To compare the genetic susceptibility of elk (Cervus elaphus nelsoni) with various alleles of the PRNP gene, which encodes the normal cellular prion protein, to chronic wasting disease (CWD), eight 8-month-old elk calves of 3 genotypes (2 132MM, 2 132LM, and 4 132LL) were orally dosed with CWD-infected brain material from elk. During postinoculation (PI) month 23, both 132MM elk had lost appetite, developed clinical signs of weight loss and central nervous system (CNS) dysfunction, and were euthanized. Two other elk (both 132LM) developed similar clinical signs of disease and were euthanized during PI month 40. All 4 affected elk had microscopic lesions of spongiform encephalopathy (SE), and PrPres, the disease-associated form of the prion protein, was detected in their CNS and lymphoid tissues by use of immunohistochemical (IHC) and Western blot (WB) techniques. These findings indicate that elk with MM and LM at codon 132 are susceptible to orally inoculated CWD. All 4 LL elk are alive at PI year 4 and are clinically normal, which suggests that 132LL elk may have reduced susceptibility to oral infection with CWD-infected material or may have prolonged incubation time.  相似文献

Radionuclide ventriculography has been used in humans to evaluate valvular incompetency. The stroke volume ratio, derived from the radionuclide ventriculogram, is used to quantify the severity of mitral regurgitation (MR). Previous studies conducted in humans have shown that left to right stroke volume ratio increases as the severity of MR increases. In this study, we evaluated radionuclide ventriculography as a noninvasive method to detect MR in dogs with surgically created mitral insufficiency. Six male and three female adult, conditioned mongrel dogs were used. Scintigraphic studies were performed prior to and 4 weeks after surgically created MR. Because of the overlap of the left and right ventricles when viewed from a left lateral position, we combined data from a first-pass radionuclide angiocardiogram with the radionuclide ventriculogram to obtain a corrected stroke volume ratio. Blood flow transit parameters were also derived from the first-pass radionuclide angiocardiogram. Standard left ventricular functional indices were also measured from the radionuclide ventriculogram. On the left lateral view of the heart, 25 to 30% of the right ventricular volume overlaps the left ventricle. After correcting for the overlap, the stroke volume ratio of normal dogs was 1.17±0.178 (mean±SD), which increased to 2.06±0.41 (mean±SD) (p<.001) 4 weeks after creation of MR. The was no significant change in left ventricular ejection fraction or peak rate of ejection following MR. The transit times of blood through the left ventricle were measured from the first-pass radionuclide angiocardiogram and were expressed as half-time clearance, peak clearance rate, and time to peak clearance rate. The baseline half-time clearance was 2.07±0.71 s (mean±SD), which increased to 6.70±4.89 s (mean±SD) (p=.02) after creation of MR. The baseline peak clearance rate was 49.75±8.96 cts/s (mean±SD), which decreased to 23.12±6.84 cts/s (mean±SD) (p<.001) after creation of MR. Stroke volume ratios significantly increased following creation of MR. Blood flow transit through the left ventricle slowed following creation of MR. The variability of these parameters were small in the baseline studies, suggesting these techniques may be clinically useful to gauge the severity of MR in dogs.  相似文献

Transportation and handling stress of white suckers raised in cages   总被引:1,自引:0,他引:1  
Changes in plasma cortisol concentrations were measured in juvenile white suckers, Catostomus commersoni (Lacepede), as a measure of their response to transportation (8 h) and chase challenge stressors. The study attempted to evaluate the ability of this species to withstand standard aquacultural husbandry practices. Plasma cortisol concentrations were elevated during and following transportation. Recovery from transportation (as plasma cortisol levels reached baseline values) was prolonged in fish transported under winter conditions compared with fish transported during the summer months. The plasma cortisol response to a 5 min chase challenge in well-fed juvenile white suckers was typically of 2 to 3 h periodicity with the peak values between 15 and 30 min. This pattern was similar in fish fed three commercial diets (although rates of recovery differed), and was unaffected by fasting. The return of plasma cortisol concentrations to baseline values following the chase-challenge stressor was prolonged in fasted groups. The results of this study showed that juvenile white suckers require several days to recover from transportation and under the acclimation conditions applied, the recovery was faster in the fish transported during the summer.  相似文献

Influenza is a viral disease that affects human and several animal species. In Brazil, H1N1, H3N2 and 2009 pandemic H1N1 A(H1N1)pdm09 influenza A viruses (IAV) circulate in domestic swine herds. Wild boars are also susceptible to IAV infection but in Brazil until this moment there are no reports of IAV infection in wild boars or in captive wild boars populations. Herein the occurrence of IAV in captive wild boars with the presence of lung consolidation lesions during slaughter was investigated. Lung samples were screened by RT-PCR for IAV detection. IAV positive samples were further analyzed by quantitative real-time PCR (qRRT-PCR), virus isolation, genomic sequencing, histopathology and immunohistochemistry (IHC). Eleven out of 60 lungs (18.3%) were positive for IAV by RT-PCR and seven out of the eleven were also positive for A(H1N1)pdm09 by qRRT-PCR. Chronic diffuse bronchopneumonia was observed in all samples and IHC analysis was negative for influenza A antigen. Full genes segments of H1N2 IAV were sequenced using Illumina's genome analyzer platform (MiSeq). The genomic analysis revealed that the HA and NA genes clustered with IAVs of the human lineage and the six internal genes were derived from the H1N1pdm09 IAV. This is the first report of a reassortant human-like H1N2 influenza virus infection in captive wild boars in Brazil and indicates the need to monitor IAV evolution in Suidae populations.  相似文献

In an attempt to identify appropriate diets for use in intensive baitfish culture, the efficacy of three commercial diet formulations, a high-protein, high-lipid, low-carbohydrate diet [a semi-moist salmonid diet (SD)] and two low-protein, low-lipid, high-carbohydrate diets [a catfish diet (CD) and a tilapia diet (TD)] on growth performance of juvenile white suckers, Catostomus commersoni, was examined. All three diets permitted growth during the 16 week trial. The SD promoted highest growth and somatic protein and lipid deposition, whereas the CD and TD formulations promoted markedly higher accumulations of glycogen in the carcass and liver. The high hepatic glycogen content was associated with an elevated hepatosomatic index in the CD- and TD-fed groups. Plasma T3 levels were lower in the CD- and TD-fed groups, possibly a response to the low dietary protein or indicative of dietary insufficiency in these fish. Of the three diets evaluated, the SD was considered to be the best in terms of the growth performance parameters measured, and offers an economical and appropriate diet for intensive culture of juvenile white suckers.  相似文献

This research examined the haematological and immunological responses of quadruplicate groups of juvenile (~400 g initial weight) Atlantic salmon (Salmo salar L.) that had each been fed daily to satiation for 12 weeks one of three high‐energy extruded diets of identical composition except for the supplemental dietary lipid (234.7 g kg?1) source. The three experimental diets varied in the composition of supplement lipid; diet 1 contained 100% anchovy oil (AO), while diets 2 and 3 replaced 29.8% and 59.7% of the AO (respectively) with a 1:1 blend of canola oil (CO) and poultry fat (PF). Immediately following the feeding trial, a random sample of fish from each diet was sampled for determination of baseline levels of various haematological and immunological parameters. Thereafter, duplicate diet groups were vaccinated (against Listonella anguillarum) and reared on their respective experimental diets for an additional 4 weeks. At that time, the remaining fish were sampled similarly, and the different parameters were measured again. Comparisons between the different diet treatment groups were made before and after vaccination. There were no significant diet treatment effects at either sample time, for haematocrit, differential leucocyte counts, erythrocyte counts, serum hemolytic activity or head kidney leucocyte respiratory burst activity. The fish fed diet 1 however, did show significantly higher post‐vaccination levels of peripheral blood leucocyte respiratory burst activity and higher serum antibody titres against L. anguillarum. The results suggest that the relatively low n‐6/n‐3 fatty acid ratios in the muscle and presumably other tissues of fish fed diet 1, may have resulted in a reduced production of immunocompromising eicosanoids than were produced in fish ingesting the other two diets that were based in part on the different amounts of the CO and PF blend. Long‐term studies are required to confirm this possibility.  相似文献

Abstract.— The aquaculture performance of mutton snapper Lutjanus analis raised in floating net cages was assessed by measuring their growth, survival, and feed conversion rates during a growout trial conducted in a 3.2‐ha saltwater lake in the Florida Keys, Florida, USA. Approximately 10,500 hatchery‐reared finger‐lings were stocked in two circular, high‐density polyethylene (HDPE) net cages of 7‐m diameter × 7‐m deep (300 m2) and 10‐m diameter × 7‐m deep (600 m3) dimensions. Cages were stocked at 25 fish/m3 (3.2 kg/m3) and 5 fish/m3 (0.72 kg/m3), respectively. Fish grew from a mean of 16.5 g to 302.8 g (25.6 cm TL) in 246 days in the former cage and from a mean of 42.3 g to 245.6 g (23.8 cm TL) in 178 d in the latter cage. Growth rates in weight were best expressed by the following exponential equations: cage 1 (high stocking density): W = 20.716 e0.0112x (r2= 0.83); cage 2 (low stocking density): W = 38.848 e0.0118x (r2= 0.81). Length‐weight data indicate that hatcheryraised, cage‐cultured mutton snapper are heavier per unit length than their wild counterparts. There was no significant difference (P < 0.05) between the slopes of the two lines, indicating that fish in the two cages grew at the same rate. The length‐weight relationships for mutton snapper stocked in cages 1 and 2 are expressed, respectively, by the equations W = 0.000009 L 3.11 (r2= 0.99) and W = 0.000005 L 3.22 (r2= 0.97). Overall feed conversion rate for both cages combined was 1.4. Approximately 10% of the fish sampled exhibited some degree of deformity, particularly scoliosis. Overall survival rate was 70%. Results suggest that L. analis has potential for aquaculture development in net cage systems.  相似文献
