Heifers managed under pastoral conditions are at risk from Streptococcus uberis mastitis infections at calving. A total of 397 heifers from six farms around New Zealand were enrolled in a study to identify and enumerate S. uberis on teat-ends of heifers in the peri-partum period, and to understand the effect of teat-spraying in the pre-calving period on the prevalence and incidence of S. uberis mastitis post-calving. Heifers were randomly assigned to Control or Sprayed groups. Sprayed heifers were teat-sprayed once, three times a week (Monday, Wednesday and Friday) with a commercial iodine-based teat sanitizer, starting at 3 weeks prior to calving and ending at day of calving. Across three farms, all glands of cows in both groups were sampled at calving to determine S. uberis intra-mammary infection (IMI) prevalence. For all farms, clinical mastitis (CM) cases detected during the week after calving were sampled and submitted for bacteriological analysis. Swabbing of teat-ends of 54 heifers from one farm showed that heifers had a pre-existing S. uberis contamination averaging 610 colony-forming units per swab (cfu/swab), at 3 weeks prior to calving. At calving, teat-end contamination was 560 cfu/swab for Sprayed heifers and 1775 cfu/swab for Control heifers. Two weeks after calving, teat-end contamination was similar between both groups, at 30 cfu/swab. The prevalence of S. uberis IMI was significantly lower in the Sprayed (3.5% glands) vs. the Control (7.4%) heifers in the first week after calving. There was a trend for Sprayed heifers (3.6% heifers) to have a lower incidence of S. uberis CM compared with Control heifers (7.4% heifers). Recurrent airway obstruction (RAO) is a chronic inflammatory condition in equine lung, which may share a common immunological basis with human asthma, in which dysregulated Th2 responses occur. Mammals express chitinases and chitinase-like proteins, two of which are active enzymes, chitotriosidase and acidic mammalian chitinase (AMCase). Both enzymes are upregulated in a range of inflammatory conditions, including asthma. We investigated the activity of chitinase in bronchoalveolar lavage fluid from horses with and without RAO in response to organic dust challenges. No significant differences were found in activity, although in one study RAO animals had elevated chitinase activity that fell short of statistical significance. The pH optimum and pH lability of the activity was consistent with the presence of chitotriosidase. RT-PCR amplification of the mRNA encoding chitotriosidase and AMCase in normal equine lung showed that chitotriosidase, but not AMCase, is expressed in trachea, bronchi, and peripheral lung tissue. The gene for chitotriosidase was identified from the Equus caballus (horse) genome 1.1 database and its similarity to the same genes from other species was determined. Azaspiracid (AZA) poisoning has been reported following consumption of contaminated shellfish, and is of human health concern. Hence, it is important to have sustainable amounts of the causative toxins available for toxicological studies and for instrument calibration in monitoring programs, without having to rely on natural toxin events. Continuous pilot scale culturing was carried out to evaluate the feasibility of AZA production using Azadinium spinosum cultures. Algae were harvested using tangential flow filtration or continuous centrifugation. AZAs were extracted using solid phase extraction (SPE) procedures, and subsequently purified. When coupling two stirred photobioreactors in series, cell concentrations reached 190,000 and 210,000 cell · mL(-1) at steady state in bioreactors 1 and 2, respectively. The AZA cell quota decreased as the dilution rate increased from 0.15 to 0.3 day(-1), with optimum toxin production at 0.25 day(-1). After optimization, SPE procedures allowed for the recovery of 79 ± 9% of AZAs. The preparative isolation procedure previously developed for shellfish was optimized for algal extracts, such that only four steps were necessary to obtain purified AZA1 and -2. A purification efficiency of more than 70% was achieved, and isolation from 1200 L of culture yielded 9.3 mg of AZA1 and 2.2 mg of AZA2 of >95% purity. Climate change spurs havoc on social-ecological system. People and places vulnerable to climate change have been the focus of many discussions. However, in the forestry sector, limited studies have been conducted that link human vulnerability to recent initiatives, such as reducing emissions from deforestation and forest degradation or REDD+, especially in highly vulnerable countries. Using case studies from the Philippines, this paper focuses on the vulnerability of two community-based forest management (CBFM) organizations, with and without REDD+ readiness intervention, to show the impacts of climate variability and extremes. Two balance-weighted approaches, the livelihood vulnerability index and the IPCC-framework, were used in the vulnerability assessments. Results revealed high vulnerability of both CBFM organizations, although Malitbog Upland Developers for Sustainable Association (MUDSA) livelihood vulnerability is largely aggravated by its exposure to disaster, climate variability and extremes, accessibility to health facilities and water supply. The long history of CBFM implementation in both organizations have minimal contribution in enhancing the adaptive capacity of members to cope with and adapt to climate change impacts. REDD+ (readiness), does not insure reduced vulnerability to climate change, unless sustainable livelihood is achieved. As the State controls forest resources, the rights of CBFM-organizations to commercially benefit from these resources are limited, a disincentive to the development of new and viable development programs in CBFM areas, such as REDD+. Tropical forests worldwide are undergoing rapid changes due to increasing human populations and varied land use practices. In an effort to protect these forests and their species, the number of protected areas has increased exponentially in recent decades. Clearly protected areas play a crucial role in conservation efforts and this strategy has been found to be successful in several studies. However, protected areas are often embedded within a matrix of other land cover types, including non-protected forests, agricultural fields, or urban areas. It is therefore critical for park management to think beyond the borders of their protected areas and work to maintain ecological integrity of the surrounding matrix and the connectivity between protected areas. This study integrates remote sensing, GIS and field observations to examine the effects of different management strategies on land cover changes. As a case study we examine a tiger reserve in Central India which is surrounded by commercially managed forests, representative of many protected areas throughout India. Findings of this study show that while protected areas play an important role in conservation, alternative approaches including favorable management policies could also be effective to extend conservation over larger areas. 相似文献
