Tendons regenerate poorly due to a dense extracellular matrix and low cellularity. Cellular therapies aim to improve tendon repair using mesenchymal stem cells and tenocytes; however, a current limitation is the low proliferative potential of tenocytes in cases of severe trauma. The purpose of this study was to develop a method useful in veterinary medicine to improve the differentiation of Peripheral Blood equine mesenchymal stem cells (PB-MSCs) into tenocytes. PB-MSCs were used to study the effects of the addition of some growth factors (GFs) as TGFβ3 (transforming growth factor), EGF2 (Epidermal growth factor), bFGF2 (Fibroblast growth factor) and IGF-1 (insulin-like growth factor) in presence or without Low Level Laser Technology (LLLT) on the mRNA expression levels of genes important in the tenogenic induction as Early Growth Response Protein-1 (EGR1), Tenascin (TNC) and Decorin (DCN). The singular addition of GFs did not show any influence on the mRNA expression of tenogenic genes whereas the specific combinations that arrested cell proliferation in favour of differentiation were the following: bFGF2 + TGFβ3 and bFGF2 + TGFβ3 + LLLT. Indeed, the supplement of bFGF2 and TGFβ3 significantly upregulated the expression of Early Growth Response Protein-1 and Decorin, while the use of LLLT induced a significant increase of Tenascin C levels. In conclusion, the present study might furnish significant suggestions for developing an efficient approach for tenocyte induction since the external administration of bFGF2 and TGFβ3, along with LLLT, influences the differentiation of PB-MSCs towards the tenogenic fate. 相似文献
Dehorning adult cattle is a surgical procedure causing distress of varying intensities that can be reflected in behavioural changes and alterations in plasma cortisol levels. Stress responses during the dehorning process were evaluated in 18 Red Pied cows. The cows were divided into 3 groups of six and kept in tie-stall housing. Those in the first group were dehorned under general anaesthesia (GA) induced by intravenous administration of xylazine and ketamine. The second group was dehorned under sedation and local anaesthesia (SLA) induced by intramuscular administration of xylazine and local anaesthesia with lidocaine. The third group was dehorned under local anaesthesia (LA) with lidocaine. Dehorning was performed with a foetotomy wire. Blood samples were taken 0.5 h before dehorning to determine cortisol levels, and, by means of a central venous catheter inserted into the jugular vein, during surgery at 0, 0.5, 1, 1.5, 2, 2.5, 3, 3.5, 4, 5, 6, 7 and 8 h post-surgery. Concurrently, occurrence of stress behaviours was assessed. Cortisol levels were measured by enzyme immunoassay (EIA). The lowest mean peak levels of plasma cortisol (82.53 +/- 6.04 nmol l(-1)), the most rapid return of plasma cortisol levels to baseline values (1.92 +/- 1.11 h), and the lowest occurrence of stress behaviours (2.38 +/- 5.83%) were noted in the SLA group. The highest mean peak levels plasma cortisol (113.86 +/- 25.65 nmol l(-1)), the slowest return of plasma cortisol levels to baseline values (3.83 +/- 2.18 h) and the most frequent occurrence of stress behaviours (65.48 +/- 28.72%) were observed in the LA group. There were significant differences between the SLA and LA groups in peak plasma cortisol levels (p = 0.011) and in occurrence of stress behaviours (p = 0.003). Sedation induced by intramuscular administration of xylazine in conjunction with local anaesthesia with lidocaine is considered the most suitable method of anaesthesia when dehorning adult cattle. During the last few years, many Danish dairy farmers have expressed increasing concerns regarding a group of cows, which we have chosen to term ‘loser cows’. Until now, a loser cow has not been described scientifically. We defined a loser cow on the basis of a clinical examination of the cow. A total of 15,151 clinical examinations were made on 6,451 individual cows from 39 randomly selected, large Danish dairy herds with loose-housing systems using a clinical protocol. Scores for the clinical signs lameness, body condition, hock lesions, other cutaneous lesions, vaginal discharge, condition of hair coat and general condition were converted into a loser cow score. Cows with a loser cow score of 8 or more were classified as loser cows. The overall prevalence of loser cows was 2.15%, 4.50% and 2.98% during the first, second and third round of herd visits, respectively.
The associations between the loser cow state and milk production, mortality, morbidity, culling and workload for the farmer were evaluated using data from herd visits and from the Danish Cattle Database and a number of different statistical techniques. It was concluded that the loser cow state has significant negative consequences for both the farmer and the cow. On average, loser cows yielded 0.61 to 2.24 kg energy corrected milk less per day than non-loser cows depending on parity. Hazard ratio for death or euthanasia was 5.69 for loser cows compared to non-loser cows. Incidence rate ratio for disease treatments was 0.69 for non-loser cows compared to loser cows. Loser cows were often culled in an ‘unfavourable’ way and generally caused extra workload for the farmer.
A simplified version of the loser cow score was evaluated and is recommended for future research and use in practice. 相似文献
Summary A simulation model capable of predicting the yield response of corn to a limited water supply was developed by combining two existing mathematical models. The resulting computer model was evaluated using experimental data taken under a wide range of soil moisture conditions. The soil profile water balances was simulated using SWATRE and SUCROS was used to model the crop growth in response to environmental conditions. In addition to the integration of the two existing models, some minor changes were made to each in an effort to improve the accuracy of the combined models. The model input parameters were derived entirely from published literature. The experimental data necessary for model validation were available from irrigation studies at the Sandhills Agricultural Laboratory of the University of Nebraska. These experiments not only provided the required input soil and climatic data, but also the observed irrigation levels, soil moisture distributions and crop yield required for model validation. Initial evaluation of the computer model indicates that the combined model adequately describes crop evapotranspiration, soil moisture extraction and crop yield under a fairly wide range of soil moisture stress. Additional modifications for the prediction of leaf area expansion and senescence, especially under moisture stress, are needed to improve the accuracy of the model. 相似文献
The inability to properly maintain irrigation systems over time forced the Government of Indonesia (GOI) to seek cost recovery from water users through introduction of an irrigation service fee. The plan is to introduce this fee in all of the technical irrigation systems of Indonesia, covering about 4 million hectares, over a 12 year period. Design and Introduction of this service fee in 4 pilot areas of the 4 major rice producing provinces (West, Central and East Java, and South Sulawesi) during 1989–1991 has shown that users are willing to pay. Results of first ISF collection from over 11,000 farmers in Central and East Java in pilot systems was an encouraging 95%. However that acceptance to pay by users depends on their structured and systematic involvement in defining systems needs. They need to understand a differentiation in payment if service levels are different. They appreciate use of collected funds in the system where collected. They understand the organization of water users associations but this requires their active role in fee determination and collection. They understand the establishment of federations of these associations. ISF requires also active involvement of Local Government officials as facilitators and intermediaries between service receivers (the water users) and service providers (personnel of the Public Works irrigation department) in the introduction period. This article describes the experience of the first 21 months of the ISF project, the concept developed, the principles used as basis for ISF, the introduction at the field level, issues related to acceptance by users and by the institutions involved, and the first results.Abbreviations/Acronyms DGWRD

Hyperspectral remote sensing for monitoring horticultural production systems requires the understanding of how plant physiology, canopy structure, management and solar elevation affect the retrieved canopy reflectance during different stages of the phenological cycle. Hence, the objective of this study was to set up and to interpret a hyperspectral time series for a mature and healthy citrus orchard in the Western Cape province of South Africa considering these effects. Based on the remotely sensed data, biophysical parameters at the canopy level were derived and related to known observed physiological and phenological changes at the leaf level and to orchard management. Fractions of mature fruit, flowers, and sunburnt leaves were considered, and indices related to canopy structure chlorophyll content and canopy water status were calculated.Results revealed small cover fractions of mature fruit, flowers and sunburnt leaves of respectively 2.1%, 3.1% and 7.0%, but the high spectral contrast between flowers and leaves allowed a successful classification of flowering intensity. Furthermore, it was shown that canopy level time series of vegetation indices were sensitive to changes in solar elevation and soil reflectance which could be reduced by applying an empirical soil line correction for the most affected indices. Most trends in vegetation indices at the canopy level could be explained by a combination of changes at the leaf level (chlorophyll, carotenoids, dry matter), changes in canopy structure (leaf area index and leaf angle distribution) and changes in cover fractions of vegetative flushes, flowers and sunburnt leaves. The transformed chlorophyll absorption ratio index over the optimised soil adjusted vegetation index (MCARI/OSAVI) was best related to leaf level trends in chlorophyll content. Seasonal changes in the photochemical reflectance index (PRI) were linked to inverse changes in the carotenoid-to-chlorophyll ratio. Canopy structure indices (the modified triangular vegetation index or MTVI2 and the standardized leaf area index determining index or sLAIDI) were sensitive to changes in leaf area index, average leaf angle as well to management interactions (pruning and harvest). Canopy water status was highly impacted during the spring flush due to expanding leaves that concealed trends in the underlying mature leaves. Seasonal trends in soil and weeds reflectance were related to changes in volumetric soil water content and to the earlier and reduced growth period of non-irrigated weeds. 相似文献
Application of sequential solvent extraction, followed by HPLC combined with the taste dilution analysis, enabled the localization of the most intense velvety astringent, drying, and puckering astringent, as well as bitter-tasting, compounds in red wine, respectively. Isolation of the taste components involving gel adsorption chromatography, ultrafiltration, and synthesis revealed the identification of 26 sensory-active nonvolatiles, among which several hydroxybenzoic acids, hydroxycinnamic acids, flavon-3-ol glycosides, and dihydroflavon-3-ol rhamnosides as well as a structurally undefined polymeric fraction (>5 kDa) were identified as the key astringent components. In contradiction to literature suggestions, flavan-3-ols were found to be not of major importance for astringency and bitter taste, respectively. Surprisingly, a series of hydroxybenzoic acid ethyl esters and hydroxycinnamic acid ethyl esters were identified as bitter compounds in wine. Taste qualities and taste threshold concentrations of the individual wine components were determined by means of a three-alternative forced-choice test and the half-mouth test, respectively. 相似文献
