Leaf litter respiration (R LL) was directly measured in situ to evaluate relationships with the water content in leaf litter (WC), which is distributed heterogeneously under natural conditions. To do so, we developed a small, closed static chamber system using an infrared gas analyzer, which can measure instantaneous R LL. This study focuses on the measurement of CO2 effluxes from leaf litter using the chamber system in the field and examines the relationship between R LL and WC among seven broadleaf species in a temperate forest. The measurements focused on the position of leaves within the litter layer, finding that both R LL and WC were significantly higher in the lower layer. The value of R LL increased with increasing WC, and the response of R LL to WC was similar among all seven species. Moreover, the temporal variation in WC differed among three species and was associated with leaf litter thickness. The observed heterogeneity in WC induced by the physical environment (e.g., position and thickness of leaf litter) affects the variation in WC and, therefore, both R LL and the decomposition rates of organic matter in the litter layer.  相似文献

We investigated testicular and circulating levels of dimeric inhibins in Holstein bulls from the infantile to postpubertal periods (5 to 50 weeks of age) and examined the relationship between the profiles of circulating dimeric inhibins and FSH. Concentrations of total inhibin and inhibin B in the testis were highest at 4 to 5 weeks of age but decreased gradually as the bulls aged. Testicular inhibin A levels showed a gradual decline to a nadir at 15 to 26 weeks of age, but by 39 weeks, they were high again. The contents of total inhibin, inhibin A, and inhibin B per testis generally increased with age. Fractionation of testicular homogenates obtained from 15-week-old bulls by a combination of immunoaffinity chromatography and SDS-PAGE confirmed the presence of two major molecular weight forms (32 and 45 kDa) of dimeric inhibins in the testes. Circulating levels of total inhibin and inhibin A showed a significant increase in bulls at around 10 to 14 weeks of age compared to the levels between 5 and 7 weeks of age but decreased thereafter. However, immunoreactivity for inhibin B was not detected in the peripheral circulation, probably because of low sensitivity of the inhibin B assays. The concentrations of plasma FSH were high at 5 weeks of age but declined to lower levels between 11 and 40 weeks, and then increased from 41 weeks onward. There was no significant correlation between the plasma levels of FSH and inhibin A or total inhibin. The results clearly indicate that the bull testis produces inhibin A and B and secretes at least inhibin A into the circulation during postnatal development. However, the profile of circulating FSH in bulls shows no reciprocal relationship with the inhibin A or total inhibin profile during the postnatal period.  相似文献

The effect of addition of Acacia mangium bark powder on the thermosetting processes of two commercial phenol resins, PF-A and PF-B, was examined by bond strength test, torsional braid analysis, and differential scanning calorimetry. When the bark powder was added to PF-A, the bond strength of plywood pressed at 110°C increased and was comparable to that of plywood pressed at 120 and 130°C. However, when the bark powder was added to PF-B, the bond strength of plywood pressed at 110°C was still lower than that of plywood pressed at 120 and 130°C. The relative rigidity and loss tangent of PF-A cured with the bark powder obtained by heating at 100°C were comparable to those at 120 and 140°C, and the reaction enthalpy was increased by bark powder addition. In contrast, chemical reactions for cured PF-B were not enhanced by bark powder addition.  相似文献

The polymerase chain reaction (PCR) is a rapid, precise method for detecting and identifying pathogenic bacteria. In addition to the published primers for identification of Agrobacterium tumefaciens up to species level, two sets of primers were designed to identify the nopaline and octopine types of Agrobacterium tumefaciens. The RBF-RBR primer set designed based on the nopaline type T-DNA right border detected the nopaline type A208 and R225f strains, and the ocsF-ocsR primer set derived from the ocs gene of the octopine type A. tumefaciens detected the octopine type A348 strain. After polymerase Chain reaction (PCR) amplification by the RBF-RBR primers, the A208 and R225f strains could be differentiated from each other by restriction fragment length polymorphism digestion using the restriction enzymes DraI and XbaI. Multiple colonies can be screened at one time in a single PCR tube with satisfactory efficiency, thereby allowing rapid detection of pathogenic A. tumefaciens. Following a rough screening by classical biovar medium and -methyl-d-glucoside medium, the developed PCR system was introduced to identify isolates collected from soil and crown gall samples. Of 42 isolates determined to be A. tumefaciens, 7 were found to be octopine type; all the rest were R225f type.  相似文献

SUMMARY: The effects of nutrient salts eluted from artificial algal reef blocks made with sea-bottom sediment (hedoro) and cement to the growth and survival rate of the gametophytes and juveniles of Eisenia bicyclis were investigated. When each sample block was submerged in filtered seawater for 28 days, the nutrient salt content from the hedoro increased with time and, in particular, the amounts of NO3− N and NO2− N reached a constant level on day 10. We observed that the lower the cement ratio, the higher the concentrations of eluted NH4+− N and PO43−− P. The growth of gametophytes as well as juveniles was highest in the elution of the block with the lowest cement ratio of 30%. In contrast, the survival rate of gametophytes and juveniles decreased linearly with time and the survival rate was higher for the hedoro blocks than for the block made of mortar.  相似文献

ABSTRACT: Poly(α- L -guluronate)lyase, as one of alginate lyases, was purified from the culture medium of a marine bacterium, Pseudomonas sp. strain F6, to an electrophoretically homogeneous state. The enzyme was shown to have a molecular mass of 36 kDa by sodium dodecylsulfate–polyacrylamide gel electrophoresis (SDS-PAGE), and was most active at around pH 7.5 and was stable between pH 6.5 and pH 8.5. In the thermal stability experiments, the enzyme's activity diminished through an intermediate state with increasing incubation temperatures and was finally lost when heated at 100°C for 15 min. The addition of hen egg-white lysozyme to the enzyme decreased thermal stability dramatically. The apparent retention of enzyme activity (approximately 50%) was observed after the addition of 6 M guanidine hydrochloride and 8 M urea. Enzyme activity was lost completely with 10 mMSDS, while the ordered structure, which is considered likely to be β-structure, was markedly created. The similar conformational feature has also been created in marine bacterial and mollusc enzymes and the β-structure is commonly observed in polyuronate lyases. The divalent cation (Ca2+) promoted the activity of the calcium chelator-treated enzyme significantly, suggesting that Ca2+ is involved in the formation of the active intermediate between the acidic uronate(s) and amino acid side-chain(s) of the enzyme.  相似文献

ABSTRACT

The influence of the long-term combination of rice straw removal and rice straw compost application on methane (CH4) and nitrous oxide (N2O) emissions and soil carbon accumulation in rice paddy fields was clarified. In each of the initial and continuous application fields (3 and 39?51 years, respectively), three plots with different applications of organic matter were established, namely, rice straw application (RS), rice straw compost application (SC) and no application (NA) plots, and soil carbon storage (0?15 cm), rice grain yield and CH4 and N2O fluxes were measured for three years. The soil carbon sequestration rate by the organic matter application was higher in the SC plot than in the RS plot for both the initial and continuous application fields, and it was lower in the continuous application field than in the initial application field. The rice grain yield in the SC plot was significantly higher than those in the other plots in both the initial and continuous application fields. Cumulative CH4 emissions followed the order of the NA plot < the SC plot < the RS plot for both the initial and continuous application fields. The effect of the organic matter application on the N2O emissions was not clear. In both the initial and continuous application fields, the increase in CH4 emission by the rice straw application exceeded the soil carbon sequestration rate, and the change in the net greenhouse gas (GHG) balance calculated by the difference between them was a positive, indicating a net increase in the GHG emissions. However, the change in the GHG balance by the rice straw compost application showed negative (mitigating GHG emissions) for the initial application field, whereas it showed positive for the continuous application field. Although the mitigation effect on the GHG emissions by the combination of the rice straw removal and rice straw compost application was reduced by 21% after 39 years long-term application, it is suggested that the combination treatment is a sustainable management that can mitigate GHG emissions and improve crop productivity.  相似文献

Eight fatty acid esters of triterpene alcohols (1-8), four free triterpene alcohols (9, 12, 17, and 18), four diterpene acids (19-22), two tocopherol-related compounds (23 and 24), four estolides (25-28), three syn-alkane-4,6-diols (29-31), one 1,3-dioxoalkanoic acid (32), and one aliphatic ketone (33), along with the mixture of free fatty acids, were isolated from the diethyl ether extract of the pollen grains of sunflower (Helianthus annuus). Among these compounds, 14 (2-8, 12, 23, 25-28, and 33) were new naturally occurring compounds, and their structures were determined on the basis of spectroscopic methods. Twenty-four terpenoids and lipids (1-4, 6-9, 12, and 19-33) and six free triterpene triols (10, 11, and 13-16), derived from their fatty acid esters (2, 3, and 5-8) by alkaline hydrolysis, were evaluated with respect to their inhibitory effects on the induction of Epstein-Barr virus early antigen (EBV-EA) induced by the tumor promoter, 12-O-tetradecanoylphorbol-13-acetate (TPA), in Raji cells, which is known to be a primary screening test for antitumor promoters. Among the 30 compounds tested, 21 compounds possessing a di- or a polycyclic ring system in the molecule (1-4, 6-16, and 19-24) showed potent inhibitory effects on EBV-EA induction (91-100% inhibition at 1 x 10(3) mol ratio/TPA).  相似文献

Fog/cloud and rain water were collected at the mountainside of Hachimantai range in northern Japan and rain water was also collected at Akita City in order to investigate the air pollutant scavenging mechanism. The concentrations of various ions in these samples were analyzed, and the fog drop size and the wind direction were measured at each fog event. The fog at Hachimantai range had a very high total ion concentration, and was considerably acidified by non sea salt (nss-) SO4 2? and NO3 ?, compared with the rain at Akita and all sites in Hachimantai range. Using the oblique rotational factor analysis, three factors were extracted as the air pollutants; A: (NH4)2SO4+H2SO4, B: sea salts+HNO3+H2SO4, C: NH4NO3+OH?. These salts are well-known as the cloud condensation nuclei (CCN). Combining the factor analysis with the 72h back trajectory at 850hPa level, the contribution of Factor A was closely connected to the long-range transportation of anthropogenic or natural aerosol in air masses of continental origin.  相似文献

We investigated the vertical profile of element distributions in the soil column on a granite substrate at three sites on the island of Yakushima, a world natural heritage site in southwestern Japan, where stream acidification due to acid rain is of increasing concern. Statistical analysis shows that the elements in the granitic soil can be classified into three groups according to their mineralogical derivations: Group 1 (K, Mg, Mn, Rb, and Nb), group 2 (Fe, Ti, Zr, Cr, and Pb), and group 3 (Ca, Na, Sr, and P), which are related to the presence of K-feldspar and K-mica, Fe-Ti oxides, and plagioclase and apatite, respectively. The pattern of distribution of group-1 elements in the soil column differs slightly among the sites because of differences in the proportions of K-feldspar and K-mica, The organic surface soil is enriched in group-2 elements and shows a strong magnetic susceptibility, indicating the presence of biogenetic magnetite. At all sites, Ca and Sr are intensely depleted due to the selective dissolution of plagioclase. Ca depletion compared to the original granite reaches more than 90 % in the C horizon, resulting in a decreasing capacity for the neutralization of acid rain and an increasing contribution of atmospheric Ca to the surface soil and vegetation.  相似文献
