Growth and photosynthetic performance were analyzed in alloplasmic tomato at a high- (25/17 °C; HTR) and low-temperature regime (12/6 °C; LTR) in order to establish the role of cytoplasmic variation on low-temperature tolerance of tomato (Lycopersicon esculentum Mill.). Four alloplasmic tomato lines, containing the nuclear genome of tomato and the plastome of L. hirsutum LA 1777 Humb. & Bonpl., an accession collected at high-altitude in Peru, were reciprocally crossed with 11 tomato entries
with a high inbreeding level and a wide genetic variation, resulting in a set of 44 reciprocal crosses. Irrespective of growth
temperature, alloplasmic families with alien chloroplasts of L. hirsutum (h) were on average characterized by a high shoot biomass, a large leaf area, and a low specific leaf area in comparison with
their euplasmic counterparts. These results do not directly point to an advantageous effect of h-chloroplasts on biomass accumulation at low temperature but rather towards a small general beneficial effect on growth and/or
distribution of assimilates. Significant chloroplast-related differences in photosynthetic performance, however, were not
detected at both temperature regimes, indicating that h-chloroplasts can properly function in a variable nuclear background of L. esculentum. It is concluded that chloroplast substitution is not an effective method for breeding tomato plants with improved low-temperature
tolerance.

Displacement imaging is a recent, powerful NMR method with which distributions of displacements can be acquired of e.g. fluids within a porous medium. Both motion parallel and perpendicular to the flow direction may be observed within a time window of a few milliseconds to several seconds. By combining displacement imaging with the line scan technique, one-dimensionally resolved measurements with a high temporal resolution ( < 1 min) of the spatial dependency of motion can be obtained. Here we present displacement images of flow through two simple model systems for soil: an unconsolidated glass bead water system and a sintered glass bead filter. It is demonstrated that the combination of displacement imaging and spatial resolution along a line is important to access both bulk displacement and local displacements in relation to the local porosity. 相似文献

Carvacrol is a component of several essential oils and has been shown to exert antimicrobial activity. The structural requirements for the activity of carvacrol were determined by comparison to structurally related (nonessential oil) compounds. Removal of the aliphatic ring substituents of carvacrol slightly decreased the antimicrobial activity. The effect of the hydroxyl group of carvacrol on activity could not be determined by simply comparing it to p-cymene, because this compound is immiscible with water; therefore, 2-amino-p-cymene, the amino analogue of carvacrol, which has a similar hydrophobicity and structural characteristics, was used. 2-Amino-p-cymene had similar membrane disruption and bacterial killing characteristics as carvacrol showing that, contrary to previous reports, the hydroxyl group of carvacrol itself is not essential for the antimicrobial activity. However, the observed 3-fold lower activity for 2-amino-p-cymene as compared to carvacrol indicates special features in the antimicrobial mode of action of carvacrol due to the hydroxyl group. 相似文献

In this paper the determination of the physical/rheological characteristics is described for a series of commercial galactomannans of which the structural details have been reported previously. Both solubility of the galactomannans and rheological properties of galactomannan solutions and galactomannan/xanthan mixtures were determined. Using a statistical analysis approach an attempt was undertaken to recognize correlations between structural and rheological data. The best correlation found was between the abundance of galactose substituents at a regular distance (type of galactomannan) and the storage modulus (G') of mixed galactomannan/xanthan gels, underscoring the hypothesis that branching hinders the formation of a network with xanthan gum. Also, the G' for the group of locust bean gums correlated with the degree of blockiness, that is, the size and occurrence of nonsubstituted regions on the mannose backbone. In addition, galactomannans displayed an apparent decrease in gelling ability with increasing average molecular weight. That G' also relates to the type of galactomannan can therefore partly be attributed to differences in average molecular weight for the various galactomannan types. However, within the series of locust bean gums tested, also an increase of G' with molecular weight was observed. This can be explained by the decreasing number of loose ends of the polymers and the concomitant increasing efficiency in network participation with increasing molecular weight. 相似文献

The course of the Maillard reaction between α-lactalbumin and various mono- and oligosaccharides in the solid state was studied using UPLC-ESI-TOF-MS. Individual reaction products were monitored for their degree of substitution per protein molecule (DSP). The Maillard reaction rate depended on the saccharide type and decreased when the saccharide size increased. Conjugation with charged saccharides was hindered when a specific average DSP was reached, probably resulting from electrostatic repulsion. The DSP varied between 0 and 15, and the standard deviation of the average DSP, which is a measure for product dispersity, increased to 1.9. Similar experiments were performed with a dipeptide. Relative reaction rates in these experiments were 1 for glucose, 0.28 for maltose, and 0.16 for maltotriose. Comparison of the results obtained using α-lactalbumin and the dipeptide made clear that the Maillard reaction rate is determined by a number of factors, including saccharide reactivity and lysine accessibility. 相似文献

Livestock is one of the growth sectors in the rural economy. In the third world the provision of livestock services for smallholders has generally been in the hands of the governments, leading to erratic, insufficient, and unreliable delivery systems. Especially in cases where the benefits of services accrue to the owners of the animals, privatization of some of the animal services may improve the delivery system. In order to explore the impact of such a private system, a group of sheep farmers in North Sumatra, Indonesia, were organized in an Animal Health Delivery Network (AHDN). The experience with AHDN indicates that the scope of private sector involvement is promising. Farmers were able to recognize the benefits and willing to pay for the drugs. However, they seem to be reluctant to apply the drugs themselves, thereby relying on the help of extension agents and traders. Selling drugs via poultry shops to farmers was not successful because drug wholesaler firms do not make their products available in small quantities. The study confirmed that the role of Government and private sector in the delivery of livestock services needs to be redefined.The authors are all members of the Small Ruminant Collaboration Research Support Program's (SR-CRSP) Economics Project being implemented by Winrock International Institute for Agricultural Development (WI) in the United States and the Center of Research Institutes for Animal Sciences (CRIAS) in Indonesia. The pig sector is struggling with negative attitudes of citizens. This may be the result of conflicting attitudes toward pig husbandry between citizens and other stakeholders. To obtain knowledge about these attitudes, the objectives of this study were (1) to determine and compare attitudes of various stakeholders toward animals, humans and the environment in the context of pig husbandry and (2) to determine and compare the acceptability of publically discussed issues related to pig husbandry of various stakeholders. A questionnaire was distributed to citizens, conventional pig farmers, organic pig farmers, pig husbandry advisors and pig veterinarians. Respondents could indicate their attitude toward aspects related to animals, humans and the environment in the context of pig husbandry and they could indicate their opinion about the acceptability of issues of pig husbandry, e.g. piglet mortality and inside pig housing. Based on measured attitudes and the acceptability of issues, the studied stakeholders could be divided into three distinctive groups. The group of citizens and organic pig farmers showed negative attitudes toward all aspects of pig husbandry, the group of conventional pig farmers and pig husbandry advisors only showed negative attitudes toward aspects related to economics and the group of pig veterinarians showed negative attitudes to specific aspects of pig husbandry. This indicates that stakeholders have different interests and different perspectives with regard to pig husbandry. The fungus Cladosprium fulvum is a biotrophic leaf pathogen of tomato. The fungus develops in the intercellular space without forming specialized feeding structures and does not affect the leaf tissue. The outcome of the C. fulvum-tomato interaction can be described by the gene-for-gene model. Failure of infection is expressed by a hypersensitive response. Two fungal proteins, ECP1 and ECP2, have been isolated and their corresponding genes have been cloned. In a compatible interaction including many physiological races ECP1 and ECP2 are highly produced and a role in pathogenicity is suggestive. The ecp1 gene shows some homology with tumor necrosis factor receptors (TNFRs) while the ecp2 gene shows no homology with sequences known in data bases. However, disruption of one of the two genes showed no reduced pathogenicity of the fungus. Two race-specific elicitors, AVR4 and AVR9, have been isolated and their corresponding genes have been cloned. The avirulence genes Avr4 and Avr9 are only present in C. fulvum avirulent on Cf-4 and Cf-9 cultivars, respectively. The expression of these two genes is, like the expression of the ecp genes, highly induced when the fungus grows in planta. Disruption of the Avr9 gene in wild type avirulent races leads to virulence on tomato genotypes carrying the complementary resistance gene Cf-9. A single base-pair change in the avirulence gene Avr4 leads to virulence on tomato genotypes carrying the Cf-4 resistance gene. Isolation, characterization and possible function of ECP1, ECP2, AVR4, and AVR9 will be discussed. 相似文献
