建立了同时测定番茄中多菌灵及其主要代谢物(2-氨基苯并咪唑和苯并咪唑)的超高效液相色谱-串联质谱(UPLC-MS/MS)分析方法。番茄样品用乙腈超声提取,经浓缩后用初始流动相稀释并定容,过0.22 μm滤膜后进行UPLC-MS/MS分析。采用多反应监测(MRM)模式测定,外标法定量。结果表明:在添加水平为0.05~5 mg/kg下,多菌灵及其主要代谢物的平均回收率为70.5%~104.9%,相对标准偏差为2.6%~9.5%。方法的检出限(LOD)为0.16~4.73 μg/kg,定量限(LOQ)为0.29~4.99 μg/kg。 相似文献

AIM: To investigate the aggravating effect of albumin overload on the kidney injury induced by lipid nephrotoxicity, and to observe the renoprotective effect of simvastatin (SIMV) on adriamycin nephropathy (ADR) mice.METHODS: SPF healthy male BALB/c mice were randomly divided into control group, ADR group, ADR with bovine serum albumin (BSA) overload (ADR+BSA) group, and ADR with both BSA overload and SIMV treatment (ADR+BSA+SIMV) group. All mice were uninephrectomized under general anesthesia 2 weeks before setting up ADR model. ADR+BSA model started to be set up 4 weeks later. At the end of the 0th, 2nd, 6th, 10th and 14th weeks, 24 h urinary protein was evaluated. At the end of the 14th week, the serum biochemical indexes and the kidney pathological changes were observed, and glomerulosclerotic index (GSI) was also evaluated. The cholesterol in the kidney was measured by enzymic colorimetric method and oil red O staining. The expression of IL-1β, TGF-β1 and low-density lipoprotein receptor (LDLr) in the kidney tissues was determined by real-time PCR. The expression of IL-1β and IL-17 was measured by immunohistochemistry and the expression of IL-17 in the kidney was measured by ELISA.RESULTS: Compared with control group, the expression of IL-1β, TGF-β1, IL-17 and LDLr, and cholesterol content in the kidney and the GSI were all significantly increased in ADR group (P<0.05). Compared with ADR group, 24 h urinary protein, serum creatinine, the expression of IL-1β, IL-17 and LDLr, and cholesterol content in the kidney were all significantly increased in ADR+BSA group (P<0.05). Treatment with SIMV significantly decreased the expression of IL-1β, TGF-β1, IL-17 and LDLr. The accumulation of cholesterol in the kidney and the GSI were also decreased (all P<0.05).CONCLUSION: Inflammation aggravates the lipid deposition and glomerular sclerosis by increasing the expression of LDLr in ADR mice. Albumin overload further accelerates the progressive kidney damage by regulating the expression of IL-1β, TGF-β1 and IL-17, which promotes the increase in LDLr. The beneficial effect of SIMV might be mediated by its anti-inflammatory effect. 相似文献

Improved protocol for Agrobacterium mediated transformation of tomato (Lycopersicon esculentum) Micro-Tom was developed to use in corporation of the carotenoid biosynthetic genes CsZCD (Crocus zeaxanthin 7,8-cleavage dioxygenase). From these experiment, a transformation methodology using explants from cotyledons cultured for 1 day on the medium with zeatin 2 mg/L, IAA 0.1 mg/L, carefully submerged in the Agrobacterium inoculum for 20 min, then concultured with the agrobacterium for 3 days on the same medium, followed by a transfer to the same medium with 500 mg/L cefotaxin for 3 days and then by a transfer to the same medium with 100 mg/L kanamycin and 500 mg/L carabenillin for 6–8 weeks and resulted in a greater than 20% transformation efficiency in the concentration of Agrobacterium OD600 = 0.2 tested. In this transformation method, no feeder layers were used and the subculture media was minimal. Among the Agrobacterium concentrations of OD600 = 0.2, 0.5 and 1.0, the best transformation efficiency, 20.87%, was obtained by using OD600 = 0.2, which was significantly higher than that of OD600 = 1.0. The presence of the inserted target genes was checked using a rapid and efficient PCR test. The protocol was successfully employed in the production of transgenic Micro-Tom tomato containing the carotenoid biosynthesis CsZCD under constructive promoter. 为探讨犬的乳腺肿瘤组织中PTEN和VEGF蛋白表达与临床病理学特征关系的相关性,采用免疫组织化学SP染色法检测了32例乳腺肿瘤组织及6例正常乳腺组织中上述2种蛋白的表达。结果发现:1)PTEN蛋白在正常乳腺组织和良性乳腺肿瘤高表达率为100%,在恶性乳腺肿瘤组织中的高表达率为67%(8/12),两者比较差异极显著(P<0.01);乳腺肿瘤组织中PTEN蛋白表达与犬的年龄、肿瘤大小无关(P>0.05),而与淋巴结转移、组织学分级有关(P<0.01)。2)VEGF蛋白在正常乳腺组织中高表达率为33.3%(2/6),在乳腺肿瘤组织中高表达率为78%(25/32),两者比较差异极显著(P<0.01);乳腺肿瘤组织中VEGF蛋白表达与患病犬的年龄、肿瘤大小无关(P>0.05),而与淋巴结转移有关(P<0.05)。联合检测PTEN、VEGF表达可作为乳腺癌生物学行为和预后判断的重要指标。 相似文献

Because of its high nutritional value and health benefits, aquaculture production of Apostichopus japonicus in China is the largest of any single species. Therefore, the development of new farming methods is of considerable significance. In this study, discarded oyster shells have been used to create an artificial reef for the culture of this species. The results have shown that from 6th March 2009 to 26th November 2009, the wet weight of sea cucumber increased from 49.57 ± 1.16 to 79.87 ± 1.46 g ind?1. Between 16th July and 18th October, the specific growth rate and daily weight gain of A. japonicus differed significantly from other periods. Population density was higher within the reef compared with outside the reef area, and the difference was significant (P < 0.01). The maximum distance between A. japonicus individuals within the reef area on 3rd March and 16th July was 65.0 ± 3.3 and 62.9 ± 4.4 cm, respectively, but the difference was not significant (P > 0.05). The diatom species attached to the oyster shells were similar to those found in the stomach content of A. japonicus. In conclusion, the oyster‐shell reef provides a suitable habitat and shelter for the culture of the sea cucumber, A. japonicus. 相似文献
