Plasmodium spp. protozoa cause malaria and are known to infect humans and a variety of animal species including macaque monkeys. Here we report both our experience with malaria recrudescence in cynomolgus monkeys (Macaca fascicularis) in a toxicity study and the results of a survey on Plasmodium infection in cynomolgus monkeys imported to Japan for laboratory use. A cynomolgus monkey from the toxicity study presented with severe anemia and Plasmodium protozoa in erythrocytes on a thin blood smear and was subsequently diagnosed with symptomatic malaria. In this animal, congestion and accumulation of hemozoin (malaria pigment) in macrophages were noted in the enlarged and darkly discolored spleen. As a follow-up for the experience, spleen sections from 800 cynomolgus monkeys in toxicity studies conducted between 2003 and 2013 were retrospectively examined for hemozoin deposition as a marker of Plasmodium infection. The origin of the animals included Cambodia, China, Indonesia, and Vietnam. Hemozoin deposition was confirmed in 44% of all examined monkeys. Monkeys from Indonesia showed the highest incidence of hemozoin deposition (approx. 80%). A high prevalence of Plasmodium infection in laboratory monkeys was also confirmed with polymerase chain reaction (PCR) by using Plasmodium genus-specific primers. Although Japan is not a country with endemic malaria, it is important to be aware of the prevalence and potential impact of background infection with Plasmodium spp. and recrudescence of symptomatic malaria in imported laboratory monkeys on pharmaceutical toxicity studies.  相似文献

Bending strength and toughness of heat-treated wood   总被引:9,自引:0,他引:9  
The load-deflection curve for static bending and the force-time curve for impact bending of heat-treated wood were examined in detail. The effect of oxygen in air was also investigated. Sitka spruce (Picea sitchensis Carr.) was heated for 0.5–16.0h at a temperature of 160°C in nitrogen gas or air. The dynamic Young's modulus was measured by the free-free flexural vibration test, the static Young's modulus and work needed for rupture by the static bending test, and the absorbed energy in impact bending by the impact bending test. The results obtained were as follows: (1) The static Young's modulus increased at the initial stage of the heat treatment and decreased later. It decreased more in air than in nitrogen. (2) The bending strength increased at the initial stage of the heat treatment and decreased later. It decreased more in air than in nitrogen. (3) The work needed for rupture decreased steadily as the heating time increased. It decreased more in nitrogen than in air. It is thought that heat-treated wood was more brittle than untreated wood in the static bending test because W12 was reduced by the heat treatment. This means that the main factors contributing to the reduction of the work needed for rupture were viscosity and plasticity, not elasticity. (4) The absorbed energy in impact bending increased at the initial stage of the heat treatment and decreased later. It decreased more in air than in nitrogen. It was concluded that heat-treated wood became more brittle in the impact bending test becauseI 12 andI 23 were reduced by the heat treatment.  相似文献

The proliferative potential of a spinal nephroblastoma was studied in a young dog. A 4-month-old, female golden retriever showed developing deterioration in her gait and subsequent paralysis of her hind legs. At necropsy, a well-demarcated grayish brown tumor mass was found in the lumbar spinal cord segments between L2 and L3. Histologically, a blastemal cell tumor with a tubule- or glomeruli-like structure was found to be infiltrating intradurally. Proliferating cells at the S-phase, assessed using the bromodeoxyuridine (BrdU) labeling method, were seen occasionally in the tubular cells and glomeruli-like structures and were frequently seen in the blastemal cells. Immunohistochemically, the tubular epithelial cells were positive for cytokeratin, and the blastemal cells were positive for vimentin. The present tumor showed a high potential for growth and invasion, which suggests that it the potential to expand into the adjacent spinal cord.  相似文献

Six clones of monoclonal antibodies (Mabs) to fowl adenovirus (FAV) serotype 1 were produced. All Mabs reacted positively by enzyme-linked immunosorbent assay. Three Mabs recognized the putative 100-kD hexon protein and reacted to serotype 1 specifically by western blot analysis but did not react to other FAV serotypes (2, 3, 4, 5, 6, 7, and 8a). These Mabs will be useful for immunodiagnosis of FAV serotype 1 infection in chickens with gizzard erosion and in further research studies involving the genomes and proteins of FAV serotype 1.  相似文献

Band 3 deficiency with hereditary spherocytosis and hemolytic anemia in Japanese black cattle, band 3(Bov.Yamagata), is caused by a total lack of band 3 protein with an autosomal dominant inheritance. Genotyping for band 3 deficiency and sexing were successfully achieved in biopsied embryo cells with efficiencies of 98.4% and 97.4%, respectively. Transfer of the embryo that was determined as homozygous for the mutant allele into a recipient cow resulted in the production of a fetus exhibiting the genotype and red cell phenotypes characteristic of band 3(Bov.Yamagata). These results demonstrate that our procedure is reliable and applicable to produce animals free from or homozygous for the mutant allele by breeding carrier animals.  相似文献

SUMMARY: The effects of nutrient salts eluted from artificial algal reef blocks made with sea-bottom sediment (hedoro) and cement to the growth and survival rate of the gametophytes and juveniles of Eisenia bicyclis were investigated. When each sample block was submerged in filtered seawater for 28 days, the nutrient salt content from the hedoro increased with time and, in particular, the amounts of NO3− N and NO2− N reached a constant level on day 10. We observed that the lower the cement ratio, the higher the concentrations of eluted NH4+− N and PO43−− P. The growth of gametophytes as well as juveniles was highest in the elution of the block with the lowest cement ratio of 30%. In contrast, the survival rate of gametophytes and juveniles decreased linearly with time and the survival rate was higher for the hedoro blocks than for the block made of mortar.  相似文献

Summary In our previous report of a series of experiments involving ordinary grafting, virus-inoculated grafting and DNA treatment in Capsicum annuum L., we described the variants as well as gene analyses of individual variants, together with the characteristics of the graft-induced genetic changes obtained.Microhistological analysis of the stock stems revealed that chromatin masses, stained to greenish blue and in various sizes and shapes, were moving through cell wall and intercellular space from the lignifying and dying cells, stained to lighter purplish brown, towards the vascular bundles (Fig. 1). I discussed the mechanisms of chromatin transfer from the stock dying cells through the vascular system across the graft-union to the growing point(s) of the scion (Fig. 3), and how that process causes transformation in the fast dividing scion flower primordia (Fig. 4).The significance of chromatin translocation can be understood in the following way: Genetic information is never transmitted from the stock to the scion, unless a DNA molecule larger than a functional unit such as a gene translocates from the stock, and unless the DNA molecules per se are integrated into new cell nuclei of the scion. In other words, no transmission of genetic information will take place, if DNA molecules of the stock disintegrated to nucleotides, as when a protein molecule is digested into amino acids, and are utilized by the scion simply as raw material in the formation of new cell nuclei with its own DNA strand as template.Also discussed were the significance of mentor methods, and the importance of distinguishing between horticultural graft and genetical graft (Fig. 2).  相似文献

Electrolysis of a 0.1% (17.1 mM) solution of NaCl using separate anode and cathode compartments gives rise to solutions containing active chemical species. The strongly acidic "anode solution" (EW+) has high levels of dissolved oxygen and available chlorine in a form of hypochlorous acid (HOCl) with a strong potential for sterilization, which we have investigated here. Exposing Aspergillus parasiticus at an initial density of 10(3)spores in 10 microL to a 50-fold volume (500 microL) of EW+ containing ca. 390 micromol HOCl for 15 min at room temperature resulted in a complete inhibition of fungal growth, whereas the cathode solution (EW-) had negligible inhibitory effects. Moreover, the mutagenicity of aflatoxin B(1) (AFB(1)) for Salmonella typhimurium TA-98 and TA-100 strains was strongly reduced after AFB(1) exposure to the EW+ but not with the EW-. In high-performance liquid chromatography analysis, the peak corresponding to AFB(1) disappeared after treatment with the EW+, indicating decomposition of the aflatoxin. In contrast, the routinely used disinfectant sodium hypochlorite, NaOCl, of the same available chlorine content as that of EW+ but in a different chemical form, hypochlorite (OCl-) ion, did not decompose AFB(1) at pH 11. However, NaOCl did decompose AFB(1) at pH 3, which indicated that the principle chemical formula to participate in the decomposition of AFB(1) is not the OCl- ion but HOCl. Furthermore, because the decomposition of AFB(1) was suppressed by pretreating the EW+ with the OH radical scavenger thiourea, the chemical species responsible for the AFB(1)-decomposing property of the EW+ should be at least due to the OH radical originated from HOCl. The OH in EW+ was proved by electron spin resonance analysis.  相似文献
