ABSTRACT The mechanism of the effects of flumorph (a novel fungicide) was investigated by analyzing alterations of hyphal morphology, cell wall deposition patterns, F-actin organization, and other organelles in Phytophthora melonis. Calcofluor white staining suggested that flumorph did not inhibit the synthesis of cell wall materials, but disturbed the polar deposition of newly synthesized cell wall materials during cystospore germination and hyphal growth. After exposure to flumorph, zoospores were able to switch into cystospores accompanied with the formation of a cell wall, whereas cystospores failed to induce the isotropic-polar switch and did not produce germ tubes but continued the isotropic growth phase. In flumorph-treated hyphae, the most characteristic change was the development of periodic swelling ("beaded" morphology) and the disruption of tip growth. Newly synthesized cell wall materials were deposited uniformly throughout the diffuse expanded region of hyphae, in contrast to their normal polarized patterns of deposition. These alterations were the result of F-actin disruption, identified with the fluorescein isothiocynate (FITC)-phalloidin staining. The disruption of F-actin also was accompanied by disorganized organelles: each swelling of subapical hyphae was associated with a nucleus. Vesicles did not undergo polarized secretion to the apical hyphae, but diffused around nuclei for the subapical growth; thus, the cell wall was thickened with periodic expansion along the hyphae. Upon removing flumorph, normal tip growth and organized F-actin were observed again. These data, as well as data published earlier, suggest that flumorph may be involved in the impairment of cell polar growth through directly or indirectly disrupting the organization of F-actin. Ulcerative colitis (UC) is a lifelong illness with profound emotional and social impacts, and could cause serious damage to large intestine, especially in colon. However, the pathogenesis of UC remained unclear. The present study attempts to find out the role of matrix metalloproteinases-7 (MMP-7) and lysozyme in the pathogenesis of UC through a mice model induced by dextran sulfate sodium (DSS). The UC model was evaluated both by disease activity index (DAI) and the intestinal histopathology. The results show that there is a high correlation between the DAI score and the pathological changes of colon. Interleukin-6 (IL-6) serum levels and large intestinal fluids levels in UC mice are always higher than that of the control groups, which might be associated with the degree of the inflammation damage in the colon. The change tendency of the MMP-7 mRNA and protein expressions are both up-regulated firstly and then down-regulated from 1 to 5 d in the colon, but only the MMP-7 protein is up-regulated at 7 d again. The up-regulated MMP-7 levels in the early stage of UC may play a protective role through the activated defensins, while the down-regulated levels in the mid-later stage of UC may be connected with the severe lesions in the colon. However, the up-regulated MMP-7 levels in the later stage of UC in the colon may also contribute to the tissue repair or be served as a marker to CRC (colorectal cancer). The distribution of lysozyme protein indicates that there may be Paneth-like cells in the colon. Both the changes of MMP-7 and lysozyme in the small intestine may play a protective role for the safe environment of the whole gut, especially to the colon of UC. 相似文献

采用PCR-SSCP和聚丙烯酰胺凝胶电泳(PAGE)技术检测类固醇21-羟化酶基因(steroid 21-hydroxylase,CYP21)10个外显子在山羊(Capra hircus)低繁殖力品种(安哥拉山羊和内蒙古绒山羊)、中等繁殖力品种(波尔山羊)和高繁殖力品种(济宁青山羊)中的碱基变异,同时研究该基因对济宁青山羊高繁殖力的影响.结果表明,10对引物中仅引物P10扩增片段存在多态性.对于P10扩增产物,在内蒙古绒山羊、波尔山羊和济宁青山羊中检测到AA、AB和BB基因型,在安哥拉山羊中检测到AA和AB基因型;测序分析发现,BB与AA基因型相比发生了A2789C、C2791A、G2818A、G2851C、G2852T和G2854A的突变,在第2821与2 822位发生了两个碱基CG的缺失突变,导致426~448位共有19个氨基酸发生改变;济宁青山羊BB和AB基因型产羔数最小二乘均值分别比AA基因型的多0.81只(P<0.05)和0.47只(P<0.05),BB和AB基因型之间差异不显著(P<0.05).研究结果初步表明CYP21基因的B等位基因是提高山羊产羔数的一个潜在有效的DNA标记. 相似文献

锦鲤选美的不同等级之间具有高相似度的特点,目前都是人工进行选美分级。为解决人工选美所存在的效率低、主观性强、成本高的问题,提出了一种基于迁移学习和改进ResNeXt50残差网络的锦鲤选美方法。本文首先构建了红白、大正、昭和3种锦鲤的选美等级数据集。其次,采用迁移学习策略提高训练速度,并从SE注意力模块、Hardswish激活函数和Ranger优化器3方面对ResNeXt50模型进行了改进,构建了SH-ResNeXt50锦鲤选美分级模型。试验结果表明:SH-ResNeXt50模型有效提升了锦鲤选美的等级分选能力,模型准确率达95.6%,损失值仅0.074,优于常用的AlexNet、GoogLeNet、ResNet50和ResNeXt50网络模型。最后,采用Grad-CAM分析SH-ResNeXt50模型的可解释性,结果表明SH-ResNeXt50模型和人工识别的感兴趣区域基本一致。本文所提出的方法实现了具有高相似度的锦鲤不同等级的智能分选,对其它具有高相似度的生物等级分选具有借鉴意义。 相似文献

为研究光照周期对北京油鸡雏鸡采食行为与生长性能的影响,本试验选择1日龄北京油鸡162只,随机分为三个处理(1、2、3),每处理54只,设3个重复,每个重复18只。1~3日龄均采用23L∶1D的光周期,4日龄起三个处理的光周期分别设置为8L∶16D、12L∶12D、16L∶8D,自雏鸡2周龄开始试验,6~7周龄观察记录各处理目标鸡的采食行为,并记录2~7周龄生长性能。结果表明:采食频次各处理间差异不显著(P>0.05),但处理2、3比处理1呈增加趋势;采食行为的最大、最小与平均持续时间处理3显著大于处理1(P<0.05),处理1和处理2之间无显著差异(P>0.05);采食在所有观测行为中所占时间比例均表现为处理3显著大于处理1(P<0.05),3个处理分别为18.41%、21.5%和24.74%。5周龄后采食量均为处理3显著高于处理1、2(P<0.05),7周龄日增重处理3显著高于处理1、2(P<0.05),而料肉比则在第6~7周龄时处理1显著低于处理3(P<0.05),其它各周各处理间无显著差异(P>0.05)。因此,北京油鸡雏鸡(7周龄前)在16L∶8D光周期下比12L∶12D、8L∶16D的采食时间长,利于雏鸡的采食量和体重的增加,但8L∶16D时的饲料利用效率比16L∶8D高。 相似文献
