A wilt disease of the model legume Lotus japonicus was observed in a greenhouse in Tokyo, Japan in May 2004. Roots of diseased plants were rotted and dark brown with lesions
spreading to lower stems and leaves, resulting in rapid plant death. The causal agent was identified as Fusarium solani based on the morphology. Sequence analysis of rDNA supported the identification. Inoculation of roots of healthy plants with
conidia reproduced characteristic disease symptoms, and F. solani was reisolated from lesions, satisfying Koch’s postulates. The isolate also caused chlorotic to necrotic lesions on leaves
of healthy plants after wound-inoculation. Infection by F. solani of leaves of L. japonicus was confirmed histologically. Mycelia were observed in the intercellular spaces of parenchymatous tissues in the lesion area
and the surrounding tissues. This is the first report of fungal disease on L. japonicus satisfying Koch’s postulates. We named it “Fusarium root rot of L. japonicus” as a new disease. The compatibility of L. japonicus and F. solani is expected to form a novel pathosystem for studying interactions between legumes and fungal pathogens.

‘Candidatus Liberibacter asiaticus’ is the most widespread of the three species of ‘Ca. Liberibacter’ that cause citrus greening disease (huanglongbing). To ascertain the phylogenetic relationships among Indian
isolates that have higher diversity in the 16S rDNA than Asian isolates of this species, we collected symptomatic leaves from
Northeast India, Papua New Guinea and Timor-Leste (East Timor) and detected ‘Ca. L. asiaticus’ by PCR using primers specific for nusG–rplK genes and 16S rDNA. Phylogenetic analysis with 16S rDNA sequences and single nucleotide polymorphisms of the omp gene region revealed that the Northeast Indian isolates were genetically closer to Asian-common isolates from Japan, Taiwan,
and Vietnam than to Indian isolates reported previously. Thus, the Asian-common strains of ‘Ca. L. asiaticus’ are apparently also present in Northeast India. 相似文献

It has been reported that the fruits and leaves of berries such as the blackberry, raspberry, and strawberry contain a high level of scavenging activity for chemically generated active oxygen species. This study investigated the antioxidative activities of black chokeberry fruit (Aronia melanocarpa Elliot) both in vitro and in vivo using the DPPH stable radical and rats with ethanol-induced gastric injury, respectively. The red pigment fraction of the black chokeberry contained three main components, one of which was identified as cyanidin 3-O-beta-glucoside by HPLC analysis and (1)H NMR. The black chokeberry red pigment fraction scavenged >44% of DPPH radicals at a concentration of 25 microg/mL compared to the control solution. The black chokeberry extract and its hydrolysate administrated at 2 g/kg of body weight each had nearly the same protective effect as quercetin administrated at 100 mg/kg of body weight in suppressing the area of gastric mucosal damage caused by the subsequent application of ethanol to <30% compared to the control group. The black chokeberry red pigment fraction had a similarly significant protective effect on gastric mucosa in a dose-dependent manner when administered at 30-300 mg/kg of body weight, and the administration of 30 mg/kg of body weight could suppress ethanol-induced gastric mucosal damage by approximately 50% (ID(50) = 30 mg/kg of body weight). 相似文献

Seven types of processed foods, namely, cornstarch, cornmeal, corn puffs, corn chips, tofu, soy milk, and boiled beans, were trial produced from 1 and 5% (w/w) genetically modified (GM) mixed raw materials. In this report, insect resistant maize (MON810) and herbicide tolerant soy (Roundup Ready soy, 40-3-2) were used as representatives of GM maize and soy, respectively. Deoxyribonucleic acid (DNA) was extracted from the raw materials and the trial-produced processed food using two types of methods, i.e., the silica membrane method and the anion exchange method. The GM% values of these samples were quantified, and the significant differences between the raw materials and the trial-produced processed foods were statistically confirmed. There were some significant differences in the comparisons of all processed foods. However, our quantitative methods could be applied as a screening assay to tofu and soy milk because the differences in GM% between the trial-produced processed foods and their raw materials were lower than 13 and 23%, respectively. In addition, when quantitating with two primer pairs (SSIIb 3, 114 bp; SSIIb 4, 83 bp for maize and Le1n02, 118 bp; Le1n03, 89 bp for soy), which were targeted within the same taxon specific DNA sequence with different amplicon sizes, the ratios of the copy numbers of the two primer pairs (SSIIb 3/4 and Le1n02/03) decreased with time in a heat-treated processing model using an autoclave. In this report, we suggest that the degradation level of DNA in processed foods could be estimated from these ratios, and the probability of GM quantification could be experimentally predicted from the results of the trial producing. 相似文献

It was recently reported that kokumi substances such as glutathione are perceived through the calcium-sensing receptor (CaSR). In addition, screening by the CaSR assay and sensory evaluation revealed that γ-glutamyl-valyl-glycine (γ-Glu-Val-Gly) was a potent kokumi peptide. In this study, the quantities of γ-Glu-Val-Gly in various commercial fish sauces originating from Vietnam (Nuoc Mum), Thailand (Nampra), China (Yu-lu), Korea, Japan (Shottsuru and Ikanago-shoyu), and Italy (Garum) were investigated using a LC/MS/MS method followed by derivatization with 6-aminoquinoyl-N-hydroxysuccinimidyl-carbamate (AQC). The analyses revealed γ-Glu-Val-Gly at concentrations ranging from 0.04 to 1.26 mg/dL, indicating that γ-Glu-Val-Gly is widely distributed among various commercial fish sauces. 相似文献

High-level accumulation of the target recombinant protein is a significant issue in heterologous protein expression using transgenic plants. Miraculin, a taste-modifying protein, was accumulated in transgenic tomatoes using an expression cassette in which the miraculin gene was expressed by the cauliflower mosaic virus (CaMV) 35S promoter and the heat shock protein (HSP) terminator (MIR-HSP). The HSP terminator was derived from heat shock protein 18.2 in Arabidopsis thaliana . Using this HSP-containing cassette, the miraculin concentration in T0 transgenic tomato lines was 1.4-13.9% of the total soluble protein (TSP), and that in the T1 transgenic tomato line homozygous for the miraculin gene reached 17.1% of the TSP. The accumulation level of the target protein was comparable to levels observed with chloroplast transformation. The high-level accumulation of miraculin in T0 transgenic tomato lines achieved by the HSP terminator was maintained in the successive T1 generation, demonstrating the genetic stability of this accumulation system. 相似文献
