Background
Measurement of salivary cortisol has been used extensively as a non-invasive alternative to blood sampling to assess adrenal activity in ruminants. However, there is evidence suggesting a considerable delay in the transfer of cortisol from plasma into saliva. Previous studies in cattle have used long sampling intervals making it difficult to characterise the relationship between plasma and salivary cortisol (PLCort and SACort, respectively) concentrations at different time points and determine whether or not such a time lag exist in large ruminants. Therefore, the objective of this study was to characterise the relationship between plasma and salivary cortisol and determine if there is a significant time lag between reaching peak cortisol concentrations in plasma and saliva across a 4.25 h time-period, using short sampling intervals of 10–15 min, following social separation in dairy cattle.Five cows were separated from their calves at 4 days after calving, and six calves were separated from a group of four peers at 8 weeks of age. Following separation, the animals were moved to an unfamiliar surrounding where they could not see their calves or pen mates. The animals were catheterised with indwelling jugular catheters 1 day before sampling. Blood and saliva samples were obtained simultaneously before and after separation.Results
In response to the stressors, PLCort and SACort increased reaching peak concentrations 10 and 20 min after separation, respectively. This suggested a 10 min time lag between peak cortisol concentrations in plasma and saliva, which was further confirmed with a time-series analysis. Considering the 10 min time lag, SACort was strongly correlated with PLCort (P < 0.0001).Conclusions
Salivary cortisol correlates well with plasma cortisol and is a good indicator of the time-dependent variations in cortisol concentrations in plasma following acute stress. However, there is a time lag to reach peak cortisol concentrations in saliva compared to those in plasma, which should be considered when saliva samples are used as the only measure of hypothalamic-pituitary-adrenal axis response to stress in cattle. 相似文献12.

This study was conducted to investigate the effects of increasing levels of extracts of Byrsonima crassifolia, Celtis pallida, Enterolobium cyclocarpum, Fraxinus excelsior, Ficus trigonata, Phoradendrom brevifolium and Prunus domestica on in vitro gas production (GP) and ruminal fermentation of a high concentrate diet. Plant extracts were prepared at 1 g dry matter (DM)/8 mL of solvent mixture (methanol : ethanol : water, 1:1:8) and added at levels of 0, 0.6, 1.2 and 1.8 mL/g DM of a high concentrate diet. In vitro GP was recorded at 2, 4, 6, 8, 10, 12, 24, 48 and 72 h of incubation. Increasing addition of extracts linearly increased (P < 0.001), the GP24, GP48 and GP72 (mL/g DM), and linearly decreased (P < 0.001), the discrete GP lag time. Moreover, increasing extract doses linearly increased (P < 0.001) the asymptotic GP and decreased (P < 0.001) the rate of GP. GP6 was not impacted by treatments and GP12 increased linearly (P = 0.01) with increasing addition of extracts. Rumen pH declined linearly (P < 0.05) with increasing doses of extracts added. As no interactions (P > 0.05) occurred between the extracts and doses, it could be conclude that all extracts positively modified rumen fermentation at doses of 1.2 to 1.8 mL extract/g diet DM. 相似文献

Background
Heifers not used as breeding stock are often implanted with steroids to increase growth efficiency thereby altering hormone profiles and potentially changing the environment in which ovarian follicles develop. Because bovine granulosa cell culture is a commonly used technique and often bovine ovaries are collected from abattoirs with no record of implant status, the objective of this study was to determine if the presence of an implant during bovine granulosa cell development impacts follicle stimulating hormone-regulated steroidogenic enzyme expression. Paired ovaries were collected from 16 feedlot heifers subjected to 1 of 3 treatments: non-implanted (n = 5), Revalor 200 for 28 d (n = 5), or Revalor 200 for 84 d (n = 6). Small follicle (1 to 5 mm) granulosa cells were isolated from each pair and incubated with phosphate buffered saline (n = 16) or 100 ng/mL follicle stimulating hormone (n = 16) for 24 h.Results
Granulosa cells of implanted heifers treated with follicle stimulating hormone produced medium concentrations of progesterone similar (P = 0.22) to non-implanted heifers, while medium estradiol concentrations were increased (P < 0.10) at 28 and 84 d compared to non-implanted heifers indicating efficacy of treatment. Additionally, real-time PCR analysis in response to follicle stimulating hormone treatment demonstrated a decrease in steroidogenic acute regulatory protein (P = 0.05) mRNA expression in heifers implanted for 84 d and an increase in P450 side chain cleavage mRNA in granulosa cells of heifers implanted for 28 (P < 0.10) or 84 d (P < 0.05) compared to non-implanted females. However, no difference in expression of 3-beta-hydroxysteroid dehydrogenase (P = 0.57) and aromatase (P = 0.23) were demonstrated in implanted or non-implanted heifers.Conclusions
These results indicate follicles which develop in the presence of high concentrations of androgenic and estrogenic steroids via an implant tend to demonstrate an altered capacity to respond to follicle stimulating hormone stimulation. The dietary protein requirement of the freshwater prawn Macrobrachium rosenbergii to obtain the maximum body protein retention was assessed by the estimated protein needed daily for maintenance (M) and body protein increment (G). The body protein increment was estimated by quantifying body nitrogen increments when the prawn was fed high‐protein diets composed primarily of casein. The protein required for maintenance was estimated by regressing weight gains in feeding experiments using graded dietary levels of casein. The true daily retention (R) or increase in body protein of the prawn corresponds to the sum of M and G. The values of M, G, and R determined in the present study were 3.86, 0.24, and 4.10 g/kg body weight/d, respectively. On the basis of the daily body protein increment and the net protein utilization of casein (58%), dietary protein requirement of the juvenile M. rosenbergii for maximum body protein retention was suggested to be about 7.1 g/kg body weight/d. 相似文献

We describe the seasonal migrations of Argentine chub mackerel ( Scomber japonicus ) into the fishing ground in relation to sea surface temperature conditions and analyse the temperature evolution during the fishing season on the basis of 1955–97 time-series. The upper temperature limit for the presence of schools was around 19°C. At higher values, most schools leave coastal waters (< 50 m) for the shelf. An inverse relationship was observed between catches per unit of effort (CPUE) of the inshore fishery and sea surface temperature. The highest standardized CPUE values corresponded to temperatures lower than 19°C. Above this temperature, the probability of obtaining a standardized CPUE higher than 1 was extremely low, and the standard deviations were minimal. The economic benefit from catches was also analysed through the different months of the fishing season. It was found that the mean benefit corresponding to temperatures lower than 19°C exceeded approximately 15 times that obtained at higher temperatures. A simple fishing strategy was proposed to the commercial fleet in order to improve the economic benefit of the catches. The migratory pattern followed by mackerel during the spawning period was also analysed in relationship to other environmental factors like food availability. Zooplankton composition, size structure and abundance in coastal and shelf waters are described, and their implications for feeding ecology of larvae and adults of the species are discussed. 相似文献

OBJECTIVE: To assess the efficacy of prophylactic hoof health examination and trimming during midlactation at reducing the incidence of lameness during late lactation in dairy cows. DESIGN: Randomized field trial. ANIMALS: 333 Holstein cows. PROCEDURES: Cows without apparent lameness were randomly allocated into 1 of 2 groups approximately 204 days after calving. Cows allocated to the treatment group (n = 161) were examined on a tilt table for diagnosis and underwent hoof-trimming procedures, if needed, for treatment of hoof disorders or lesions. Cows in the control group (n = 172) were not examined. Cows were assigned a locomotion score weekly for 28 weeks after allocation to a group. The number of cows classified as lame during late lactation (approx 205 to 400 days after calving) was compared between groups to assess the efficacy of prophylactic examination and trimming. RESULTS: Incidence of lameness during late lactation was 24% in cows in the control group and 18% in cows in the treatment group. CONCLUSIONS AND CLINICAL RELEVANCE: The 25% decrease in number of new cases of lameness in cows undergoing prophylactic hoof health examination and trimming during midlactation may be relevant for the well-being of dairy cows and should not represent a major economic burden to producers. 相似文献

The single seed descent (SSD) method of selection has been evaluated in safflower (Carthamus tinctorius L.). The initial population of this study was a BC2F1 from a cross between a well adapted and a high oil cultivar. Transgressive segregation for yield and oil content producing lines with considerably higher values than the better parent was observed in BC2F4 and BC2F5 generations. These results and the great saving of time and effort makes the SSD method a very desirable tool for safflower breeders. 相似文献

Genetic diversity within and between the maintainer (B) and restorer (R) lines used in hybrid breeding programs of the Philippine
was investigated with information from analysis of pedigree record, quantitative traits and SSR assays. Mean coefficients
of coancestry were calculated as 0.11 within R lines, 0.27 within B lines, and 0.04 between R and B lines, indicating greater
diversity among R lines than among B lines, and the significant divergence between B and R lines. These results are consistent
with those obtained from quantitative trait analysis and SSR marker assays. Relative gene diversity for 37 random SSR markers
averaged 0.20 within B lines,0.28 within R lines, and 0.52 between two groups of lines. There were no consistent associations
among various genetic diversity measures. Random sets of SSR marker and pedigree based diversity measures had no significant
correlation with mid-parent heterosis for grain yield and biomass, indicating that prediction of heterosis for complex traits
based on these two genetic diversity estimates is difficult.

Lipid hydroperoxides are important factors in lipid oxidation due to their ability to decompose into free radicals. In oil-in-water emulsions, the physical location of lipid hydroperoxides could impact their ability to interact with prooxidants such as iron. Interfacial tension measurements show that linoleic acid, methyl linoleate, and trilinolein hydroperoxides are more surface-active than their non-peroxidized counterparts. In oil-in-water emulsion containing surfactant (Brij 76) micelles in the continuous phase, linoleic acid, methyl linoleate, and trilinolein hydroperoxides were solubilized out of the lipid droplets into the aqueous phase. Brij 76 solubilization of the different hydroperoxides was in the order of linoleic acid > trilinolein > or = methyl linoleate. Brij 76 micelles inhibited lipid oxidation of corn oil-in-water emulsions with greater inhibition of oxidation occurring in emulsions containing linoleic acid hydroperoxides. Surfactant solubilization of lipid hydroperoxides could be responsible for the ability of surfactant micelles to inhibit lipid oxidation in oil-in-water emulsions. 相似文献
