The APSIM model was used to assess the impact of legumes on sorghum grown in rotation in a nutrient-limited system under dry conditions in south-western Zimbabwe. An experiment was conducted at Lucydale, Matopos Research Station, between 2002 and 2005. The model was used to simulate soil and plant responses in the experiment. Sequences of cowpea (Vigna unguiculata), pigeonpea (Cajanus cajan), groundnut (Arachis hypogaea) and sorghum (Sorghum bicolor) were used in the rotations. Legumes accumulated up to 130 kg of N ha−1 which was potentially available for uptake by sorghum in the following season. The APSIM model predicted total biomass, grain and N yields of the legume phase within the experimental error and performed well in predicting sorghum yield and N supplied in the rotation after cowpea and groundnut. The model generally under-predicted sorghum total biomass and grain yield after pigeonpea. Observed patterns of crop water use, evaporative losses during the dry season and re-charge of soil profile at the start of the rainy season were generally well predicted by the model. An assessment of output on sorghum N and water stresses in the rotation indicated that the legume–cereal rotation is more driven by soil nitrogen availability than water availability even under semi-arid conditions. Further legume–cereal rotation analysis using the model will assist in the understanding of other processes in the rotations in dry environments.  相似文献

Jatropha curcas seed-cake was evaluated for use as a solid state fermentation substrate for production of cellulolytic and xylanolytic enzymes by Aspergillus niger. Supplementation of the seedcake with 10% thatch grass (Hyperrhaenia sp.) resulted in a fivefold increase in xylanase production. Ammonium chloride supplementation increased production of xylanase by 13%. Under the same conditions, cellulase production was not influenced by supplementation with grass or the nitrogen sources used. Maximum xylanase was produced at 25 °C whilst cellulase was maximally produced at 40 °C. Highest xylanase activity was obtained when the cultures had an initial pH of 3 whereas cellulase was maximally produced at an initial pH of 5. Under optimised conditions, 6087 U and 3974 U of xylanase and cellulase respectively were obtained per gram of substrate. Zymograms of crude enzyme extracts showed six active bands ranging from 20 kDa to 43 kDa for cellulase and a 31 kDa active band for xylanase.  相似文献

The utilization of plant‐based diets in fish is limited by the presence of anti‐nutrients, which bind to dietary nutrients and reduce their bioavailability. Exogenous enzyme supplementation may alleviate the effects of anti‐nutrients and improve feed utilization. In this study, a commercial multi‐enzyme Natuzyme50® was added to a kikuyu‐based diet at a rate of 0 (control), 0.25, 0.5, 0.75 and 1.00 g kg?1 DM. Each diet was fed to triplicate groups of Oreochromis mossambicus for 60 days. All fish fed diets containing Natuzyme50® had higher (P < 0.05) growth performance compared with those fed the control diet. Fish fed the diet containing 0.50 g kg?1 had the best growth performance, highest protein digestibility and the highest levels of digestive enzyme activities. When Natuzyme50® was supplemented above 0.50 g kg?1 in the diet, growth performance and enzyme activities decreased. The improved growth performance observed with Natuzyme50® supplementation was attributed to the presence of enzymes such as cellulase, xylanase and phytase in the cocktail that are not naturally produced by fish. The presence of these enzymes released bound nutrients that would have not been available to the fish. Natuzyme50® supplementation also increased (P < 0.05) the activities of endogenous enzymes, improving the efficacy of the digestive process. The optimal dietary level of Natuzyme50® for optimal growth performance in O. mossambicus was 0.62 g kg?1 DM feed. Cost benefit analysis indicate that adding up to 0.5 g Natuzyme50® kg?1 in the diet results in higher profits as the increased growth attained offset the additional cost of Natuzyme50®.  相似文献

World-wide, enterotoxigenic Escherichia coli (ETEC) and verotoxigenic E. coli (VTEC)-induced diarrhea are economically important for porcine producers. Our aim was to investigate the prevalence of toxin and fimbrial genes among E. coli isolated from diarrheic piglets from randomly selected piggeries in Zimbabwe.We used multiplex PCR for screening STa, STb, LT, and Stx-2e toxins. Subsequently F4, F5, F6, F18 and F41 fimbriae genes were screened in toxin positive isolates. Toxin positive strains lacking tested fimbriae genes were characterized using transmission electron microscopy, agglutination and agglutination inhibition tests. Approximately 32% of the 1,984 isolates tested positive for STa, STb, LT or Stx-2e genes. Of these, approximately 81% had F4, F5, F6, F18 or F41 fimbriae genes. The remaining toxin positive strains lacked tested fimbriae genes and appeared to either express F1-like fimbriae, or lacked fimbriae. The data constitute an important framework for implementation of prevention measures, such as using relevant fimbriae-based vaccines against ETEC induced diarrhea or VTEC-induced edema.  相似文献
