How well does a botanical garden collection of a rare palm capture the genetic variation in a wild population? 总被引:1,自引:0,他引:1

Conservation is increasingly central to the botanic garden mission. Living plant collections are important components of conservation. Critical evaluation of living conservation collections with population genetic analysis can directly inform ex situ conservation strategy. Here, we quantify the degree of genetic variation captured through a population-based collection protocol, and explore optimal sampling for ex situ conservation. An extensive living collection derived from one population of Leucothrinax morrisii (Arecaceae) provided a model system. We compared 58 specimens from the ex situ collection with 100 individuals from throughout the parent population via 6 ISSR loci. Random bootstrapped resamples of the data were made to model differently structured ex situ collections. Mean diversity (He) differed little between the collection (0.204) and the population (0.216), and genetic distance (D) was very close (0.036). Very few private alleles were found between the collection and the population. Allelic capture, as measured by percent of private alleles, was greater than 94%. Resampled collections of different sizes captured from 48% to 94% of alleles. Pairwise comparison of bootstrapped resamples suggests that increasing the representation of half-sibling groups does not significantly increase allele capture. Increase in allele capture with increasing sample size is greatest at low resample sizes, and showed diminishing returns as resample size increased. No appreciable increase in allele capture was gained through maintaining different half-sibling groups. These data inform sampling for ex situ conservation purposes, and recommend sample sizes of at least 15 individuals, with the upper limit based on resources. 相似文献

CENH3 is a centromere-specific histone H3 variant essential for kinetochore assembly. Despite its central role in centromere function, there has been no conclusive evidence supporting CENH3 as sufficient to determine centromere identity. To address this question, we artificially targeted Drosophila CENH3 (CENP-A/CID) as a CID-GFP-LacI fusion protein to stably integrated lac operator (lacO) arrays. This ectopic CID focus assembles a functional kinetochore and directs incorporation of CID molecules without the LacI-anchor, providing evidence for the self-propagation of the epigenetic mark. CID-GFP-LacI-bound extrachromosomal lacO plasmids can assemble kinetochore proteins and bind microtubules, resulting in their stable transmission for several cell generations even after eliminating CID-GFP-LacI. We conclude that CID is both necessary and sufficient to serve as an epigenetic centromere mark and nucleate heritable centromere function. 相似文献

The hydrolysis of trans-permethrin and α-naphthyl acatate by midgut, fat body, and cuticle homogenates from Pseudoplusia includens (Walker) was monitored during the development of the last instar. The midgut homogenates appeared to have two pH optima (7.6 and 8.6) for the hydrolysis of trans-permethrin, the fat body homogenates had one optimum (7.4–7.8), and the cuticle homogenates had a major optimum at 6.6. Hydrolysis of both substrates peaked during the late feeding stages for midgut and cuticle homogenates, although relative changes were not the same. Hydrolysis of trans-permethrin peaked during the late feeding stage in fat body homogenates, while hydrolysis of α-naphthyl acetate continually increased through the prepupal stage. Thus, the hydrolysis of α-naphthyl acetate is not necessarily associated with the hydrolysis of trans-permethrin. The LD50 values for trans-permethrin on the different stages appeared to reflect the influence of hydrolysis. 相似文献

The aim of this study was to determine the pharmacokinetics of dexamethasone in broiler chickens. Dexamethasone sodium phosphate (0.3 mg/kg bodyweight) was injected IV or IM and blood samples were collected at 0, 0.25, 0.5, 0.75, 1, 2, 4, 6, 8, 10, 12 and 24 h after administration. Dexamethasone in the plasma samples was measured using a liquid chromatography–tandem mass spectrometry method and the pharmacokinetics analysed according to a one-compartmental model.The maximum plasma concentration after IM administration occurred at 0.37 h. The elimination half-life for dexamethasone was 0.46 h and 0.70 h following IV and IM administration, respectively, which was shorter than other species, while the clearance (1.26 L/h kg) was higher than has been reported for other species (<0.5 L/h kg). The volume of distribution (~1 L/kg) was similar to values reported for other species and the bioavailability of dexamethasone after IM administration was 100%. The results from this study will be useful in investigating whether inflammatory disease may affect the pharmacokinetic parameters of dexamethasone in chickens. 相似文献

- 1. Chalk stream floodplains are extensively managed and the high degree of connectivity associated with water meadows has been severely reduced. The main contributor to aquatic biodiversity is perceived to be the river itself. Ditches in these areas have been largely ignored as a contributor to total diversity.
- 2. Separate surveys of instream, bankside and ditch macroinvertebrates in a small area of floodplain provided a unique opportunity to assess the relative contribution of each locality to total aquatic diversity. The fauna and flora of a ditch on the lower River Frome floodplain was examined and compared with studies of adjacent bankside and instream habitat. A total of 145 macroinvertebrate taxa and 61 plant species were recorded from the ditch. Faunal abundance and richness were positively related to the diversity of aquatic plants. The ditch showed a clear linear trend in community type that appeared to be partly related to the acidic seepage from tertiary deposits at the source and percentage cover of vegetation.
- 3. There were large differences in the numbers of taxa collected in ditch, bankside, instream mesohabitat and RIVPACS surveys. Of the total 202 taxa recorded in the four surveys, the ditch contributed 59 unique taxa with 25 from the bankside and two and three from the mesohabitat and RIVPACS surveys, respectively.
- 4. The large contribution of the Rushton Ditch to aquatic biodiversity on the floodplain is due to three main factors: the chemically different and slow‐flowing environment, the connection to the main river, and a management regime which has halted hydroseral succession at a point where vegetation structure is for the most part open and able to support a wide range of ecological niches. The maintenance of these conditions requires sensitive management and further knowledge of the needs and interactions of faunal communities with vegetation.

Almaco jack (Seriola rivoliana Valenciennes) is an excellent candidate for aquaculture due to its fast growth rate and high market value. While S. rivoliana have adapted well to captivity, survival at early life stages can be improved to increase profitability during production. A wide range of variables cause larval mortalities but high bacterial loads in rearing tanks are often correlated with these losses. The aim of this study was to investigate the effect of egg disinfection on bacterial load and hatch rate of S. rivoliana. Disinfectants tested included formalin (F100 and F200; 100 and 200 mg/L, respectively, for 60 min), hydrogen peroxide (HPO; 300 mg/L for 10 min) and peracetic acid/hydrogen peroxide (PAA/HPO; 15.7 mg/L/39.6 mg/L for 1 min). Concentrations and contact times were determined based on current use in marine aquaculture and preliminary trials. Eggs treated with HPO and F100 had significantly higher hatch rates than the untreated control group. All treatments significantly decreased total Vibrio counts compared to untreated eggs; however, total bacterial counts were only decreased following treatments with PAA/HPO and F200. To prevent egg mortality due to bacterial overgrowth, consideration should be given to the use of surface disinfection using HPO or F100. Future studies should investigate the use of peracetic‐based products at lower doses. 相似文献

To determine the effect of swine hepatitis E virus (HEV) infection on pregnant gilts, their fetuses, and offspring, 12 gilts were intravenously inoculated with swine HEV. Six gilts, who were not inoculated, served as controls. All inoculated gilts became actively infected and shed HEV in feces, but vertical transmission was not detected in the fetuses. There was no evidence of clinical disease in the gilts or their offspring. Mild multifocal lymphohistiocytic hepatitis was observed in 4 of 12 inoculated gilts. There was no significant effect of swine HEV on fetal size, fetal viability, or offspring birth weight or weight gain. The offspring acquired anti-HEV colostral antibodies but remained seronegative after the antibodies waned by 71 days of age. Swine HEV infection induced subclinical hepatitis in pregnant gilts, but had no effect on the gilts' reproductive performance, or the fetuses or offspring. Fulminant hepatitis associated with HEV infection was not reproduced in gilts. 相似文献

In Eurasian perch (Perca fluviatilis), the variability in spawning quality is a major limiting factor for successful production, especially when breeders are fed with an artificial diet. The influence of the dietary DHA/EPA/AA ratio on the egg and larval quality and on the fatty acid and lipid class composition of eggs has been investigated in perch broodstock. Two experimental diets (16% lipids) with two different DHA/EPA/AA ratios, D1 (3/2/2) and D2 (23/9/1), were compared with a natural diet consisting of cultured carp juveniles, CC (10/10/1) and with a commercial diet for salmonids, CDS (14/16/1). Percentages of fertilization and hatching were comparable between fish fed D1, D2 and CC, with the highest hatching rate observed for D1 (63.5 ± 3.8%). These diets supported better values than the CDS. Larval survival and TL50 observed after osmotic stress were higher for the D1 group, followed by larvae produced by fish fed D2 and CC. Larvae from fish fed D1, D2 and CC were significantly more robust than larvae from the CDS group. Differences were observed regarding the fatty acid (FA) profile in the eggs, which was related to the dietary FA composition. The results indicate that a ratio of 3/2/2 seemed to be effective for obtaining eggs and larvae of good quality. 相似文献

Hospital-based infection control in veterinary medicine is emerging and the role of the environment in hospital-acquired infections (HAI) in veterinary hospitals is largely unknown. This study was initiated to determine the recovery of Escherichia coli and selected veterinary and zoonotic pathogens from the environments of 101 community veterinary hospitals. The proportion of hospitals with positive environmental swabs were: E. coli--92%, Clostridium difficile--58%, methicillin-resistant Staphylococcus aureus (MRSA)--9%, CMY-2 producing E. coli--9%, methicillin-resistant Staphylococcus pseudintermedius--7%, and Salmonella--2%. Vancomycin-resistant Enterococcus spp., canine parvovirus, and feline calicivirus were not isolated. Prevalence of antimicrobial resistance in E. coli isolates was low. Important potential veterinary and human pathogens were recovered including Canadian epidemic strains MRSA-2 and MRSA-5, and C. difficile ribotype 027. There is an environmental reservoir of pathogens in veterinary hospitals; therefore, additional studies are required to characterize risk factors associated with HAI in companion animals, including the role of the environment. 相似文献
