were cultured for up to 10 weeks. After 2 weeks of culture, Sertoli cells, which were identified by their inhibin-positive reaction in immunohistochemistry, and Leydig cells, which were identified by their morphological characteristics, were observed in the cords. Neither undifferentiated nor differentiated types of germ cells were detected. The number of cells in the cords progressively decreased during the culture period. In order to discover the fate of the Sertoli cells, the level of inhibin in the spent media was determined. Inhibin in the media was at a detectable level after 2 days of culture. The levels increased and peaked at 2 weeks. When frozen-thawed testicular cells were applied to the culture, the peak level was maintained for over 8 weeks, in contrast to the gradual decrease of inhibin level when fresh cells were cultured. These results indicate that the culture conditions can sustain the survival of Sertoli cells. Further improvement is required for proliferation and differentiation of germ cells. 相似文献

Male pronucleus (MPN) formation is a very important physiological event during fertilization, which affects in vitro production of transferrable embryos. The aim of this study was to find out the correlation between the number of penetrated sperm and the occurrence of failure of MPN formation in porcine oocytes. In vitro matured porcine oocytes were fertilized in vitro with frozen epididymal sperm. Two different frozen sperm lots were tested in this study, which were different in terms of polyspermy rates. The numbers and the status of penetrated sperm in oocytes were evaluated 10 h after insemination. Under high polyspermy condition, the polyspermy rate was 83.5% with an average mean of 3.5 sperms per penetrated oocyte, whereas the percentage of polyspermy was 65.5% with an average mean of 2.4 sperms per penetrated oocyte under moderate polyspermic condition. Correlation analysis revealed a negative correlation between the number of penetrated sperm and their MPN formation percentage both in the sperm lot of high polyspermy (R = −0.560, p < 0.05) and in the sperm lot of moderate polyspermy (R = −0.405, p < 0.05) which suggests that penetration of excessive spermatozoa disables the oocyte cytoplasm to promote MPN formation. 相似文献
