Sagittaria trifolia L. is one of the most serious weeds in paddy fields in Japan. Since the late 1990s, severe infestations of S. trifolia have occurred following applications of sulfonylurea herbicides in Akita prefecture. In this study, two accessions of S. trifolia, R1 and R2, were collected from paddy fields with severe infestations and their resistance profiles were determined in comparison to a susceptible accession, S1. R1 and R2 were highly resistant to bensulfuron‐methyl. R1 was also highly resistant to pyrazosulfuron‐ethyl, but R2 was susceptible. Relative to S1, R1 had an amino acid substitution at the Pro197 residue of acetolactate synthase (ALS), a well‐known mutation that confers sulfonylurea resistance, suggesting that R1 has a target‐site‐based resistance (TSR) mechanism. The sequence of the ALS gene in R2 was identical to that in S1. A Southern blot analysis indicated that there was only one copy of the ALS gene in S1 and R2. These results suggest that R2 has a non‐target‐site‐based resistance (NTSR) mechanism. R2 was moderately resistant to imazosulfuron but susceptible to thifensulfuron‐methyl. R2 and S1 were susceptible to pretilachlor, benfuresate, MCPA‐ethyl and bentazon. The results reveal the occurrence of two sulfonylurea‐resistant biotypes of S. trifolia that show different mechanisms of cross‐resistance to sulfonylureas related to TSR in R1 and NTSR in R2.  相似文献

Vigorous cold tolerance at the fertilization stage (CTF) is a very important characteristic for stable rice production in cold temperature conditions. Because CTF is a quantitatively inherited trait, pyramiding quantitative trait loci (QTLs) using marker-assisted selection (MAS) is effective for improving CTF levels in rice breeding programs. We previously identified three QTLs controlling CTF, qCTF7, qCTF8 and qCTF12, using backcrossed inbred lines derived from a cross between rice cultivar Eikei88223 (vigorous CTF) and Suisei (very weak CTF). However, pyramiding of these QTLs for the application of MAS in practical rice breeding programs have not yet been elucidated. In this study, we examined the effect of pyramiding QTLs for improvement of CTF level using eight possible genotype classes from the 152 F3 population derived from a cross between Eikei88223 and Suisei. Increasing of CTF levels in combinations between qCTF7 and qCTF12 and between qCTF8 and qCTF12 were detected. Furthermore, we compared the haplotype pattern around the QTLs for CTF among the rice cultivars from Hokkaido. These results are useful for improvement of new cultivars with high CTF levels using MAS and identification of genetic resources with the novel QTL(s) for CTF.  相似文献

The valuable marine carotenoid, astaxanthin, is used in supplements, medicines and cosmetics. In this study, crustacyanin, an astaxanthin-binding protein, was used to solubilize and concentrate astaxanthin. The recombinant crustacyanin of European lobster spontaneously formed an inclusion body when it was over-expressed in Escherichia coli. In this study, fusing the NusA-tag to the crustacyanin subunits made it possible to express in a soluble fraction and solubilize astaxanthin in aqueous solution. By cutting off the NusA-tag, the crustacyanin subunits generated the pure insoluble form, and captured and concentrated astaxanthin. Overall, the attaching and releasing NusA-tag method has the potential to supply solubilized carotenoids in aqueous solution and concentrated carotenoids, respectively.  相似文献

To evaluate the available silicon (Si) content in agricultural soils in Japan and to investigate the determining factors of this content, we collected 180 soil samples from the surface layer of paddies and upland fields in Japan and determined their available Si contents. A phosphate buffer (PB; 0.02 M, pH 6.9) or an acetate buffer (AB; 0.1 M, pH 4.0) was used to extract available Si from the soil samples, and the Si concentrations in the extracts were determined by inductively coupled plasma-atomic emissions spectroscopy (ICP-AES). The total Si content and selected physicochemical properties were also determined for the soil samples. The median values of the available Si contents by the PB and AB methods were 48.8 and 79.7 mg kg?1 and corresponded to 0.017% and 0.027% of the total Si content, respectively. The overall data showed log-normal distributions. The available Si content of the upland soils was significantly higher than that of the paddy soils by both the PB (p < 0.01) and AB methods (p < 0.05). The available Si contents by the PB and AB methods had a significant positive correlation (p < 0.01) and they had significant negative correlation with the total Si content (p < 0.01). The values of the available Si contents by the PB and AB methods correlated positively with the pH, total carbon (C) content, and dithionite-citrate bicarbonate extractable iron (Fed) and aluminum (Ald), acid oxalate extractable iron (Feo) and aluminum (Alo), Fed-Feo and Alo+1/2Feo values (p < 0.01). A multi-regression analysis indicated that pH, amorphous minerals and crystalline iron (Fe) oxides were the dominant determining factors of available Si in the soils, and these three variables explained approximately two thirds of the variation of available Si content in agricultural soils in Japan. In terms of soil type, Terrestrial Regosols, Dark Red soils and Andosols had relatively high available Si contents, whereas Sand-dune Regosols, Red soils and Gray Lowland soils had relatively low contents. In terms of region, the soils in the Kanto and Okinawa regions had relatively high available Si contents and those in the Kinki, Shikoku and Chugoku regions had relatively low contents. In conclusion, the available Si content and its determining factors for agricultural soils in Japan were quantitatively elucidated, and this will contribute to the establishment of rational soil management?—including the application of silicate materials, taking into account the Si-supplying power of the relevant soils—for sustainable and productive agriculture in Japan.  相似文献

The possibility of restoring natural broadleaf forests may be decreased by the effects of plantation management, particularly in sites that undergo repeated rotation. We investigated the following two working hypotheses about the effects of repeated plantation of conifers on the natural regeneration of woody saplings in cool-temperate Japanese cedar plantations: (1) that repeated plantation of conifers decreases sapling species richness, and (2) that repeated plantation of conifers changes sapling species compositions. Our result supported the first hypothesis, because species richness was significantly lower in second-rotation plantations than in first-rotation plantations. The second hypothesis was not supported, because no significant or substantial differences in species composition were observed between plantations with different numbers of rotations. However, the abundance of tree (nonshrub) and gravity-dispersed species decreased after the second rotation of large saplings, albeit not those of small saplings, suggesting that response to repeated rotation depended on sapling size. Our results suggest that it is important to consider factors affecting the maintenance of a species in the plantations, such as distance from natural forests and seed sources, to minimize the effects of repeated plantation.  相似文献

Climate change has been an increasingly significant factor behind fluctuations in the yield and quality of rice (Oryza sativa L.), particularly regarding chalky (white-back, basal-white, and milky-white) grain, immature thin grain, and cracked grain. The development and use of heat-tolerant varieties is an effective way to reduce each type of grain damage based on the existence of each varietal difference. Cultivation methods that increase the available assimilate supply per grain, such as deep-flood irrigation, are effective for diminishing the occurrence of milky-white grains under high temperature and low solar radiation conditions. The application of sufficient nitrogen during the reproductive stage is important to reduce the occurrence of most heat damage with the exception of milky-white grain. In regard to developing measures for heat-induced poor palatability of cooked rice, a sensory parameter, the hardness/adhesion ratio may be useful as an indicator of palatability within a relatively wide air–temperature range during ripening. Methods for heat damage to rice can be classified as either avoidance or tolerance measures. The timing of the measures is further divided into preventive and prompt types. The use of heat-tolerant varieties and late transplanting are preventive measures, whereas the application of sufficient nitrogen as a top dressing and irrigation techniques during the reproductive stage are prompt types which may function to lower the canopy temperature by enhancing evapotranspiration. Trials combining the different types of techniques will contribute towards obtaining more efficient and steady countermeasures against heat damage under conditions of climate change.  相似文献
