Identifying the Fusarium species cause Fusarium head blight (FHB) and produces mycotoxins in wheat and other cereal is difficult and time consuming because of confusing
phenotypic classification systems. In Japan, the F. graminearum complex, F. culmorum, F. avenaceum, and Microdochium nivale predominantly cause FHB. The internal transcribed spacer (ITS) and 5.8S of rDNA, a partial sequence of β-tubulin and mitochondrial
cytochrome b (cytb) genes of the four species were PCR-amplified and analyzed. On the basis of the ITS, β-tubulin and cytb sequences, F. avenaceum and M. nivale are distinct from the F. graminearum complex and F. culmorum, whereas the F. graminearum complex is closely related to F. culmorum. Moreover, thiophanate–methyl-resistant isolates of the F. graminearum complex and F. culmorum did not have an amino acid substitution at amino acid codon 198 or 200 of β-tubulin. In contrast, very highly or highly thiophanate–methyl-resistant
isolates of M. nivale had Glu (GAG) substituted with Ala (GCG) or Lys (AAG) at codon 198, respectively. The allele-specific PCR assay was used

Fungicide resistance in plant pathogens is often caused by a single point mutation in a gene encoding fungicide target proteins.
Such is the case for resistance to MBI-D (inhibitors of scytalone dehydratase in melanin biosynthesis) fungicides in rice
blast fungus (Magnaporthe oryzae), which is caused by a mutation in the scytalone dehydratase gene that results in a replacement of valine with methionine
at codon 75 of the fungicide target protein. PCR-Luminex, a novel system developed for high-throughput analysis of single
nucleotide polymorphisms (SNPs) was successfully introduced to diagnose MBI-D resistance using specific oligonucleotide probes
coupled with fluorescent beads. The PCR-Luminex system was further tested for its potential in identifying species causing

A class III chitinase gene (CHI2) is induced in cucumber plants (Cucumis sativa L.) in response to infection by pathogenic microorganisms. Infection of Botrytis cinerea, causal agent of gray mold disease on cucumber, also induces CHI2 expression. To investigate whether CHI2 is involved in resistance to gray mold disease, transgenic cucumber plants were produced to overexpress the CHI2 gene. One line was analyzed in detail in terms of disease resistance. The transgenic cucumber plant (CC2) constitutively expressed CHI2 and reduced the symptoms of B. cinerea for 4 days after inoculation compared with nontransgenic plants. However, this inhibitory effect was not absolute, and CC2 eventually developed serious disease symptoms. Chitinase activity of the crude extract from CC2 leaves was higher than that from nontransgenic plants. A high-molecular-weight fraction containing CHI2 from CC2 leaves had fungistatic activity against B. cinerea. Interestingly, the low-molecular-weight fraction from CC2 leaves with CHI2 removed also had fungistatic activity against B. cinerea. Not only the introduced chitinase activity but also the endogenous defense reactions activated by overexpression of CHI2 may be involved in the enhanced gray mold disease resistance in CC2. 相似文献

SummaryFruit firmness and cell wall composition were compared among five cultivars of F. × ananassa (‘Chandler’, ‘Seascape’, ‘Tudla’, ‘Hokowase’ and ‘Reiko’) and two land races of F. chiloensis (‘94BAU-1A’ and ‘97PUR-1A’) grown in central Chile. Fruits of F. chiloensis were smaller than those of F. × ananassa. Firmness of epidermal tissue did not differ significantly between F. × ananassa and F. chiloensis. Firmness of cortical tissue, however, was significantly lower in F. chiloensis than in F.× ananassa and as a result, difference in the firmness between epidermal and cortical tissues was significantly greater in F. chiloensis than in F. × ananassa. Calcium concentration was low in ‘Reiko’ and ‘Hokowase’ while high in the other varieties of F. × ananassa. F. chiloensis showed intermediate values between these two groups. Higher levels of total uronic acid and neutral sugars in cell wall fractions of F. chiloensis as compared with F. × ananassa were mainly due to its high concentration in the water soluble fraction. Total uronic acid in cell wall fraction was significantly higher in F. chiloensis than in F. × ananassa, mainly due to significantly higher concentrations in the water-soluble fraction. Non-cellulosic neutral sugars, especially arabinose in the water, CDTA, and Na2CO3-soluble fractions were significantly higher in F. chiloensis than in F. × ananassa. On the other hand, galactose in the CDTA-, Na2CO3-, and KOH-soluble fractions was significantly lower in F. chiloensis than in F. × ananassa. Moreover, cellulose concentration of F. chiloensis was also significantly lower than that of F. × ananassa. Results obtained from cell wall analysis suggest that softer cortex in F. chiloensis might be due to either lower cellulose content, higher solubilization rate of pectic substances, and/or compositional differences in the branching of neutral sugars. 相似文献

The protective effect of immunoglobulins derived from chicken egg yolk (IgY) against infection by Canine parvovirus 2 (CPV-2) was evaluated in 10 beagle dogs orally challenged with a strain of the virus. The 2-mo-old dogs were divided into 3 groups and treated with powders containing CPV-2 IgY or normal egg yolk for 7 d after the challenge. The 4 dogs receiving normal egg yolk (control group) demonstrated mild symptoms typical of CPV-2 infection, such as vomiting, diarrhea, and weight loss. No symptoms were observed by 16 d after challenge in the 3 dogs receiving 2 g of IgY powder. Of the 3 dogs receiving 0.5 g of IgY powder, 2 had clinical CPV-2 disease; however, the manifestations were less severe than in the control group. Furthermore, the IgY-treated groups had significantly greater weight gain and shorter duration of virus shedding than the control group. These results indicate that IgY is useful in protecting dogs from CPV-2-induced clinical disease. 相似文献

Heated and hydrated naturally occurring dolomite showed very strong antiviral activity. Infectivity of avian and human influenza, avian infectious bronchitis (coronavirus), Newcastle disease (paramyxovirus) and avian laryngotracheitis (herpesvirus) viruses dropped at least 1,000 fold following contact with the dolomite for five minutes at 4 degrees C. Dolomite is expected to be useful to inhibit the incidence of emerging and re-emerging infectious diseases. 相似文献
