A metabolomics based approach has been used to study the infection of the Hwacheong rice cultivar (Oryza sativa L. cv. Hwacheong) with compatible (KJ201) and incompatible (KJ401) strains of the rice blast fungal pathogen Magnaporthe grisea. The metabolic response of the rice plants to each strain was assessed 0, 6, 12, 24, 36, and 48 h post inoculation. Nuclear
Magnetic Resonance (NMR) spectroscopy and Gas and Liquid Chromatography Tandem Mass spectrometry (GC/LC-MS/MS) were used to
study both aqueous and organic phase metabolites, collectively resulting in the identification of 93 compounds. Clear metabolic
profiles were observed at each time point but there were no significant differences in the metabolic response elicited by
each pathogen strain until 24 h post inoculation. The largest change was found to be in alanine, which was ~30% (±9%) higher
in the leaves from the compatible, compared to the resistant, plants. Together with several other metabolites (malate, glutamine,
proline, cinnamate and an unknown sugar) alanine exhibited a good correlation between time of fungal penetration into the
leaf and the divergence of metabolite profiles in each interaction. The results indicate both that a wide range of metabolites
can be identified in rice leaves and that metabolomics has potential for the study of biochemical changes in plant-pathogen

We analyze the history of acid rain in China over the last three decades in the light of the relationship between China and Japan. China has been aware of the problem of acid rain and has been conducting scientific research since the late 1970s. However, little effort has been put into practical countermeasures. In the early 1990s, acid rain was re-evaluated as an international environmental problem. Meanwhile, Japan became concerned about trans-boundary air pollution from China. What is important is that Japan's concern strongly influences China's acid rain policy. In the context of growing international concern for environmental problems, China has taken remarkable steps to control acid rain. Improvement of rice (Oryza sativa L.) growth in upland conditions with deep tillage and mulch 总被引:1,自引:0,他引:1
The productivity of upland rice in Japan as well as in the world is low and unstable owing to scarce and unpredictable rainfall. The objective of this study was to examine whether agronomic methods could enhance grain yield of upland rice. Four field experiments were conducted from 2001 to 2003 in Nishitokyo, Japan, under upland conditions with different water supplies, in order to quantify the effects of deep tillage combined with deep placement of manure (50-cm depth), straw mulch (6 t ha−1), or their combinations on the growth and grain yield of rice. Mulch kept surface soil moisture higher than without mulch even at reproductive stage, and it increased yield to the greatest extent under the most favourable conditions with much rainfall before heading (i.e., 2003). Deep tillage with deep placement of manure induced deep root proliferation and higher nitrogen uptake, increasing biomass production, and panicle number, and consequently grain yield was enhanced under the two lowest yielding environments with less rainfall before heading. Rice plants with deep tillage with deep manure application without mulch tended to have lower leaf water potential and higher diffusion resistance during drought, and negative effects on grain filling and harvest index in some experiments compared with the control. When deep tillage with deep placement of manure was combined with mulching in two experiments in 2002 and 2003, grain yield always enhanced compared with the control (P < 0.10, 6.0 t ha−1 versus 5.4 t ha−1 on average), suggesting their synergetic mechanisms for yield increase and stabilization. The results showed that deep tillage or mulching can improve grain yield of rice under drought-prone rainfed upland conditions in a temperate climate on an Andosol, and their combination had more consistent and greater positive effects. 相似文献

We evaluated the developmental ability of oocytes in porcine primordial follicles xenografted into nude mice. Ovarian tissues from 20-day-old piglets, in which most of the follicles were primordial, were transplanted under the kidney capsules of ovariectomized nude mice. Forty-nine to 89 days after grafting (mean +/- SEM, 66.9 +/- 1.9 days; n = 64), the host mice showed the presence of cornified epithelial cells in their vaginal smears for the first time. The mice were then treated with 4 IU of equine chorionic gonadotropin (eCG) 60 days after first detection of vaginal cornification. Oocytes were collected from the host mice 48 h after treatment with eCG, and then matured. The maturation rates, based on the incidence of first polar body, ranged from 25.1% to 42.5%. They were then fertilized in vitro and cultured in vitro for 6 days, or transferred into estrous-synchronized recipients and recovered after 6 days. On Day 6 of culture, 15.4% of the matured oocytes had cleaved to the 2- to 8-cell stage. However, neither the embryos cultured in vitro nor those transferred and recovered developed to advanced embryonic stages, such as morulae or blastocysts. This result suggests that the developmental ability of xenografted oocytes is insufficient, even after in vitro maturation. Further strategies, such as improvement of hormonal treatment for host mice, are required to enable oocytes in xenografted ovarian tissues to acquire the cytoplasmic maturation necessary for embryonic development. 相似文献

We investigated chronological changes in immunohistochemical phenotyping in the thymus and spleen in Crl:CD rats up to the age of about one year. In the thymus, T cells increased markedly from 3 to 4 weeks of age. Proliferating cells also increased markedly at these points. B cells tended towards an increase with age. In the spleen, white pulp increased until 9 weeks of age and remained fairly stable thereafter. In the periarteriolar lymphoid sheath and marginal zone, T cells gradually increased until 9 weeks of age and became almost flat thereafter. In the lymph follicle, T cells increased with age. B cells tended towards an increase with age in all areas of the spleen. It was concluded that development of the thymus was most marked from 3 to 4 weeks of age and that both the thymus and spleen had matured by 9 weeks of age. 相似文献

To assess the effect of changes in organic litter stock on seasonal changes in heterotrophic respiration (R H), soil respiration (R S), and total ecosystem respiration (R E), we measured seasonal changes in leaf litter respiration (R LL) by the chamber method and estimated the seasonal change in total R H using the RothC model in a warm-temperate mixed deciduous?Cevergreen forest in Japan. Both R E and R S had seasonal hysteresis and were higher in spring than at the same temperature during autumn. Under warm and humid conditions, the rate of decomposition of newly supplied leaf litter in one?year was high (60% loss). Consequently, R LL and R H were higher in spring after leaf drop, when more fresh material was available, than in autumn. In this study, 42 and 88% of the difference in R E and R S between spring and autumn (soil temperature 16?C18°C) could be accounted for by the difference in R H, respectively, and 71% of the difference in R H could be accounted for by the difference in R LL. This study showed that seasonal changes in heterotrophic respiration (R LL and R H) could be a major factor in the seasonal hysteresis of R E and R S. 相似文献

ABSTRACTThe sugar-accumulating potential of global and local sweet and grain sorghum varieties were tested under the local conditions. The basis for this study was the dependency of sugar accumulation on temperature and photoperiod. Thus, the efficacy of cultivars as a bioenergy source would need to be determined based on their performance under the local environmental conditions. A strong correlation of sucrose content with brix was observed, enabling large-scale screening of varieties for high sucrose content. The morphological characteristics inherent in sweet sorghum, such as tall stems, greater number of leaves and a longer vegetative period, were found to correlate with the total stem sugar content. Assessment of sugars along the stem revealed maximum sugar accumulation in the upper intermediate to upper internodes in most of the varieties tested. The maximum theoretical ethanol yield (MTEY), a function of brix and juice yield, was determined as a better indicator of testing the performance of a variety as a potential source of bioethanol, mainly due to a negative correlation of stem juiciness and sucrose content in the varieties tested. Further, the relative expression of vacuolar invertase genes, SbINV1 and SbINV2, was studied, and a strong negative correlation of SbINV2 to stem sucrose content was observed. This reveals a possibility of involvement of vacuolar invertase gene, SbINV2, in sugar accumulation in sweet sorghum stems, and as a key candidate for molecular breeding studies for higher stem sugar content. 相似文献

A five-month-old male beagle dog suddenly became moribund. Bloody fluid accumulated in the thoracic and abdominal cavities, and soft yellow flecks were floating in the thoracic fluid. The mediastinum and pericardium became dark reddish with villous thickening. Other parietal and pulmonary pleurae were rough, and the organs adhered to each other. Histologically, most mediastinal pleura formed papillary projections covered by a single layer of mesothelial cells. Many macrophages and neutrophils infiltrated the submesothelial connective tissue. At the mediastinum adjacent to the pericardium, cuboidal mesothelial cells proliferated solidly and formed a thick surface stratum. The flecks consisted of gram-negative filamentous or small bacillary (coccoid) bacteria. In the right posterior lobe of the lung, neutrophilic infiltration and a large encapsulated abscess including a bacterial colony were present. We diagnosed this case as “bacterial pleuritis with thickened mesothelial hyperplasia”. The cause of the pleuritis might be a chronic pleural infection spread via the lung abscess. 相似文献

In porcine oocytes, the function of the zona pellucida (ZP) with regard to sperm
penetration or prevention of polyspermy is not well understood. In the present study, we
investigated the effects of the ZP on sperm penetration during in vitro
fertilization (IVF). We collected in vitro-matured oocytes with a first
polar body (ZP+ oocytes). Some of them were freed from the ZP (ZP− oocytes) by two
treatments (pronase and mechanical pipetting), and the effects of these treatments on
sperm penetration parameters (sperm penetration rate and numbers of penetrated sperm per
oocyte) were evaluated. There was no evident difference in the parameters between the two
groups. Secondly, we compared the sperm penetration parameters of ZP+ and ZP− oocytes
using frozen-thawed epididymal spermatozoa from four boars. Sperm penetration into ZP+
oocytes was found to be accelerated relative to ZP− oocytes. Thirdly, we evaluated the
sperm penetration of ZP+ and ZP− oocytes at 1−10 h after IVF (3 h gamete co-incubation).
The proportions of oocytes penetrated by sperm increased significantly with time in both
groups; however, the number of penetrated sperm per oocyte did not increase in ZP−
oocytes. Finally, we performed IVF using ZP− oocytes divided into control (3 h) and
prolonged gamete co-incubation (5 h) groups. Greater numbers of sperm penetrated in the 5
h group than in the control group. These results suggest that the ZP and oolemma are not
competent factors for prevention of polyspermy in our present porcine IVF system. However,
it appears that ZP removal is one of the possibilities for reducing polyspermic

A gene for FK506 binding protein 6 (Fkbp6) expresses during a specific stage of male and female meiosis. Disruption of the gene influences male reproduction, i.e. arrests spermatogenesis, but not female reproduction. Using the mouse model (targeted disruption), the role of the gene in homologous chromosome pairing has been demonstrated in a previous study. For further understanding the function of Fkbp6 in chromosome synapsis, we evaluated chromosome pairings during male meiosis in the as/as rat, a spontaneous null mutation, and compared them with those of the mouse model. Electron microscopy of the pachytene nuclei unveiled several types of abnormal chromosome pairing in the rat model, as shown in the mouse previously. The frequencies of aberrant pairings in the knockout mice and mutant rats were 42 of 67 nuclei (62.7%) and 20 out of 74 nuclei (27.0%), respectively. In order to clarify the mechanism of male specific infertility in Fkbp6 deficiency, the localization of gammaH2AX, a marker protein of XY chromosome inactivation during male meiosis, was examined. Immunostaining of gammaH2AX unveiled normal localization of the molecule to XY chromosomes (XY body) in both models, showing the independency of FKBP6 in sex chromosome inactivation. Besides the XY body, focal localization of gammaH2AX was observed in accordance with the unsynapsed chromosomes in both types of null animal. These results indicate the fundamental role of Fkbp6 in homologous chromosome synapsis during male meiosis. In conclusion, male specific infertility under Fkbp6 deficiency remains unsolved. 相似文献
