Use of somaclonal variation and in vitro selection for chilling tolerance improvement in rice 总被引:7,自引:0,他引:7
Embryo-derived calli of four rice varieties cultivated at high altitude in Burundi — Facagro 57, Facagro 76, Kirundo 3 and
Kirundo 9 — were submitted to different temperature regimes. The percentage of regenerating calli greatly varied depending
on variety, length of culture and callus temperature treatment. The reduction of regeneration percentages induced by low temperature
was more pronounced in the more sensitive varieties. Regenerated plants (R0) and their progenies in R1, R2 and R3 were cold-screened
together with control plants. In all varieties, significantly higher survival rates were obtained in R3 with in vitro plants
than with control plants. Such chilling tolerance improvement was not obtained following a massal selection applied during
3 successive generations onto the control plants. In vitro plants regenerated from calli cultivated either at 25 °C, either
at 4 °C, were cultivated at different altitudes in Burundi during two successive generations. For most observed traits, the
in vitro plants were characterized by lower means, larger variation and higher maximum values than the control plants. The
most chilling-tolerant somaclonal families were most usually characterized by extensive differences in fatty acid composition,
chilling-induced electrolyte leakage and chlorophyll fluorescence, compared to the varieties they derived from.

The large-scale conversion of old forests to tree plantations has made it increasingly important to understand how understory vegetation responds to such landscape changes. For instance, in some forest types a reduction in understory richness and cover is thought to result from the development of canopy closure in plantations, although there is a paucity of empirical data demonstrating this relationship. We used a 420-year forest chronosequence as a case study to assess the relationship between stand age, tree canopy cover and understory vascular plant richness and composition in the Siskiyou Mountains of Oregon. The chronosequence consisted of six young managed (age 7–44) and nine older unmanaged (age 90–427) stands. All stands were similar in underlying geology, slope, elevation, and aspect. We found a non-linear relationship between stand age and richness, in which richness was highest in the youngest stands, reached a low in mid-aged stands (∼55 years), then increased in the oldest stands. We also found that percent tree canopy cover was correlated with total understory cover, richness, diversity, and species composition. In general, young stands were characterized by high shrub and graminoid cover and old stands were characterized by an abundant herb layer. Our work suggests that a major component of our study landscape is currently entering the forest stage (canopy closure) characterized by low levels of vascular plant species richness and cover. We use our results to discuss the potential effects of future forest management on understory plants. 相似文献

We analysed metabolic diversity in apples from wild species, elite material and a F1 population, using liquid chromatography–mass spectrometry (LC‐QTOF‐MS). The evaluated elite material appeared to have strongly reduced levels of phenolic compounds, down to 1% of the concentrations in the investigated wild germplasm. In one quarter of the F1 population, the concentrations of phenolic compounds such as quercetin derivatives, procyanidin, catechin and epicatechin were further significantly reduced, due to accumulation of recessive alleles of putatively leucoanthocyanidin reductase, a structural gene that is located at the top of LG16. In another part of F1 progeny, putatively glycosylated forms of β‐glycols were up to 50 times more abundant compared to both parents. These metabolites were mapped with high logarithm of odds (LOD) scores at the top of LG8, and progeny that was homozygous recessive for the candidate gene showed the elevated levels. We hypothesize that this was caused by inheritance of non‐functional alleles of enoyl‐CoA hydratase gene. Both examples of transgressive segregation, where some progeny significantly deviated from both parents, were caused by accumulation of recessive alleles. 相似文献

Pterocarpus santalinoides is a multipurpose tree species, native to Africa and South America. However, excessive pruning for fodder is jeopardising its seed production leading to poor regeneration in the Sahel region. Efficient vegetative propagation techniques may overcome this concern. To address this issue, four experiments were conducted to determine the effects of substrate (Experiment 1), cutting type (Experiments 2 and 3), genotype (Experiment 4) and auxins (Experiments 1, 2, and 4) on rooting ability in a non-mist propagating unit in Mali. Among the four tested substrates, no significant differences were found in rooting parameters, including the percentage of rooted cuttings, the number of roots and secondary roots per rooted cutting, and the length of the longest root per rooted cutting. Semi-hardwood cuttings rooted significantly better than softwood cuttings with a higher percentage of rooted cuttings (31.7?±?6.5% vs. 9.2?±?2.7%), greater number of roots (8.2?±?4.8 vs 1.5?±?0.7), greater number of secondary roots (16.7?±?4.2 vs 4.6?±?1.8) and longer root length (17.8?±?12.0?cm vs 2.3?±?1.0?cm) per rooted cutting. In addition, one-node cuttings cut below the second and third nodes, respectively, had the highest rooting success (88% and 90%, respectively) followed by cuttings cut below the fourth node (68%) and the first node (62%). Significant differences in the percentage of rooted cuttings were found among 12 randomly selected genotypes with rooting varying from 2% to 75%.The results also showed that auxin application, especially IBA, had a significant effect on rooting success, the number of roots and secondary roots per rooted cutting. Single-node cuttings, and cuttings of 10?cm length with multiple softwood and semi-hardwood nodes, can be used with or without auxin treatment for successful clonal propagation of P. santalinoides. The results of the present study indicated that P. santalinoides is amenable to clonal propagation using low-cost propagators and sand-based rooting media. 相似文献

OBJECTIVE: To determine the onset of immunity after IM administration of a single dose of a recombinant canarypox virus vaccine against West Nile virus (WNV) in horses in a blind challenge trial. ANIMALS: 20 mixed-breed horses. PROCEDURE: Horses with no prior exposure to WNV were randomly assigned to 1 of 2 groups (10 horses/group). In 1 group, a recombinant canarypox virus vaccine against WNV was administered to each horse once (day 0). The other 10 control horses were untreated. On day 26, 9 treated and 10 control horses were challenged via the bites of mosquitoes (Aedes albopictus) infected with WNV. Clinical responses and WNV isolation were monitored for 14 days after challenge exposure; antibody responses against WNV after administration of the vaccine and challenge were also assessed in both groups. RESULTS: Following challenge via WNV-infected mosquitoes, 1 of 9 treated horses developed viremia. In contrast, 8 of 10 control horses developed viremia after challenge exposure to WNV-infected mosquitoes. All horses seroconverted after WNV challenge; compared with control horses, antibody responses in the horses that received the vaccine were detected earlier. CONCLUSIONS AND CLINICAL RELEVANCe: In horses, a single dose of the recombinant canarypox virus-WNV vaccine appears to provide early protection against development of viremia after challenge with WNV-infected mosquitoes, even in the absence of measurable antibody titers in some horses. This vaccine may provide veterinarians with an important tool in controlling WNV infection during a natural outbreak or under conditions in which a rapid onset of protection is required. 相似文献

The increasing international movement of horses combined with the relaxation of veterinary regulations has resulted in an increased incidence of equine infectious diseases. Vaccination, along with management measures, has become the primary method for the effective control of these diseases. Traditionally modified live and inactivated vaccines have been used and these vaccines have proven to be very successful in preventing disease. However, there are a number of equine infectious diseases for which conventional technology has shown its limitations. The advent of recombinant technology has stimulated the development of second generation vaccines, including gene deleted mutants, live vectored vaccines and DNA vaccines. These vaccines have in common that protective antigens are endogenously processed and presented along the molecules of the MHC I and MHC II complex, resulting in the stimulation of both humoral and cell-mediated immune responses similar to natural infection. The present paper provides a review of the vaccines being employed today against the most important equine viral diseases followed by a summary of new developments that are expected to bring improved vaccines to the market in the foreseeable future. 相似文献
