Aminoacyl-transfer RNA (tRNA) synthetases, which catalyze the attachment of the correct amino acid to its corresponding tRNA during translation of the genetic code, are proven antimicrobial drug targets. We show that the broad-spectrum antifungal 5-fluoro-1,3-dihydro-1-hydroxy-2,1-benzoxaborole (AN2690), in development for the treatment of onychomycosis, inhibits yeast cytoplasmic leucyl-tRNA synthetase by formation of a stable tRNA(Leu)-AN2690 adduct in the editing site of the enzyme. Adduct formation is mediated through the boron atom of AN2690 and the 2'- and 3'-oxygen atoms of tRNA's3'-terminal adenosine. The trapping of enzyme-bound tRNA(Leu) in the editing site prevents catalytic turnover, thus inhibiting synthesis of leucyl-tRNA(Leu) and consequentially blocking protein synthesis. This result establishes the editing site as a bona fide target for aminoacyl-tRNA synthetase inhibitors.  相似文献

Physalaemin: an amphibian tachykinin in human lung small-cell carcinoma   总被引:3,自引:0,他引:3  
Immunoreactivity to the amphibian peptide physalaemin was characterized from extracts of a human lung small-cell carcinoma by immunological, chemical, and pharmacological means. Tumor-related peptide cross-reacted with three antiserums to physalaemin to yield 1.1 to 1.6 nanomoles per gram of tissue. Physalaemin and tumor peptide had similar retention times on high-performance liquid chromatography after chemical and enzymic modifications that included pH changes, oxone oxidation, use of a hydrophilic ion-pairing reagent, and digestion with trypsin and pyroglutamate aminopeptidase. Both physalaemin and the tumor peptide produced a contractile response of isolated guinea pig ileum at threshold concentrations of approximately 100 to 150 picograms per milliliter. These data suggest that small-cell carcinoma of the lung contains a physalaemin-like peptide that has structural and biological homology to its amphibian counterpart.  相似文献

The use of composts in agricultural soils is a widespread practice and the positive effects on soil and plants are known from numerous studies. However, there have been few attempts to compare the effects of different kinds of composts in one single study. The aim of this paper is to investigate to what extent and to which soil depth four major types of composts would affect the soil and its microbiota.In a crop-rotation field experiment, composts produced from (i) urban organic wastes, (ii) green wastes, (iii) manure and (iv) sewage sludge were applied at a rate equivalent to 175 kg N ha−1 yr−1 for 12 years. General (total organic C (Corg), total N (Nt), microbial biomass C (Cmic), and basal respiration), specific (enzyme activities related to C, N and P cycles), biochemical properties and bacterial genetic diversity (based on DGGE analysis of 16S rDNA) were analyzed at different depths (0-10, 10-20 and 20-30 cm).Compost treatment increased Corg at all depths from 11 g kg−1 for control soil to 16.7 g kg−1 for the case of sewage sludge compost. Total N increased with compost treatment at 0-10 cm and 10-20 cm depths, but not at 20-30 cm. Basal respiration and Cmic declined with depth, and the composts resulted in an increase of Cmic and basal respiration. Enzyme activities were different depend on the enzyme and among compost treatments, but in general, the enzyme activities were higher in the upper layers (0-10 and 10-20 cm) than in the 20-30 cm layer. Diversity of ammonia oxidizers and bacteria was lower in the control than in the compost soils. The type of compost had less influence on the composition of the microbial communities than did soil depth.Some of the properties were sensitive enough to distinguish between different compost, while others were not. This stresses the need of multi-parameter approaches when investigating treatment effects on the soil microbial community. In general, with respect to measures of activity, biomass and community diversity, differences down the soil profile were more pronounced than those due to the compost treatments.  相似文献

While new lines developed by CIMMYT under intermediate levels ofnitrogen are more nitrogen efficient than older lines, it is not knownwhether this method of selection is the most efficient. Two lines with highN up-take efficiency (UPE) were crossed to two lines with high N utilizationefficiency (UTE). Their progenies were visually selected for superioragronomic type from the F2 till the F6 under each of fiveselection regimes. These were: always under low N (0 added) (LN), alwaysunder medium N (150 kg N ha-1 added) (MN), always under highN (300 kg N ha-1 added) (HN), alternating between low and highN starting with low N in the F2 (ALN) and alternating between highand low N starting with high N in the F2 (AHN). The resultingadvanced lines (F8) were tested in replicated yield trials under low,medium, and high levels of added N on an N deficient soil. The AHNselection regime resulted in the highest yields at intermediate and high Nlevels. The lowest yields occurred for ALN. Harvest Index (HI) did notchange. Hence all effects on yield were the result of changes in biomass.Under low N all selection regimes showed progress in yield due to animproved biomass and HI, but with no differences among regimes. UPEexpressed a higher correlation with yield and biomass than UTE under alllevels of N.  相似文献

An 8‐year‐old Labrador Retriever was presented for inspiratory efforts with stertor. A rhinoscopy and a magnetic resonance imaging scan were performed and revealed a naso‐pharyngeal mass. The mass was identified in the nasopharynx without meningeal extension. This mass was both T1‐ and T2‐hyperintense, compared to normal brain parenchyma without significant postcontrast enhancement. The mass was surgically removed and the histologic diagnosis was a mesenchymal tumor. Immunohistochemistry with antibodies was conducted and consistent with an extracranial meningioma. To the authors’ knowledge, this is the first case report documenting a primary extracranial nasopharyngeal meningioma in a dog.  相似文献

The effectiveness of adding two organic wastes (cotton gin crushed compost, CGCC, and poultry manure, PM) to a saline soil (Salorthidic Fluvaquent) in dryland conditions near Seville (Guadalquivir Valley, Andalusia, Spain) was studied during a period of 5 years. Organic wastes were applied at rates of 5 and 10 t organic matter ha−1. One year after the assay began, spontaneous vegetation had appeared in the treated plots, particularly in that receiving a high PM dose. After 5 years the plant cover in this treated plot was around 80% (compared with the 8% of the control soil). The effect on the soils physical and chemical properties, soil microbial biomass, and six soil enzymatic activities (dehydrogenase, urease, protease, β-glucosidase, arylsulfatase, and phosphatase activities) were ascertained. Both added organic wastes had a positive effect on the physical, chemical and biological properties of the soil, although at the end of the experimental period, the soil physical properties, such as bulk density, increased more significantly in the CGCC-amended soils (23%) and the exchangeable sodium percentage (ESP) decreased more significantly in the CGCC-amended soils (50%) compared to the unamended soil. Water soluble carbohydrates and soil biochemical properties were higher in the PM-amended soils compared to the CGCC-amended soils (by 70% for water soluble carbohydrates, and by 34, 18, 37, 39, 40 and 30% for urease, protease, β-glucosidase, phosphatase, arylsulfatase and dehydrogenase activities, respectively). After 5 years, the percentage of plant cover was >50% in all treated plots and 8% in the control soil.  相似文献

The major Ricinus communis allergens are the 2S albumins, Ric c 1 and Ric c 3. These proteins contain a trypsin/α-amylase inhibitor family domain, suggesting that they have a role in insect resistance. In this study, we verified that Ric c 1 and Ric c 3 inhibited the α-amylase activity of Callosobruchus maculatus, Zabrotes subfasciatus, and Tenebrio molitor (TMA) larvae as well as mammalian α-amylase. The toxicity of 2S albumin was determined through its incorporation in C. maculatus larvae as part of an artificial diet. Bioassays revealed that 2S albumin reduced larval growth by 20%. We also analyzed the tridimensional structures of Ric c 1 and Ric c 3 by (a) constructing a comparative model of Ric c 1 based on Ric c 3 NMR structure and (b) constructing the theoretical structure of the Ric c 1-TMA and Ric c 3-TMA complexes. Our biological and theoretical results revealed that Ric c 1 and Ric c 3 are a new class of α-amylase inhibitors. They could potentially be used to help design inhibitors that would be useful in diverse fields, ranging from diabetes treatment to crop protection.  相似文献

Despite the critical role of biological soil crusts (BSCs) in arid and semi-arid ecosystem function, few studies are found concerning the most important environmental variables affecting their distribution and physiology. This study seeks to determine soil and microenvironmental factors affecting the spatial distribution and pigment production of BSC-forming lichens and mosses in open patches of a semi-arid Mediterranean kermes oak thicket. We measured late-successional BSC cover, shrub cover, distance to nearest kermes oak (to test for effects of kermes oak thicket microenvironment on BSC), and pigment concentration of one lichen (Cladonia foliacea) and one moss (Pleurochaete squarrosa) species in the Nature Reserve El Regajal-Mar de Ontígola (Central Spain). At the macroscale (>0.5 m), results showed that BSC distribution and pigments were tightly coupled to a suite of soil properties, in particular soil pH, Fe, and Ca. Specifically, soil pH had a positive relationship with the cover of five individual BSC-forming lichen species and was negatively related to pigment production in C. foliacea. When pH was excluded from the analysis, Ca appeared as the main soil variable and was correlated with total BSC cover and total lichen cover. The micronutrient Fe had a significant positive relationship with the concentration of eight pigments in P. squarrosa and was also coupled with the cover of two BSC-forming lichens. Manganese, previously proposed as a key limiting micronutrient for BSCs, affected lichen diversity in a negative way. At the microscale (∼0.5 m), kermes oak microenvironment, shrub cover, and moss cover were determinants of BSC distribution, and total lichen and total BSC cover were overrepresented on N and E-facing shrub microsites. Our findings suggest that soil chemical variability and microsite diversity created by neighbouring vegetation affect BSC distribution in complex and essential ways and that studies aiming to explore BSC-environment relationships should be conducted at various spatial scales. Studies based on species- or group-specific responses are, thus, inadequate to unveil the main factors determining the distribution of the diverse organisms that constitute BSCs and/or to propose potential tools aiming to restore BSC in arid and semiarid ecosystems.  相似文献

A 55-d feeding experiment was conducted to evaluate the effects of dietary vitamin A on growth of juvenile red sea bream Chrysophrys major . Nine semi-purified diets with supplemental retinol palmitate at levels of 0, 300, 600, 1,500, 3,000, 4,500, 6,000, 15,000 and 30,000 retinol eq/kg diet, were fed twice daily to triplicate groups of 10 juveniles per tank with an initial weight of 1.178 ± 0.001 g. Weight gain and specific growth rate increased with the increase of supplemental retinol palmitate in the diet up to a level of 6,000 retinol eq/kg diet, beyond which those indices tended to decrease slightly or to plateau. Significantly lower weight gain and specific growth rate were observed in fish fed with 0 and 300 retinol eq/kg diet. No significant differences were found in survival rates among the different levels of supplementation. The crude protein, lipid, moisture and ash contents in the whole body were not affected by the different vitamin A supplemental levels. Besides the reduced growth, fish fed with low levels of vitamin A supplement showed no other signs of deficiency. No hypervitaminosis A symptoms were observed in fish fed on high levels of supplementation. Fish fed on diets with 0 and 300 retinol eq/kg diet showed a significantly lower content of vitamin A (as a total retinol) in the liver than those in groups fed with higher levels of supplementation. Vitamin A was stored in the liver mainly as retinyl esters. Total retinal content of the eye showed no significant differences among the treatments; however, fish fed with 0 and 300 retinol eq/kg of diet had slightly higher concentrations. According to the results of the mean weight gain, specific growth rate and liver retinol content, the requirement of dietary vitamin A for juvenile red sea bream was estimated to be between 1,500 and 6,000 retinol eq/kg.  相似文献
