Cercospora leaf spot (CLS) poses a high economic risk to sugar beet production due to its potential to greatly reduce yield and quality. For successful integrated management of CLS, rapid and accurate identification of the disease is essential. Diagnosis on the basis of typical visual symptoms is often compromised by the inability to differentiate CLS symptoms from similar symptoms caused by other foliar pathogens of varying significance, or from abiotic stress. An automated detection and classification of CLS and other leaf diseases, enabling a reliable basis for decisions in disease control, would be an alternative to visual as well as molecular and serological methods. This paper presents an algorithm based on a RGB‐image database captured with smartphone cameras for the identification of sugar beet leaf diseases. This tool combines image acquisition and segmentation on the smartphone and advanced image data processing on a server, based on texture features using colour, intensity and gradient values. The diseases are classified using a support vector machine with radial basis function kernel. The algorithm is suitable for binary‐class and multi‐class classification approaches, i.e. the separation between diseased and non‐diseased, and the differentiation among leaf diseases and non‐infected tissue. The classification accuracy for the differentiation of CLS, ramularia leaf spot, phoma leaf spot, beet rust and bacterial blight was 82%, better than that of sugar beet experts classifying diseases from images. However, the technology has not been tested by practitioners. During a previous longitudinal study, performed on four farrow-to-finish farms (A to D), samples were taken from twelve sows, their offspring, and the environment on various occasions over six months to study the MRSA presence. During the present study, a selection of the obtained MRSA isolates were typed by multiple-locus variable-number tandem-repeat analysis (MLVA), Pulsed Field Gel Electrophoresis (PFGE), spa typing, and SCCmec typing to study the genetic diversity of LA-MRSA isolates and to determine possible MRSA sources for pig(let)s. PFGE, spa typing, and SCCmec typing revealed the presence of one or few dominant genotype(s) per farm. In contrast, 212 MLVA types were detected on the four farms, forming one cluster on farm A, three on farm B, four on farm C and two on farm D. The genotype, found on farm A was unique for this farm. Farms B, C and D shared one cluster. In general, MLVA types from these clusters were isolated from piglets, sows, and the environment on various sampling events. Piglets carried MLVA types both related and unrelated to their mother sows’ MLVA types at farrowing and onwards. In conclusion, molecular typing revealed that within a farm one or a few dominant strain(s) are widespread. Potential MRSA sources for piglets were mother sows, the environment and other piglets.

From September 1995 to February 2001, blood samples were collected from 865 neotropical felids belonging to 8 different species. These animals were housed in 86 institutions located in 78 cities of 20 Brazilian states. Our goal was to identify the risk factors associated with sero-positivity to Toxoplasma gondii in captive neotropical felids from Brazil. All serum samples were tested by the modified agglutination test (MAT), using formalin-fixed whole tachyzoites and mercaptoethanol. For each animal an individual questionnaire was filled with questions about tattoo number, felid species, age, sex, origin, number of animals in the group, introduction of new animals in the group, time in the institution, eating meat previously frozen for a period <7 days in the last 6 months, eating meat of run-over or euthanized animals in the last 6 months, predation of rodents or birds in the last 6 months and presence of domestic cats near the enclosures in the last 6 months. The total sero-prevalence was 55% (95% CI: 52%, 57%). We estimated a prevalence of 46% (95% CI: 40%, 54%) for jaguarundi (Puma yagouaroundi); 58% (95% CI: 53%, 63%) for ocelot (Leopardus pardalis); 50% (95% CI: 45%, 56%) for oncilla (L. tigrinus); 54% (95% CI: 46%, 62%) for margay (L. wiedii); 12% (95% CI: 4%, 31%) for Pampas-cat (L. colocolo); 83% (95% CI: 65%, 93%) for Geoffroy's-cat (L. geoffroyi); 64% (95% CI: 50%, 68%) for jaguar (Panthera onca) and 48% (95% CI: 42%, 54%) for puma (Puma concolor). Multiple logistic regression was used to evaluate the association between the variables in the questionnaire and sero-positivity to T. gondii. We concluded that the independent risk factors for toxoplasmosis were: age >3 years (OR = 4.75 [2.75; 8.2]), eating meat previously frozen for a period <7 days (OR = 2.23 [1.24; 4.01]), and consumption of animals that were run-over or euthanized (OR = 1.64; [1.14; 2.37]). 相似文献
This study assesses the potential of two contrasted fragrant Pelargonium cultivars to induce pH and dissolved organic carbon (DOC) changes in the soil solution, Pb speciation, and their subsequent effects on rhizosphere phytoavailable Pb.
Materials and methods
Rooted plantlets were grown in special devices, floating on aerated nutrient solution in PVC tanks. This setup allows roots to be physically separated, through a mesh, from a 3-mm soil matrix layer that can be considered as rhizosphere soil. Two contrasted soils, each spiked with Pb-rich particles, emitted from a battery recycling industry, were used at total burdens of 500 and 1500 mg Pb kg?1 in addition to a control unspiked soil. Soil solution pH, phytoavailable Pb, DOC, Pb adsorption, precipitation on roots, and Pb phases in soil and plant were investigated.
Results and discussion
Attar of Roses (Attar) cultivar acidified its rhizosphere by 0.4 pH units in both spiked soils. Concolor Lace (Concolor) was unable to change soil solution pH on soil-1 and increased it by 0.7 units on soil 2. Concentrations of Pb in soil solution from Attar plants were always higher than those of Concolor ones. DOC contents of both unspiked soil-1 and soil-2 without plants were not significantly different. In the case of spiked samples, DOC contents in the rhizosphere soil were increased by three and two times for Attar and Concolor, respectively, compared to the unspiked soil without plant. Both cultivars were able to increase DOC contents, independent of soil type and level of contamination. Accumulation of Pb in shoots and roots was higher in Attar as compared to Concolor due to enhanced available Pb as a result of pH and DOC modifications of the rhizosphere soil. Significant amounts of Pb were adsorbed on roots of both cultivars. X-ray elemental analysis of precipitates on roots revealed the association of Pb with P in cylinder-like structures. Extended X-ray absorption fine structure (EXAFS) spectroscopy revealed that Pb was present, to a major extent in the inorganic form, mainly as PbSO4 in the soil, whereas it was complexed with organic species within plant tissues. The conversion of Pb into organic species could decrease toxicity, may enhance plant tolerance, and could increase translocation.
Conclusions
Plant-induced changes were responsible for the modification of lead phases within the soil. Immobile forms present in the source leaded particles as well as in the soils were converted into soluble species, ultimately improving the phytoavailable or soil solubilized Pb.
Abstract: An 11‐year‐old female spayed domestic shorthair cat was referred to The Ohio State University Veterinary Teaching Hospital (OSU‐VTH) for evaluation of a 6 × 4 × 3.5 cm mass in the left midcervical region causing increased respiratory sounds and lateral deviation of the trachea. A fine needle aspirate of the mass was obtained before referral and the cytology results were compatible with a reactive lymph node. Immunocytochemistry showed increased numbers of CD3+ T lymphocytes and small numbers of CD20+ and CD79a+ medium to large lymphocytes. Differential diagnoses from the referral pathologist were T‐cell‐rich B‐cell lymphoma and feline Hodgkin's‐like lymphoma. A subsequent fine needle aspirate performed at the OSU‐VTH showed similar results. On flow cytometry the majority of cells were CD3+ T lymphocytes that were double positive for CD4 and CD8 (73%), compatible with either a double‐positive (CD4+CD8+) T‐cell lymphoma or lymphocytes from ectopic thymic tissue. The mass was surgically removed. Histopathology and immunohistochemistry of the mass revealed a predominant population of CD3+ small lymphocytes and small numbers of medium to large lymphocytes with moderate anisocytosis and anysokaryosis. A population of cytokeratin‐positive epithelial cells surrounded small microcystic structures filled with eosinophilic material and structures interpreted as Hassall's corpuscles. These findings were consistent with thymic tissue and a diagnosis of ectopic thymoma was made. PCR results for lymphocyte antigen receptor rearrangement (PARR) were negative. The cat had no evidence of disease 16 months after removal of the mass. To our knowledge this is the first report of an ectopic cervical thymoma in a cat. The clinical and diagnostic features of this unusual case will be useful in helping veterinarians and pathologists obtain a presurgical diagnosis and establish a prognosis for similar lesions. 相似文献
Oxidative stress represents a challenge during sperm manipulation. We have tested the effect of increasing hydrogen peroxide (H2O2) levels on red deer spermatozoa after cryopreservation, and the role of male‐to‐male variation in that response. In a first experiment, eight thawed samples were submitted to 0, 25, 50, 100 and 200 μm H2O2 for 2 h at 37°C. Intracellular reactive oxygen species (H2DCFDA‐CM) increased with H2O2 concentration, but we only detected a decrease in sperm function (motility by CASA and chromatin damage by sperm chromatin structure assay) with 200 μm . Lipoperoxidation assessed by the thiobarbituric acid reactive substance (TBARS) method increased slightly with 50 μm H2O2 and above. In a second experiment, samples from seven males were submitted to 0 and 200 μm H2O2 for 2 h, triplicating the experiment within each male. Males differed at thawing and regarding their response to incubation and H2O2 presence. We found that the kinematic parameters reflected male‐to‐male variability, whereas the response of the different males was similar for lipid peroxidation and viability. A multiparametric analysis showed that males grouped differently if samples were assessed after thawing, after incubation without H2O2 or after incubation with H2O2. Red deer spermatozoa are relatively resilient to H2O2 after thawing, but it seems to be a great male‐to‐male variability regarding the response to oxidative stress. The acknowledgement of this individual variability might improve the development of optimized sperm work protocols. 相似文献
A waxy spring wheat (Triticum aestivum L.) genotype was fractionated into flour and starch by roller and wet‐milling, respectively. The resultant flour and starch were evaluated for end‐use properties and compared with their counterparts from hard and soft wheats and with commercial waxy and nonwaxy corn (Zea mays L.) starches. The waxy wheat flour had exceptionally high levels of water absorption and peak viscosity compared with hard or soft wheat flour. The flour formed an intermediate‐strength dough that developed rapidly and was relatively susceptible to mixing. Analysis by differential scanning calorimetry and X‐ray diffractometry showed waxy wheat starch had higher gelatinization temperatures, a greater degree of crystallization, and an absence of an amylose‐lipid complex compared with nonwaxy wheat. Waxy wheat and corn starches showed greater refrigeration and freeze‐thaw stabilities than did nonwaxy starches as demonstrated by syneresis tests. They were also similar in pasting properties, but waxy wheat starch required lower temperature and enthalpy to gelatinize. The results show analogies between waxy wheat and waxy corn starches, but waxy wheat flour was distinct from hard or soft wheat flour in pasting and mixing properties. 相似文献
