The present work investigated the ability of inactive brown seaweed, Turbinaria conoides, to biosorb aluminum(III) and cadmium(II) ions in both single and binary systems. Initial experiments were undertaken to determine the influence of pH and biosorption isotherms of each metallic ion. Owing to the presence of carboxylic groups, T. conoides exhibited high uptake capacity towards Al(III) and Cd(II) through ion-exchange mechanism. In the case of Al(III), T. conoides exhibited maximum biosorption at pH 4 with a capacity of 2.37?mmol/g, whereas the highest Cd(II) biosorption occurred at pH 5 with a capacity of 0.96?mmol/g. For both metal ions, T. conoides exhibited fast kinetics. Several models were used to describe isotherm (Langmuir, Freundlich, Redlich-Peterson, and Toth) and kinetic (pseudo-first and pseudo-second order) data. Desorption and reuse of T. conoides biomass in three repeated cycles was successful with 0.1?M HCl as elutant. In binary systems, the presence of Cd(II) severely affected Al(III) uptake by T. conoides. Compared to single-solute systems, Al(III) uptake was reduced to 56% compared to only 27% for Cd(II). Based on the model parameters regressed from the respective monometal systems, multicomponent Langmuir and Freundlich models were used to predict binary (Al + Cd) system of which the multicomponent Freundlich model was able to describe with good accuracy.  相似文献

A field experiment was conducted to evaluate the effect of integrated use of farmyard manure and bio-inoculants on wheat productivity for two years in succession. Increasing levels of farmyard manure (FYM) up to 15 t ha?1 significantly (p ≤ 0.05) improved the dry matter accumulation, effective tillers per m row length, and grain weight per spike in both the years. Application of 15 t ha?1 FYM caused significant increase in spikelets per spike and grains per spike over control and 5 t ha?1 during two consecutive years. Inoculation with MSX-9 strain of Azotobacter chroococcum produced significantly higher dry matter accumulation to 25.63, 13.33, 7.78 and 23.66, 8.35, 5.50% over uninoculation, Azospirillum brasilense (SP-7) and Azospirillum lipoferum (A-5) at harvest during 1999–2000 and 2000–2001, respectively. Incorporation of 15 t ha?1 FYM significantly (p ≤ 0.05) enhanced grain and straw yield to 62.45 and 38.05%; 56.66 and 36.28%; 59.42 and 37.52% over control in 1999–2000, 2000–2001 and pooled analysis, respectively. The grain and straw yield of wheat significantly (p ≤ 0.05) enhanced to 26.51, 10.10, 7.54 and 14.45, 5.77, 3.16% through A. chroococcum (MSX-9), A. brasilense (SP-7) and A. lipoferum (A-5) over uninoculation.  相似文献

A sequence characterized amplified region (SCAR) marker tagged to an Agropyron elongatum‐derived leaf rust resistance (Lr) gene Lr19 was validated on 18 known alien Lr gener in near‐isogenic lines (NILs) in the variety ‘Thatcher’, along with three wheat cultivers carrying Lr24 and two carrying Lr19. The marker was expressed only in the Lr24 lines confirming that the marker tagged the geneLr24. The monomorphic expression of the SCAR marker in 10NIL pairs for Lr19 and Lr24 revealed that each NIL pair possessed the same gene, Lr24. The donor parents used in the NIL pairs for Lr19 (‘Sunstar*6/C80‐1′) and Lr24 (‘TR380‐14*7/3Ag#14′) amplified the same fragment. Nonsegregation for leaf rust in the F2 population of the cross between the above donor parents confirmed the presence of the same gene in the two parents. Apparently, a genuine parent stock of ‘Sunstar*6/C80‐1’ was not involved in the development of the NIL pairs for Lr19 due to an improper maintence bredding protocol either at source or destination which went undetected in the absence of signs of virulence for either gene in the region.  相似文献

An in vitro propagation method for female plants of Momordica dioica (Roxb.) has been established. The nodal segments were harvested and the cut ends of the explants were sealed with wax and then surface sterilized and cultured. Bud breaking occurred on Murashige and Skoog’s (MS) agar-gelled medium + 2.0 mg L−1 6-Benzylaminopurine (BAP) + 0.1 mg L−1 Indole-3 acetic acid (IAA). The cultures were amplified by passages on MS medium supplemented with 1.0 mg L−1 BAP + 0.1 mg L−1 IAA. Further, shoot amplification (29.2 shoots per vessel) was achieved by subculturing of in vitro regenerated shoot clump on MS medium + 0.5 mg L−1 BAP + 0.1 mg L−1 IAA. The micropropagated shoots were subsequently transferred for root formation on half-strength MS medium + 2.0 mg L−1 Indole-3 butyric acid (IBA) with 89% success rate. The in vitro-regenerated shoots were also rooted ex vitro with 34% success. These plantlets were hardened in the greenhouse and transferred to the field. The established protocol is suitable for true to type cloning of mature female plant of M. dioica.  相似文献

Reactions to eight isolates of Karnal bunt, Neovossia indica, collected from seven different locations in northern India were studied on 13 host lines, including cultivars and breeding lines of Triticum aestivum, Triticum durum and Triticosecale in all possible combinations. The incidence of Karnal bunt varied from zero in PBW 34 and PBW 248 with isolates Ni8 and Ni2, respectively, to as high as 66.8% in a highly susceptible cultivar WL 711 with isolate Ni5. The differences in disease incidence among cultivars and isolates were highly significant. All the isolates could be distinguished on the basis of differential reactions on one or more of the host lines. Even the most resistant lines of durum (PDW 215), triticale (TL 1210) and wheat (HD 29) could be distinguished by the differential disease reaction with one or more of the eight isolates. The cultivar-isolate interaction for disease score was highly significant, indicating the probable existence of a gene-for-gene relationship in this host-pathogen system.  相似文献

Introgression of the winter gene pool into spring wheat is being considered as one of the strategies to break through the yield plateau. However, little information is available on the combining ability of these two important but distinct groups of wheats in Indian conditions. Therefore, the present study was undertaken to determine the combining ability and gene action of yield and yield attributes in winter × spring wheat crosses. Seventy F1 progenies developed by 14 winter and five spring wheat lines using a line × tester design were evaluated, along with their parents, for yield and yield attributes in a randomized complete block design under field conditions. The mean squares for all the characters studied showed highly significant differences. The mean squares due to female × male interactions were significant for all the characters studied except for grains per ear and grain weight per ear. Additive genetic effects were found to play a key role in controlling the expression of days to heading, plant height and spikelets per ear.‘MV 19’ and ‘Stepniak’/‘Karvuna’ among winter and ‘PBW 65’ among spring wheats were good general combiners for most of the yield attributes studied. The estimates for specific combining ability effects suggested that, although general combining ability (GCA) effects of most winter wheats are either average or poor, their combination can give desirable genotypes with spring wheat parents possessing a high GCA.  相似文献

The effects of water stress on physiological attributes of drought‐sensitive (Kalyansona) and drought‐tolerant (C‐306) wheat cultivars were studied in a pot experiment. Water stress was imposed by withholding irrigation at boot and anthesis stages. Leaf water potential, leaf osmotic potential and leaf turgor potential (measured with pressure chamber and osmometer), as well as leaf diffusive resistance, leaf transpiration rate and leaf‐to‐air transpiration gradient (measured with a steady‐state porometer) were measured diurnally. Growth and yield parameters were recorded after harvesting of the crop. Triplicate data were analysed using a completely randomized design and correlations amongst these parameters were computed. Water stress was found to reduce diurnal leaf water potential and leaf osmotic potential in both the genotypes but leaf osmotic potential was significantly higher in the drought‐tolerant cultivar C‐306 than in the drought‐sensitive cultivar Kalyansona. Positive turgor was recorded in both the genotypes under water stress and non‐stress conditions. Water‐stressed plants showed significantly lower turgor potential than control plants. In diurnal observations, water‐stressed plants exhibited significantly higher leaf diffusive resistance in both genotypes at both stages. The diffusive resistance of C‐306 was predominantly higher than that of Kalyansona. Water stress decreased leaf transpiration rate at both stages but the reduction was higher at the anthesis stage. The leaf‐to‐air temperature gradient was much higher in C‐306 than in Kalyansona at the boot stage but at the anthesis stage genotypic variation was non‐significant. The capacity to maintain cooler foliage was lower at the anthesis stage than at the boot stage in both the cultivars. Shoot dry weight, number of grains, test weight, grain yield, biological yield and harvest index decreased to a greater extent when water stress was imposed at the anthesis stage, while imposition of water stress at the boot stage caused a greater reduction in plant height and number of tillers. Similarly, water stress caused a smaller reduction in growth, yield and yield attributes in C‐306 than in Kalyansona. In general, the correlation coefficient of grain and biological yield with water potential and its components was positive and highly significant. Similarly, turgor potential was also correlated positively and significantly with grain yield at both the stages, but with biological yield it was significant only at the anthesis stage. A negative and significant correlation was obtained for diffusive resistance and leaf‐to‐air temperature gradient with grain yield at the boot and anthesis stages. The rate of transpiration was also positively and significantly correlated to grain and biological yields at both the stages. Amongst the yield attributes, number of leaves and number of tillers were positively correlated at the anthesis stage, whereas leaf area and shoot dry weight were significantly correlated with grain and biological yields at both the stages.  相似文献

Corn leaf aphids (Rhopalosiphum maidis Fitch) are found throughout the year on maize in Hawaii and occasionally cause yield loss. Sweet corn inbred Hi38-71 was observed to have high field resistance to aphids and was chosen for this genetic study. An artificial infestation technique was developed using hair-pin clip cages (2.2 cm diameter) which we devised and built. The cages were applied to field-grown plants into which three wingless viviparous adults were placed. Aphid populations were classified on a 1–10 rating scale after about 2 weeks based on digital images of the cages. Mean aphid coverage ratings were 2.97 for the resistant parent and 7.28 for the susceptible parent (representing >200 insects per cage). The F1 hybrids showed similar susceptibility (6.72), showing resistance to be recessive in nature. Six generations of the cross between Hi38-71 and susceptible inbred Hi27 were artificially infested to provide a generation mean analysis of 360 treated plants over two growing seasons. A joint scaling test showed that the fit to a 3-parameter additive-dominance model satisfactorily explained the observed variability with no assumed linkage or epistasis. We conclude that resistance to corn leaf aphid in Hi38-71 is conferred by a single recessive gene labeled aph. These results concur with a previous study under uncontrolled natural infestation in a single environment. The hair-pin clip cage method was most effective in distinguishing resistant and susceptible genotypes under diverse natural growing conditions in Hawaii.  相似文献

Wild Lens species/subspecies are a potential source for increasing genetic diversity in cultivated lentil. Four intraspecific crosses were attempted between cultivated and wild lentils. Viable hybrids were produced between L. culinaris ssp. culinaris × L. culinaris ssp. orientalis and L. culinaris ssp. culinaris × L. culinaris ssp. odomensis. Normal meiosis and pollen fertility were observed in the first set of crosses, whereas chromosomal abnormalities and reduced pollen fertility were observed in the second set of crosses. These crosses were also studied for some quantitative traits. The range, mean and coefficient of variation were calculated in parents, F1, F2 and BC1 generations to determine the extent of variability generated in the cultivated lentil through introgression of genes from wild lentil. The cultivated lentil × L. culinaris ssp. orientalis crosses showed substantially higher variability for all the traits than crosses involving cultivated lentil ×L. culinaris ssp. odomensis. The results of the present study indicated that these wild subspecies can be exploited for breeding purposes and their variation can easily be utilized to widen the genetic base of the cultivated lentil.  相似文献
