The effects of temperature and autotomy of chelipeds on survival, growth and moulting of mangrove crab (Scylla serrata) juveniles were investigated under laboratory conditions for 60 days. Hatchery‐produced crabs with 2.0–2.3 cm internal carapace width (1.7–2.2 g body weight) at intermoult stage were exposed to one of four temperature treatments (constant 29, 32 or 35°C, or ambient [24–31°C]) or subjected to cheliped autotomy. All crabs held at 35°C had 100% mortality due to incomplete moulting during first moult. The mean survival of crabs at termination was 58%, 64% and 50% for ambient temperature, 29 and 32°C respectively. Specific growth rate (SGR) of crabs in the ambient and 29°C were comparable but significantly lower than those at 32°C. The moult interval of the crabs was significantly shorter in treatments with constant water temperature of 29 and 32°C compared with ambient temperature. The survival of crabs with intact chelipeds was comparable with those with one or two autotomized chelipeds. Crabs with intact or one autotomized chelipeds had significantly higher SGR than crabs with both chelipeds autotomized in the first moult. On the second moult, however, high SGR was observed in crabs with two chelipeds autotomized. The moult interval was significantly shorter in the autotomized crabs compared with crabs with intact chelipeds. The results suggest that the optimum water temperature for rearing Sserrata juveniles ranges from 29 to 32°C. Likewise, autotomy of chelipeds can promote moulting without adversely affecting survival of crabs.  相似文献

Biscogniauxia mediterranea is one of the most frequent fungal pathogens involved in cork oak decline in the Mediterranean Basin, causing charcoal canker. In Portugal, this disease is widespread on adult declining trees but nowadays it increasingly affects young trees and exhibits atypical symptoms, leading to the hypothesis that some change in the fungus may have occurred. In order to evaluate the genetic diversity and phylogenetic relationship of B. mediterranea associated with cork oak, 102 isolates were obtained from young and adult trees of Quercus suber and other hosts species with different disease expression, from several Mediterranean countries. The collection of isolates was analyzed by individual and multigene phylogenies using Maximum-Likelihood approach based on nucleotide sequences of the internal transcribed spacers of ribosomal DNA, translation elongation factor 1-α and β-tubulin genes, and by microsatellite-primed PCR profiles. Sequence analyses separated the Mediterranean isolates from those from other regions, while MSP-PCR analysis revealed relevant but unstructured diversity among the Mediterranean isolates under study, making this a monophyletic but diverse population. Considering the adaptive capacity of the fungus in the Mediterranean-climate ecosystems and the present climatic change scenario, all conditions are gathered to favor aggravation of the disease in cork oak stands.  相似文献

Context

Deforestation is a major driver of biodiversity loss, mainly due to agriculture. As rice is among the world’s most important crops, determining how agricultural communities are shaped is imperative. However, few studies have addressed the factors that alter community assembly in human-modified landscapes. We aim to quantify taxonomic, functional, trait and phylogenetic diversity of an anuran community from rice crops on a biodiversity hotspot.

Objectives

Identify local and landscape characteristics responsible for variations in multiple dimensions of anuran diversity in rice crops.

Methods

This study was performed in Tocantins, Brazil. We chose 36 lentic waterbodies on rice fields for anuran sampling. We quantified taxonomic diversity (TD), functional diversity (FD) and phylogenetic diversity (PD) for each waterbody. We also estimated the mean functional differences among species for each trait separately. To evaluate how local and landscape scale features affect anurans, we performed generalized linear mixed models in 500, 1000 and 1500 m buffers around each waterbody.

Results

We found increased PD and FD in waterbodies closer to many other waterbodies and large forest patches. Anuran biomass decreased with increasing distance to the closest waterbody. Trait diversity varied with waterbody abundance and closeness, percentage of bare ground and marginal vegetation.

Conclusions

Our study emphasizes the importance of waterbody and forest patch networks for maintaining high anuran FD and PD in agricultural landscapes. As both metrics are known to be related to ecosystem resilience, understanding these patterns is pivotal for biodiversity management, especially in the tropics, where agricultural expansion is unrelenting and biodiversity is especially unique.

Maize is a staple food crop in many developing countries, hence becoming an attractive target for biofortification programs toward populations at risk of micronutrient deficiencies. A South African white endosperm maize inbred line was engineered with a carotenogenic mini-pathway to generate high-carotenoid maize, which accumulates β-carotene, lutein and zeaxanthin. As maize porridge is a traditional meal for poor populations in sub-Saharan African countries, high-carotenoid maize was used as raw material to prepare different maize meals. The objective of this work was to assess the impact of popular home-cooking techniques and different cooking parameters (temperature, time and pH) on the final carotenoid content in the cooked product, using a spectrophotometric technique based on the mean absorption of carotenoids at 450 nm. Carotenoid levels were not only preserved, but also enhanced in high-carotenoid maize porridges. The carotenoid content was increased when temperatures ≤95 °C were combined with short cooking times (10–60 min). The most optimum thermal treatment was 75 °C/10 min. When treated under those conditions at pH 5, high-carotenoid maize porridges doubled the initial carotenoid content up to 88 μg/g dry weight. Regarding to cooking techniques, the highest carotenoid content was found when unfermented thin porridges were prepared (51 μg/g dry weight of high-carotenoid maize porridge). We conclude that high-carotenoid maize may contribute to enhance the dietary status of rural populations who depend on maize as a staple food.  相似文献

Osteoarthritis (OA) remains a prevalent chronic disease without effective prevention and treatment. Amentadione (YP), a meroditerpenoid purified from the alga Cystoseira usneoides, has demonstrated anti-inflammatory activity. Here, we investigated the YP anti-osteoarthritic potential, by using a novel OA preclinical drug development pipeline designed to evaluate the anti-inflammatory and anti-mineralizing activities of potential OA-protective compounds. The workflow was based on in vitro primary cell cultures followed by human cartilage explants assays and a new OA co-culture model, combining cartilage explants with synoviocytes under interleukin-1β (IL-1β) or hydroxyapatite (HAP) stimulation. A combination of gene expression analysis and measurement of inflammatory mediators showed that the proposed model mimicked early disease stages, while YP counteracted inflammatory responses by downregulation of COX-2 and IL-6, improved cartilage homeostasis by downregulation of MMP3 and the chondrocytes hypertrophic differentiation factors Col10 and Runx2. Importantly, YP downregulated NF-κB gene expression and decreased phosphorylated IkBα/total IkBα ratio in chondrocytes. These results indicate the co-culture as a relevant pre-clinical OA model, and strongly suggest YP as a cartilage protective factor by inhibiting inflammatory, mineralizing, catabolic and differentiation processes during OA development, through inhibition of NF-κB signaling pathways, with high therapeutic potential.  相似文献

Microalgae have become a promising novel and sustainable feedstock for meeting the rising demand for food and feed. However, microalgae-based products are currently hindered by high production costs. One major reason for this is that commonly cultivated wildtype strains do not possess the robustness and productivity required for successful industrial production. Several strain improvement technologies have been developed towards creating more stress tolerant and productive strains. While classical methods of forward genetics have been extensively used to determine gene function of randomly generated mutants, reverse genetics has been explored to generate specific mutations and target phenotypes. Site-directed mutagenesis can be accomplished by employing different gene editing tools, which enable the generation of tailor-made genotypes. Nevertheless, strategies promoting the selection of randomly generated mutants avoid the introduction of foreign genetic material. In this paper, we review different microalgal strain improvement approaches and their applications, with a primary focus on random mutagenesis. Current challenges hampering strain improvement, selection, and commercialization will be discussed. The combination of these approaches with high-throughput technologies, such as fluorescence-activated cell sorting, as tools to select the most promising mutants, will also be discussed.  相似文献

The fungus Sydowia polyspora is frequently isolated from conifers worldwide and is considered a pathogen on several hosts. Stone pine (Pinus pinea) is one of the most important forestry species throughout the Mediterranean basin due to the value of the edible pine nut. Stone pines showing tip dieback, needles with tan‐ to yellow‐coloured lesions and shoot death, observed in stands in Portugal, were sampled for analysis. Fungal colonies covered with cream‐coloured spore masses, were consistently obtained. Morphological and phylogenetic analyses of the ITS rDNA region enabled identification of these isolates as S. polyspora. Inoculation tests showed that the fungus caused lesions on excised P. pinea shoots. The symptoms observed might have a negative effect on pine nut production, and thus, evaluation of the impact of this disease is of relevance to future research. This paper is the first to report S. polyspora causing disease on P. pinea.  相似文献

Dothistroma and Lecanosticta needle blight are among the most damaging foliage diseases in plantations and natural pine stands worldwide, and are therefore listed as quarantine organisms in numerous countries. The aim of this study was to validate different methodologies used for the detection of both diseases. Based on multiplex quantitative PCR (qPCR), simultaneous detection and discrimination of the three pathogens Dothistroma septosporum, Dothistroma pini and Lecanosticta acicola were possible in symptomatic needles. Additionally, the same set of needles was analysed for the presence of all three pathogens using novel end‐point PCR assays followed by enzymatic digestion of PCR products. Results were compared with the qPCR data, and for validation, collected needles were screened morphologically for the presence of typical fruiting bodies and spores. Differences in detection sensitivity were found between the detection tools. The qPCR‐based detection allowed for specific and efficient identification and quantification of all three pathogens simultaneously. At the same time, conventional PCR assays, which target the multicopy ITS region, showed increased detection sensitivity and can be conducted without the use of expensive infrastructure and reagents.  相似文献

The effect of the addition of Bacillus probiotic during larvae and postlarvae culture of the white shrimp Litopenaeus vannamei was examined in three separate experiments: (I) Nauplius4‐5 to Zoea3, which were exposed to probiotic in the water (Pw), only in the microalgae (Pm), in the water and microalgae (Pwm) and a control with no probiotic (C); (II) Mysis1 to Mysis3, which were exposed to probiotic in the water (Pw), only in Artemia (Pa), in the water and Artemia (Pwa) and a control (C); (III) PL1 to PL10, which were exposed to similar treatments for mysis experiment. The use of probiotic Bacillus spp. resulted in an increase in the survival and growth of zoea and mysis phases, especially when the probiotic was added only in the water. For postlarvae, the use of the probiotic had no influence on the zootechnical parameters, however, there was a reduction in the count of presumptive Vibrio both for water and shrimp.  相似文献
