The Estremadura Spur pockmarks are a unique and unexplored ecosystem located in the North Atlantic, off the coast of Portugal. A total of 85 marine-derived actinomycetes were isolated and cultured from sediments collected from this ecosystem at a depth of 200 to 350 m. Nine genera, Streptomyces, Micromonospora, Saccharopolyspora, Actinomadura, Actinopolymorpha, Nocardiopsis, Saccharomonospora, Stackebrandtia, and Verrucosispora were identified by 16S rRNA gene sequencing analyses, from which the first two were the most predominant. Non-targeted LC-MS/MS, in combination with molecular networking, revealed high metabolite diversity, including several known metabolites, such as surugamide, antimycin, etamycin, physostigmine, desferrioxamine, ikarugamycin, piericidine, and rakicidin derivatives, as well as numerous unidentified metabolites. Taxonomy was the strongest parameter influencing the metabolite production, highlighting the different biosynthetic potentials of phylogenetically related actinomycetes; the majority of the chemical classes can be used as chemotaxonomic markers, as the metabolite distribution was mostly genera-specific. The EtOAc extracts of the actinomycete isolates demonstrated antimicrobial and antioxidant activity. Altogether, this study demonstrates that the Estremadura Spur is a source of actinomycetes with potential applications for biotechnology. It highlights the importance of investigating actinomycetes from unique ecosystems, such as pockmarks, as the metabolite production reflects their adaptation to this habitat. 相似文献

Six cultivars (Franquette, Marbot, Mayette, Mellanaise, Lara, and Parisienne) of walnuts (Juglans regia L.) were collected during the 2001 crop, from Bragan?a, Portugal. Chemical composition, including moisture, total oil content, crude protein, ash, carbohydrates, and nutritional value, was evaluated. Fat was the predominant component, ranging from 62.3 to 66.5%. Total oil was extracted and analyzed for fatty acids, sterols, oxidative stability, and peroxide value. Fatty acids and sterols were determined by gas-liquid chromatography coupled to a flame ionization detector. Eighteen fatty acids were quantified. Polyunsaturated fatty acids and, in particular, linoleic acid were predominant. Beta-Sitosterol, delta(5)-avenasterol, and campesterol were the major sterols found. Differences were observed among the studied cultivars, especially in peroxide values and in the sterol profile. 相似文献

Larval resistance to temporary starvation is considered a key factor for successful development in the wild. Subjecting larvae to temporary starvation during early and/or late development is occasionally used in larviculture to reduce production costs.Mithraculus forceps is a popular species in the marine aquarium industry for their ability to control nuisance algae in aquarium tanks; a larval culture methodology was previously proposed in order to avoid collection from the wild. In an attempt to reduce production/feeding costs of M. forceps larval culture (two zoea stages and a megalopa), larvae (megalopa stage) were starved after 7 and 8 days post-hatch (DPH); starvation treatments were compared with those of the control treatment where larvae were fed continuously. No differences were found in survival to juvenile, metamorphosis synchronism or larval duration between the treatments, which suggest that at least by day 7 DPH, megalopae have already achieved the point-of-reserve-saturation (PRS) and are able to successfully metamorphose to crab stage without feeding (facultative lecithotrophic); newly metamorphosed juveniles are slightly smaller (particularly the ones starved after 7DPH). This flexible way of development might be very advantageous in the wild and might allow the reduction of costs of larval culture in captivity. 相似文献

Arabica coffee production is based on highly productive cultivars; however, these cultivars are susceptible to infestation by several biotic agents, including root-knot nematodes. Collections of wild Coffea arabica germplasm represent an important source of genetic variability for resistant cultivar development. In this study, 1046 plants derived from 71 wild coffee trees were evaluated with respect to Meloidogyne paranaensis resistance. In addition to information on plants reactions, we also evaluated the genetic parameters related to resistance. Progenies from the five most promising plants were also evaluated regarding resistance to M. incognita and M. exigua. The yield potential of selected plants was estimated through analysis of data for fruits harvested in 4 different years. Forty-seven plants were considered resistant based on reproduction factor values. The estimated heritability was high for all analyzed variables leading to substantial selection gain, mainly at the progeny mean level. On the basis of heritabilities and genetic correlations, we conclude that selection could be performed based on values of the gall and egg mass index. However, higher genetic gain could be obtained based on nematode count variables. A second experiment confirmed the reactions of the selected five plants to M. paranaensis, and multiple resistance was detected in three of them. Three different HPLC detection systems were compared for the determination of tocopherols and tocotrienols in olive oil: fluorescence and diode array connected in series, ultraviolet, and evaporative light scattering. The best results were obtained with the fluorescence detector, which was successfully applied in the quantification of tocopherols and tocotrienols in 18 samples of Portuguese olive oils. To support the validity of the method, the parameters evaluated were linearity, detection limits, repeatability, and recovery. All of the studied samples showed similar qualitative profiles with six identified compounds: alpha-T, beta-T, gamma-T, delta-T, alpha-T3, and gamma-T3. Alpha-tocopherol (alpha-T) was the main vitamin E isomer in all samples ranging from 93 to 260 mg/kg. The total tocopherols and tocotrienols ranged from 100 to 270 mg/kg. Geographic origin did not seem to influence the tocopherol and tocotrienol composition of the olive oils under evaluation. 相似文献

One of the actions that has been implemented to support the recovery of the panmictic population of European eel is stocking of waterbodies where natural recruitment is low or null. However, growth conditions of the stocked eels can vary greatly. This circumstance emphasises the importance to determine ideal habitat conditions to contribute to the success of stocking actions and, consequently, to increase the production of silver eels. This study aims to evaluate the early settlement and growth of stocked glass eels in the upper reaches of a fragmented river. Stocking was carried out, in 2014, at three sites of an inland tributary of the Mondego river basin (Central Portugal), and its monitoring was conducted during the following two years, until 2016, along with the collection of environmental and hydromorphological parameters. The results showed a successful dispersion throughout the study area. Growth varied spatially, although environmental parameters have not clearly explained this variation, but overall with high growth rates and a positive allometric growth in this early stage of stocking, suggesting a good condition of the stocked individuals. This study showed that these upper reaches of fragmented watercourses, a shared feature amongst most European rivers that are currently inaccessible for natural recruitment, may be suitable habitats for eel stocking. 相似文献

Methicillin-resistant Staphylococcus aureus (MRSA) was first described in horses in 1996. The frequency of MRSA colonization in horses varies among European countries, but it is unknown in Portugal. The aim of this study was to screen for MRSA nasal carriage in a sample of horses entering the Equine Unit, Large Animal Veterinary Teaching Hospital of the Faculty of Veterinary Medicine, Lisbon, Portugal. Seventy-one horses were swabbed, and MRSA was identified by selective isolation on a chromogenic medium. Two S aureus isolates showed resistance to oxacillin (minimum inhibitory concentration >4 μg/mL) and contained the mecA gene. Both strains were isolated from purebred Lusitano horses that lived in farms with more than 20 equines. These MRSA strains represented two different clones: isolate FMVA3/10 was an MRSA sequence type ST5 with a staphylococcal cassette chromosome mec VI, coresistant to erythromycin and clindamycin; and isolate FMVA16/10 was sequence type ST398, with a staphylococcal cassette chromosome mec IV, coresistant to tetracycline, gentamicin, and trimethoprim. Isolate FMVA3/10 represents a human epidemic clone not previously reported among horses in Europe, which once again reinforces the fact that transmission of MRSA clones between horses and humans occurs. Isolate FMVA16/10 represents the first report of the detection of MRSA ST398 among horses in Portugal. Lusitano horses can carry animal and human MRSA in the nostrils, acting as reservoirs, which can potentially be transmitted to humans. 相似文献

We describe the genome sequence of the protist Trichomonas vaginalis, a sexually transmitted human pathogen. Repeats and transposable elements comprise about two-thirds of the approximately 160-megabase genome, reflecting a recent massive expansion of genetic material. This expansion, in conjunction with the shaping of metabolic pathways that likely transpired through lateral gene transfer from bacteria, and amplification of specific gene families implicated in pathogenesis and phagocytosis of host proteins may exemplify adaptations of the parasite during its transition to a urogenital environment. The genome sequence predicts previously unknown functions for the hydrogenosome, which support a common evolutionary origin of this unusual organelle with mitochondria. 相似文献
