Cystic ovarian disease (COD) is one of the main causes of infertility in dairy cattle. It has been shown that intra‐ovarian factors may contribute to follicular persistence. Transforming growth factor‐beta (TGFB) isoforms are important paracrine and autocrine signalling molecules that regulate ovarian follicle growth and physiology. Considering the importance of these factors in the ovarian physiology, in this study, we examined the expression of TGFB isoforms (TGFB1, TGFB2 and TGFB3) in the ovary of healthy cows and animals with spontaneous and adrenocorticotrophic hormone (ACTH)‐induced COD. In the oestrous‐synchronized control group, the expression of TGFB1 in granulosa and theca cells was higher in spontaneous cysts than in atretic or tertiary follicles. When we compared TGFB2 expression in granulosa cells from atretic or tertiary follicles from the oestrous‐synchronized control group with that in ACTH‐induced or spontaneous follicular cysts, we found a higher expression in the latter. The expression of the TGFB isoforms studied was also altered during folliculogenesis in both the spontaneous and ACTH‐induced COD groups. As it has been previously shown that TGFB influences steroidogenesis, ovarian follicular proliferation and apoptosis, an alteration in its expression may contribute to the pathogenesis of this disease.  相似文献

While, for a long time, the role of mitochondria in sperm physiology and pathology has been largely ignored, recent research points out the mitochondria as a major organelle with key roles in sperm function both under physiological and biotechnological conditions. This paper briefly reviews these novel findings regarding the role of mitochondria in sperm, paying special attention to the most practical, readily applicable, aspects of the topic such as their role as a major source of the sublethal damage that sperm experiments after cryopreservation.  相似文献

According to our previous investigations, resistance to Phytophthora capsid in Capsicum annuum genotypes, ‘Line 29’, ‘PI201232’, ‘PI201234’ and Serrano Criollo de Morelos 334 (‘SCM334’), seems to be controlled by three genes. In order to determine the genie relationships between these four sources of resistance, three experiments were conducted which included the four genotypes, their F1s, F2s, F3s and BC1 generations together with the susceptible pepper genotype ‘Morron INI A 224’. Inoculations were made, when plants had 4—6 leaves, by irrigating the culture substrate with a zoospore suspension of P. capsici isolate ‘Bl’. Though the four genotypes showed percentages of resistance close to a 100%, none of them actually reached this level in the three experiments. ‘SCM334’ was the most resistant genotype, transmitting a high level of resistance to its F1, F2 and BQ generations. ‘Line 29’ was more resistant than ‘PI201232’ and ‘PI201234’. However, the F1 F2 and BQ generations of these three lines showed similar degrees of resistance. The four genotypes seem to have one of the three genes postulated for their resistance in common. All genes displayed a similar level of resistance, except the specific genes of ‘SCM334’, the effect of which was slightly higher. Several working procedures are suggested for breeding programmes.  相似文献

MOPECO: an economic optimization model for irrigation water management   总被引:2,自引:0,他引:2  
Water is a natural, sometimes scarce, and fundamental resource for life, essential both for agriculture in many regions of the world and also to achieve sustainability in production systems. Maximizing net returns with the available resources is of the utmost importance, but doing so is a complex problem, owing to the many factors that affect this process (e.g. climatic variability, irrigation system configuration, production costs, subsidy policies). The MOPECO model is a tool for identifying optimal production plans, and water irrigation management strategies. The model estimates crop yield, production and gross margin as a function of the irrigation depth. Finally, these gross margin functions are used to determine an optimum cropping pattern and irrigation strategy to maximize the gross margin on a farm in a specific scenario. Since the relationships between the variables are generally non-linear and the number of alternative strategies is quite large, the optimum process is complex and computationally intensive. Genetic algorithms are therefore used to identify optimal strategies. This paper describes the MOPECO model, which comprises three computing modules: (1) estimation of net water requirements; (2) derivation of the relationship between gross margin and irrigation depth; and (3) identification of the crop planning and the water volumes to be applied. The results obtained by applying the MOPECO model to a specific irrigable area in a semi-arid area of Spain, with great deficits and high water costs, are also included and discussed. These results usually show that the irrigation depth for maximum benefits is lower than that necessary to obtain maximum production. In some areas of Spain, horticultural crops are nearly always part of the optimum alternative. The crops that become part of the optimum alternative are mainly horticultural crops with a high gross margin and low water requirements. The irrigation depths selected for the ideal crop rotation are included among the irrigation depth of maximum economic efficiency and the maximum gross margin irrigation depth. Both are lower than that necessary for the maximum yield. This model helps farmers, extension services, and other agents to analyse, make decisions and optimize water management.Communicated by A. ABSTRACT: In sheep polymorphisms of the prion gene (PRNP) at the codons 136, 154 and 171 strongly influence the susceptibility to scrapie and bovine spongiform encephalopathy (BSE) infections. In goats a number of other gene polymorphisms were found which are suspected to trigger similar effects. However, no strong correlation between polymorphisms and TSE susceptibility in goats has yet been obtained from epidemiological studies and only a low number of experimental challenge data are available at present. We have therefore studied the potential impact of these polymorphisms in vitro by cell-free conversion assays using mouse scrapie strain Me7. Mouse scrapie brain derived PrPSc served as seeds and eleven recombinant single mutation variants of sheep and goat PrPC as conversion targets. With this approach it was possible to assign reduced conversion efficiencies to specific polymorphisms, which are associated to low frequency in scrapie-affected goats or found only in healthy animals. Moreover, we could demonstrate a dominant-negative inhibition of prion polymorphisms associated with high susceptibility by alleles linked to low susceptibility in vitro.  相似文献

This study assessed the intraoperative analgesic effects of intravenous lidocaine administered by a constant rate infusion (CRI) in surgical canine patients. A prospective, blinded, randomized study was designed with 2 treatment groups: A (lidocaine) and B (placebo), involving 41 dogs. All patients were premedicated with acepromazine and buprenorphine, induced with propofol and midazolam; anesthesia was maintained with isoflurane in oxygen. Group A received 2 mg/kg IV lidocaine immediately after induction, followed within 5 min by a CRI at 50 μg/kg/min. Group B received an equivalent volume of saline instead of lidocaine. Changes in heart rate and blood pressure during maintenance were treated by increasing CRI. Fentanyl was used as a supplemental analgesic when intraoperative nociceptive response was not controlled with the maximum dose of lidocaine infusion. There was a significantly lower use of supplemental intraoperative analgesia in the lidocaine than in the placebo group. Group B dogs had almost twice as high a risk of intraoperative nociceptive response as group A dogs.  相似文献

Broadleaf herbicides are commonly used in rangelands to suppress exotic weeds and release native communities from negative impacts of invasion. However, few studies have comprehensively evaluated treatment effects on differing community components across a gradient of initial invasion levels. We conducted a 6-yr experiment within grasslands of western Montana to measure local-scale effects of a broadcast application of picloram on 1) cover of the target invader, spotted knapweed (Centaurea stoebe L.), 2) prevalence of native functional groups, and 3) the secondary invader cheatgrass (Bromus tectorum L.) at differing initial levels of knapweed invasion. Treatment effectively suppressed knapweed, with cover in treated vs. control plots reduced by >60% in the sixth posttreatment year. Treatment also appeared to alleviate knapweed’s impacts on native perennial grasses, but only at the highest initial level of invasion, where cover of this group increased by >30% in treated vs. control plots to equal levels associated with noninvaded plots. In some cases, treatment appeared to exacerbate knapweed’s impacts on native forbs. At the no-invasion level, perennial forb cover declined by >20% in treated vs. control plots to match values associated with moderate or high levels of invasion, but these treatment effects were minimal at the latter invasion levels. Across initial invasion levels, species richness of perennial and/or annual forbs declined by >20% in treated vs. control plots. Treatment also promoted increases in cheatgrass cover, although differences between treated and control plots were relatively small by the sixth posttreatment year. Overall, effects of picloram application depended on initial levels of knapweed invasion, largely due to the varying strength of release effects. Selective treatment of invaded patches vs. broadcast applications would reduce side effects of broadleaf herbicide application and increase compatibility with other management measures designed to improve rangeland conditions and restore grassland communities.  相似文献

ABSTRACT

Two Chilean soils were used to evaluate the performance of the nitrification inhibitor 3,4-dimetilpirazol phosphate (DMPP) added to ammonium-sulfate-nitrate (ASN) in comparison with traditional nitrogen (N) sources and different N-application forms. Two experiments were conducted: In the first, broccoli (Brassica oleracea var. italica) plants were cultivated in pots under greenhouse conditions, and received a N-fertilization equivalent to 150 Kg N ha?1 as ASN+DMPP (one application), urea (two splits), and sodium-potassium nitrate (three splits). In the second, ryegrass (Lolium spp.) plants were grown in pots under shading conditions. In this case, ASN+DMPP and urea were applied at N rates equivalent to 150 and 300 Kg N ha?1 in a single application. In the first experiment, ASN+DMPP increased dry-matter production, maintained a higher N content in the soil (at least until the middle of the growing period), and improved fertilizer N-use efficiency (FUE) in one soil. There were no significant differences in N-leaching losses. In the ryegrass experiment, ASN+DMPP increased dry-matter production and FUE, while N-leaching losses were reduced. Treatments with ASN+DMPP maintained higher N levels in soil throughout the growing period, and there were no significant differences in the available N fraction between the two N rates. The use of DMPP-containing fertilizers may be a good alternative for increasing FUE.  相似文献

  1. The Amazon basin has been subjected to extreme climatic events and according to climate change projections this hydrosystem could face changes in the natural dynamic of flood cycles that support the feeding and reproduction of many fish species, threatening aquatic biodiversity.
  2. Protected areas (PAs) are the main tools used to safeguard the biodiversity in the long term; however, they are fixed areas that could be subject to climate change, questioning their future efficiency in protecting biodiversity.
  3. The Amazon basin currently benefits from a relatively high level of protection as 52% of its catchment area is under the form of true PAs or indigenous lands. However, the capacity of these PAs to protect freshwater biodiversity remains unclear as they have generally been assessed with little regard to freshwater ecosystems and their hydrological connectivity. Here, the aim was to evaluate the effectiveness of PAs in representing the Amazon fish fauna under current and future climatic conditions.
  4. A macroecological approach was used to estimate the minimum size of the geographical range needed by each species to achieve long-term persistence, by a combined function of range size and body size, two ecological traits known to influence species extinction risk.
  5. In future the Amazon basin could risk losing 2% of its freshwater fish fauna owing to unsuitable climatic conditions, with a further 34% adversely affected. The present Amazon network of PAs will cover the minimum required range for species persistence for more than 60% of the freshwater fish species analysed under the future climate scenario. However, more than 25% of the future susceptible species are currently concentrated in large tributaries and in the central-lower Amazon floodplain where few PAs occur, highlighting the lack of appropriate conservation actions for these specific water bodies.
