Boiled seed hardness is an important factor in the processing of soybean food products such as nimame and natto. Little information is available on the genetic basis for boiled seed hardness, despite the wide variation in this trait. DNA markers linked to the gene controlling this trait should be useful in soybean breeding programs because of the difficulty of its evaluation. In this report, quantitative trait locus (QTL) analysis was performed to reveal the genetic factors associated with boiled seed hardness using a recombinant inbred line population developed from a cross between two Japanese cultivars, ‘Natto-shoryu’ and ‘Hyoukei-kuro 3’, which differ largely in boiled seed hardness, which in ‘Natto-shoryu’ is about twice that of ‘Hyoukei-kuro 3’. Two significantly stable QTLs, qHbs3-1 and qHbs6-1, were identified on chromosomes 3 and 6, for which the ‘Hyoukei-kuro 3’ alleles contribute to decrease boiled seed hardness for both QTLs. qHbs3-1 also showed significant effects in progeny of a residual heterozygous line and in a different segregating population. Given its substantial effect on boiled seed hardness, SSR markers closely linked to qHbs3-1, such as BARCSOYSSR_03_0165 and BARCSOYSSR_03_0185, could be useful for marker-assisted selection in soybean breeding. 相似文献

This study examined the factors that influence the size of a soil seed bank in Robinia pseudoacacia stands. We proposed two hypotheses: the amount of buried seeds of R. pseudoacacia is proportional to stand biomass rather than stand age, and apicultural utilization influences the size of the soil seed bank. R. pseudoacacia generally produces seeds with various degrees of physical dormancy. In addition, this tree is short-lived, and fecundity declines after 30–40 years in relation to tree vigor. However, R. pseudoacacia is intensively used in apiculture as an important honey source, and supply of honeybees by beekeepers could influence seed crops. We investigated 25 plantations in Hokkaido (Central, Hidaka, and Oshima) and in Nagano, Japan, where many naturally regenerated R. pseudoacacia stands occur. We found great variation in the size of the soil seed bank among stands, ranging from 13,757 to 6.4 seeds m−2 per plot. A generalized linear mixed model revealed that both basal area (BARp) and apicultural utilization best explained the size of the soil seed bank. Both the positive effect of BARp and small contribution of stand age to the model implied that the soil seed bank is transitory, and will not persist for very long time. The large contribution of apicultural utilization to the size of the soil seed bank implied pollinator limitation under natural conditions. R. pseudoacacia often has large flower crops, and native pollinators can not keep up with the demand for pollination. Thus, the supply of honeybees by beekeepers should improve the pollination success of R. pseudoacacia. From these results, we conclude that we must consider stand history with respect to apicultural utilization if the land is harvested or a stand declines. 相似文献

Fowl adenoviruses (FAdVs) and avian reoviruses (ARVs) are ubiquitous in poultry farms and most of them are not pathogenic, yet often cause damage to chicks. A total of 104 chicken fecal samples were collected from 7 farms of breeder chickens (layers and broilers) in Japan from 2019 to 2021, and yielded 26 FAdV plus 14 ARV isolates. By sequencing, FAdV isolates were classified as FAdV-1, 5 and 8b. ARV isolates were classified as genotype II, IV and V. These results suggest that FAdVs and ARVs are resident in the breeder chicken farms in Japan. 相似文献

The pharmacokinetic endpoint of a 25-fold increase in human exposure is one of the specified criteria for high-dose selection for 2-year carcinogenicity studies in rodents according to ICH S1C(R2). However, this criterion is not universally accepted for 6-month carcinogenicity tests in rasH2-Tg mice. To evaluate an appropriate multiple for rasH2-Tg mice, we evaluated data for 53 compounds across five categories of rasH2-Tg mouse-positive [(1) genotoxic and (2) non-genotoxic] carcinogens and rasH2-Tg mouse-negative [(3) non-genotoxic carcinogens with clear or uncertain human relevance; (4) non-genotoxic rodent-specific carcinogens; and (5) non-carcinogens], and surveyed their tumorigenic activities and high doses in rasH2-Tg mice and 2-year rodent models. Our survey indicated that area under the curve (AUC) margins (AMs) or body surface area-adjusted dose ratios (DRs) of tumorigenesis in rasH2-Tg mice to the maximum recommended human dose (MRHD) were 0.05- to 5.2-fold in 6 category (1) compounds with small differences between models and 0.2- to 47-fold in 7 category (2) including three 2-year rat study-negative compounds. Among all 53 compounds, including 40 compounds of the rasH2-Tg mouse-negative category (3), (4), and (5), no histopathologic risk factors for rodent neoplasia were induced only at doses above 50-fold AM or DR in rasH2-Tg mice except for two compounds, which induced hyperplasia and had no relationship with the tumors observed in the rasH2-Tg mouse or 2-year rodent studies. From the results of these surveys, we confirmed that exceeding a high dose level of 50-fold AM in rasH2-Tg mouse carcinogenicity studies does not appear to be of value. 相似文献

Night break treatment was applied to Eustoma grandiflorum ‘Nail Peach Neo’ using light sources with different red (R: 660 ± 30 nm): far-red (FR: 730 ± 30 nm) ratios or FR light intensities in order to investigate growth and flowering responses. Flower initiation and induction were promoted by night break treatment with a low R:FR light source, but was delayed by a high R:FR ratio. The promotion or delay of flower bud formation was accompanied by a decrease or an increase, respectively, in the number of nodes on the main stem at anthesis to the first floret. The difference between date of visible bud with plants under night break treatment and that of the control was approximated with high accuracy by a sigmoid function of the logarithms of R:FR ratio. The threshold R:FR ratio demarcating the promotion and delay of date of visible bud was about 5.3 under the experimental conditions used. The critical R:FR ratios for promotion or delay of visible bud would be about 0.5 and 50.0, respectively. In addition, the time from planting to visible bud was approximated with an exponential function of FR light intensity. The maximum acceleration of date of visible bud by long-day treatment would be 20 days, and the critical FR light intensity would be 2.0 μmol m−2 s−1. It is concluded that growth and flowering of E. grandiflorum can be regulated by long-day treatment using light sources with different R:FR ratios or FR light intensities. 相似文献

Nitrogen and carbon dynamics in paddy and upland soils for rice cultivation and in upland soil for corn cultivation was investigated by using 13C and 15N dual-labeled cattle manure compost (CMC). In a soil with low fertility, paddy and upland rice took up carbon and nitrogen from the CMC at rates ranging from 0.685 to 1.051% of C and 17.6–34.6% of N applied. The 13C concentration was much higher in the roots than in the plant top, whereas the 15N concentration differed slightly between them, indicating that organic carbon taken up preferentially accumulated in roots. The 13C recovery in the plant top tended to be higher in upland soil than in paddy soil, whereas 15N applied was recovered at the same level in both paddy and upland soils. In the experiment with organic farming soil, paddy rice took up C and N from the CMC along with plant growth and the final recovery rates of 13C and 15N were 2.16 and 17.2% of C and N applied. In the corn experiment, a very large amount of carbon from the CMC was absorbed, accounting for at least 7 times value for rice. The final uptake rates of 13C and 15N reached about 13 and 10% of C and N applied, respectively. Carbon emission from the CMC sharply increased by 2 weeks after transplanting and the nitrogen emission was very low. It is concluded that rice and corn can take up an appreciable level of carbon and nitrogen from the CMC through roots. 相似文献

Abstract Beets were grown on soils with various exchangeable sodium percentages (ESP). A saline non-sodic soil (SA, ESP = 3.2), a saline sodic soil (SO, ESP = 23), and a saline high sodic soil (HSO, ESP = 78) were prepared from Tottori sand dune soil (CO). K-type and Ca-type artificial zeolites (50 g kg?1) were applied to these soils in order to evaluate their effects on the chemical properties of saturation extracts of the soils, water deficit, cation uptake and transport, and cation balance of beet plants. In the zeolite-free treatments, beet growth was accelerated in SA and SO, but was suppressed in HSO compared with CO. The addition of both types of zeolites ameliorated plant growth in all the soils studied, especially HSO. The relative dry weight of the soils treated by the K-type zeolite to the zeolitefree soil was 189% for CO, 125% for SA, 130% for SO, and 222% for HSO. For the soils treated with the Ca-type zeolite, the values were 169, 116, 132, and 341%, respectively. In SA, SO, and HSO, the addition of the K-type zeolite increased the K uptake due to the increase of the K concentration of saturation extracts of soils. The addition of the Ca-type zeolite increased the Ca uptake due to the increase in the Ca concentration of the saturation extracts of soils which was accompanied by an increase in the K uptake. The increase in the uptake of K or Ca and decreased in the transport of Na by the addition of both types of zeolites improved the cation balance of the plants. The Ca-type zeolite did not increase the water deficit even though it increased the electric conductivity in all the soils. The results indicated that both types of artificial zeolites were able to improve the growth performance of beets in saline and sodic soils and that the K-type zeolite could be used as a K-fertilizer as well. 相似文献

The enantiomers of 6-fluoro-, 6-bromo-, and 6-iodopericosine A were synthesized. An efficient synthesis of both enantiomers of pericoxide via 6-bromopericosine A was also developed. These 6-halo-substituted pericosine A derivatives were evaluated in terms of their antitumor activity against three types of tumor cells (p388, L1210, and HL-60) and glycosidase inhibitory activity. The bromo- and iodo-congeners exhibited moderate antitumor activity similar to pericosine A against the three types of tumor cell lines studied. The fluorinated compound was less active than the others, including pericosine A. In the antitumor assay, no significant difference in potency between the enantiomers was observed for any of the halogenated compounds. Meanwhile, the (−)-6-fluoro- and (−)-6-bromo-congeners inhibited α-glucosidase to a greater extent than those of their corresponding (+)-enantiomers, whereas (+)-iodopericosine A showed increased activity when compared to its (−)-enantiomer. 相似文献

Herbicide-resistant transgenic soybean plants hold a leading market share in the USA and other countries, but soybean has been regarded as recalcitrant to transformation for many years. The cumulative and, at times, exponential advances in genetic manipulation have made possible further choices for soybean transformation. The most widely and routinely used transformation systems are cotyledonary node–Agrobacterium-mediated transformation and somatic embryo–particle-bombardment-mediated transformation. These ready systems enable us to improve seed qualities and agronomic characteristics by transgenic approaches. In addition, with the accumulation of soybean genomic resources, convenient or promising approaches will be requisite for the determination and use of gene function in soybean. In this article, we describe recent advances in and problems of soybean transformation, and survey the current transgenic approaches for applied and basic research in Japan. 相似文献
