Although the mammalian immune system is generally thought to develop in a linear fashion, findings in avian and murine species argue instead for the developmentally ordered appearance (or "layering") of distinct hematopoietic stem cells (HSCs) that give rise to distinct lymphocyte lineages at different stages of development. Here we provide evidence of an analogous layered immune system in humans. Our results suggest that fetal and adult T cells are distinct populations that arise from different populations of HSCs that are present at different stages of development. We also provide evidence that the fetal T cell lineage is biased toward immune tolerance. These observations offer a mechanistic explanation for the tolerogenic properties of the developing fetus and for variable degrees of immune responsiveness at birth.  相似文献

Although a principal source of energy and protein for millions of the world's poorest people, the nutritional value of sorghum (Sorghum bicolor L. Moench) is diminished because of low digestibility of grain protein and starch. To address this problem, we analyzed the properties of two sorghum lines that have a common pedigree but differ in digestibility. Consistent with results based on a ruminal fluid assay, the protein and starch of one line (KS48) was more thoroughly digested than that of the other (KS51) using in vitro assays based on pepsin and α-amylase. The indigestibility of KS51 relative to KS48 was shown to be due to (i) a greater abundance of disulfide-bonded proteins; (ii) presence in KS51 of non-waxy starch and the accompanying granule-bound starch synthase; and (iii) the differing nature of the protein matrix and its interaction with starch. The current findings suggest that each of these factors should be considered in efforts to enhance the nutritional value of sorghum grain.  相似文献

Existing δ2H and δ18O values for precipitation and surface water in the Nile Basin were used to analyze precipitation inputs and the influence of evaporation on the isotopic signal of the Nile River and its tributaries. The goal of the data analysis was to better understand basin processes that influence seasonal streamflow for the source waters of the Nile River, because climate and hydrologic models have continued to produce high uncertainty in the prediction of precipitation and streamflow in the Nile Basin. An evaluation of differences in precipitation δ2H and δ18O values through linear regression and distribution analysis indicate variation by region and season in the isotopic signal of precipitation across the Nile Basin. The White Nile Basin receives precipitation with a more depleted isotopic signal compared to the Blue Nile Basin. The hot temperatures of the Sahelian spring produce a greater evaporation signal in the precipitation isotope distribution compared to precipitation in the Sahara/Mediterranean region, which can be influenced by storms moving in from the Mediterranean Sea. The larger evaporative effect is reversed for the two regions in summer because of the cooling of the Sahel from inflow of Indian Ocean monsoon moisture that predominantly influences the climate of the Blue Nile Basin. The regional precipitation isotopic signals convey to each region's streamflow, which is further modified by additional evaporation according to the local climate. Isotope ratios for White Nile streamflow are significantly altered by evaporation in the Sudd, but this isotopic signal is minimized for streamflow in the Nile River during the winter, spring and summer seasons because of the flow dominance of the Blue Nile. During fall, the contribution from the White Nile may exceed that of the Blue Nile, and the heavier isotopic signal of the White Nile becomes apparent. The variation in climatic conditions of the Nile River Basin provides a means of identifying mechanistic processes through changes in isotope ratios of hydrogen and oxygen, which have utility for separating precipitation origin and the effect of evaporation during seasonal periods. The existing isotope record for precipitation and streamflow in the Nile Basin can be used to evaluate predicted streamflow in the Nile River from a changing climate that is expected to induce further changes in precipitation patterns across the Nile Basin.  相似文献

Phytosterols (PS) lower low density lipoprotein (LDL) levels by as much as 10–14% in normal and hypercholesteromic people. High LDL levels in the blood are associated with an increased risk of coronary heart disease. Wheat germ and bran contain significant amounts of PS. The main objective of this study was to evaluate the effect of genotype and environment on PS content and composition in whole wheat grain. Protein and ash contents and mineral composition of the samples were also determined. Grain samples were collected from three cultivars, Jagger, Trego, and Intrada, grown at three locations, Alva, Balko, and Goodwell, OK in 2005. Irrigated and dryland samples were obtained from Goodwell. Total PS content and composition in the grain samples were determined using a gas chromatography system. Whole wheat grain samples varied in PS content from 202 mg/kg to 355 mg/kg. β‐Sitosterol, campesterol, and stigmasterol were the major PS compounds found in all the samples. PS exhibited a significant location‐by‐cultivar random effect (P < 0.048), indicating the presence of genotype‐by‐location interaction. Within each location, a significant cultivar effect was also observed. Protein and ash contents and mineral composition of the samples, except those collected from Goodwell‐dryland, were within the range published in the literature for other wheat cultivars. A fundamental understanding of compositional variation in wheat grain requires multi‐environment testing of genotypes, perhaps over several years. This study is a first step for achieving this goal.  相似文献

In honeybee, Apis mellifera, colonies infested with larval and adult small hive beetles (SHB), hive material, and in particular honey, tends to ferment, probably due to SHB-associated yeasts such as the predominant Kodamaea ohmeri. Here, we test the effects of organic acids on K. ohmeri and on SHB-infested honey/pollen combs. Organic acids were applied at standard concentrations used by beekeepers to control other pests. In laboratory tests, the growth of K. ohmeri was significantly inhibited by lactic, formic and acetic acids. Treatments of SHB-infested honey/pollen combs (N = 18 colonies) with acetic acid significantly increased mortality of adult SHB and treatments with formic acid significantly reduced larval infestation. Our data suggest that treatment of honeybee colonies and storage rooms with organic acids could also help in reducing SHB damage.  相似文献

Many new coleus (C. hybridus cv.) cultivars are vegetatively propagated and require different fertilization practices from seed propagated cultivars. Two experiments were conducted to evaluate growth responses of Henna, Indian Summer, Mint Mocha, New Orleans Red, Red Head, and Trusty Rusty to 0, 70, 140, 280 and 420 mg·L?1 nitrogen (N), and Henna, Mint Mocha, Red Head, and Trusty Rusty to 0, 6.2, 12.4, 24.8, or 49.6 mg·L?1 phosphorus (P) to determine optimum constant liquid feed rates to produce marketable size plants from rooted cuttings. Positive growth responses in terms of biomass were found with increasing N rates but not P rates. For medium-sized cultivars such as Henna, Indian Summer, and New Orleans Red, quadratic responses were found in aboveground biomass, and N at 280 mg·L?1 resulted in similar plant size and dry weight as those fertilized at 140 and 420 mg·L?1 N. For large-sized cultivars such as Mint Mocha, Red Head and Trusty Rusty, plant dry weight responded linearly within the N range tested and were greatest at 420 mg·L?1. However, plant visual quality was negatively affected by N rates at 280 and 420 mg·L?1 in that, leaf color became less intense at these high N rates. Plants fertilized at 70 mg·L?1 were smaller than those fertilized at 140 mg·L?1, however, they received similar visual quality ratings because of more intense leaf color. Therefore, N at 70 to 140 mg·L?1 can be used to grow most vegetative coleus for similar marketable quality. Mint Mocha and Henna were the only cultivars responded to P treatments that, 12.4 mg·L?1 P rate resulted in greater biomass than the no-P control. All other cultivars had no response to supplemental P except a linear response in tissue P%. Therefore, supplemental P is not required during the 8 week production period when there is an initial P charge in the substrate. We found that substrate pH decreases with higher P rates, therefore supplemental P fertilizer can be used for adjusting pH. Both N and P rates found optimum in this study are lower than current industry practices (N at 150 to 250 mg·L?1 and P at 24 mg·L?1) and can significantly lower production cost and potential leaching of excessive nutrient into waterway. Nutrient treatments in further study on postharvest performance of vegetative coleus will be selected based on this study.  相似文献

Non-target impacts of pesticides are well documented in crops such as soybeans and corn. However, little effort has been made to evaluate these impacts in potatoes. Field trials were conducted at the Parma and Aberdeen Research and Extension Centers during 2005 and 2006 to evaluate the potential for systemic insecticides, fungicides, and herbicides to affect disease development, crop injury, and tuber yield. The first set of trials evaluated six insecticide treatments (aldicarb, oxamyl, phorate, imidacloprid, thiamethoxam, and non-treated) in a factorial combination with in-furrow application of the fungicide azoxystrobin. Azoxystrobin significantly decreased Rhizoctonia stem canker, while aldicarb and phorate tended to increase it compared to the non-treated control. There was a significant insecticide by fungicide interaction due to a bigger reduction in disease index by the fungicide when certain insecticides were used. Despite the association of these insecticides with increased disease incidence, there was no significant impact on yield. In the second set of trials, sulfentrazone herbicide was applied just prior to plant emergence to potatoes treated with the same six insecticides to evaluate the potential for the in-furrow treatments to increase plant injury. Sulfentrazone caused stunting and visible plant injury, but the level of plant injury was influenced by insecticide treatment in only one out of four trials.  相似文献
