There is a growing interest in the links between humus forms and soil biota, and little is known about these links in Mediterranean
ecosystems. Culture-independent techniques, such as DNA extraction followed by DGGE and enzyme activities, allowed us to compare
microbial communities in two horizons of a forest soil in different seasonal conditions. Direct in situ lysis was applied
for extraction of DNA from soil; intracellular DNA was separated from extracellular and used to represent the composition
of microflora. The aims were to describe how biochemical and microbiological parameters correlate with topsoil properties
in typical Mediterranean Moder humus. Changes in bacterial and fungal community composition were evident from DGGE profiles.
Degrees of similarity and clustering correlation coefficients showed that the seasonal conditions may affect the composition
and activity of bacterial and fungal communities in the OH horizon, while in the E horizon the two communities were hardly
modified. In the same season, OH and E horizons showed a different composition of bacterial and fungal communities and different
enzyme activities, suggesting similar behaviour of eubacteria and fungi relatively to all the variables analysed. Evidently,
different organic carbon content in soil horizons influenced microflora composition and microbial activities involved in the

The polyphenol profile of an interspecific allotetraploid somatic hybrid, achieved by protoplast fusion, the 'Valencia' sweet orange ( Citrus sinensis L. Osbeck) + 'Femminello' lemon ( Citrus limon L. Burm), and three sexual hybrids obtained by backcrosses between Femminello lemon and the allotetraploid somatic hybrid ('Valencia' + 'Femminello') was studied by liquid chromatography-ultraviolet-diode array detector-mass spectrometry (LC-UV-DAD-MS). The aim of the work was to evaluate whether superior traits and improved performance can be observed in these new genotypes. Ten flavonoids (TF), comprising seven flavanones and three flavones, and four hydroxycinnamic acids (HCA) have been characterized and quantified in all hybrids and compared with those of the respective parents. The 'Valencia' + 'Femminello' somatic hybrid shows an intermediate polyphenol composition with respect to those of the parents, with a slight prevalence of lemon influence. The three sexual hybrids show, instead, different and more complex chromatographic profiles. 相似文献

In this paper molecular analysis, physical, technological, and chemical traits were used to estimate the level of variation
on five accessions of a locally adapted Sicilian faba bean (Vicia faba L.) landrace named ‘Larga di Leonforte’. DNA analysis was performed using the Amplified Fragment Length Polymorphism (AFLP)
molecular marker class and two other faba beans (cv. ‘Aguadulce Samba’—Spain—and landrace ‘Locale di Modica’—Sicily) were
used as controls. Although the accessions of ‘Larga di Leonforte’ varied significantly for most of the agronomical and physical
traits, this landrace generally had a heavy seed weight, short but large pods, and no more than two seeds per pod. This last
characteristic allowed for erect pod angle attitude at maturity. Idratation data showed difference among accessions in seed
weight at full hydration and in absorption rate at the very beginning of the hydration process, while any difference among
accessions emerged in terms of cooking properties. The six AFLP Eco+3/Mse+3 different primer combinations applied in this
research revealed different levels of polymorphism among the faba bean accessions and a total number of 346 amplicons were
generated. Around 60% of amplicons displayed a polymorphic pattern among different accessions. Cluster analysis on morphological,
technological, chemical and molecular data placed the all five ‘Larga di Leonforte’ accessions into a separated group, and
the Sicilian material shares a fairly large amount of similarity with respect to the cultivar ‘Aguadulce Samba’ selected in

Favourable prices and new technologies have recently stimulated the adoption of wheat–soybean double crops in the south-eastern Pampas. To assess the largely unknown long-term performance of this strategy, we used CERES-Wheat and CROPGRO-Soybean with slight adaptations to account for double cropping. Models and climate records were combined to analyse management practices allowing for earlier soybean sowing, which was assumed to be the key for high soybean yield and whole-system performance. Practices investigated were: (i) short-season wheat cultivars; (ii) non-selective herbicides for faster drying of wheat before harvest; (iii) relay double cropping, where soybean is sown on standing wheat before crop maturity in contrast to sequential cropping where soybean is sown after wheat harvest. 相似文献

Increasing grain yield is a key breeding goal in bread wheat. Several authors have suggested that a spike fertility index (SF), that is the quotient between grain number per unit spike (GNS) and spike chaff dry weight (SCDW), could be used as a yield‐related selection criterion, especially if molecular markers were available. Here, the effects of Ppd‐B1 and Ppd‐D1 genes on SFm, GNSm and SCDWm (measured at maturity) and the relationship between these variables were analysed in field experiments carried out during three crop seasons at Balcarce, Argentina, on an association mapping population of 100 bread wheat cultivars of diverse origin released in Argentina between 1927 and 2010. Results show that both Ppd‐B1 and Ppd‐D1 are associated with SFm with similar effects. Cultivars with insensitive alleles at both genes showed a mean SFm 9.2% greater than those with sensitive alleles at both genes; at each gene, difference in SFm between insensitive and sensitive alleles was ~4.5%. In turn, each gene showed a differential effect on GNSm and SCDWm, as Ppd‐B1 was more related to SCDWm, whereas Ppd‐D1 was only related to GNSm. Although more research needs to be carried out in order to ascertain the physiological pathway by which these genes affect spike fertility, this study represents a first approximation in order to elucidate the molecular and genetic basis underlying SF and related physiological traits. 相似文献

Five clones of sour orange (Citrus aurantium L.) showing significant morphological differences were selected from our germplasm collection and characterized both by genetic and leaf volatiles analysis. The genetic studies were undertaken by the use of molecular markers developed by PCR-based techniques (ISSR and RAPD), while the leaf essential oil patterns were obtained by chromatographic and mass spectrometric determination. Data obtained suggest that reasonably similar information can be achieved from the two techniques, supporting each other in characterization studies. 相似文献

The ultrastructural modifications that occur on the ependymal surface of the lateral ventricles of prenatal rabbit brains at 25, 27, 29 days of pregnancy, of term fetuses (30–31 days) and of 5, 10, 15 day old newborn rabbits, were studied by a SEM. On the ependymal surface the cilia, usually associated in tufts and the microvilli, variable in size and number, that surround them, show a rate of development correlated to the age. The results show the early differentiation of the surface features commensurated with early cerebrospinal fluid (CSF) functioning. 相似文献

This study evaluated the effects of rearing density on muscle growth and development in Siberian sturgeon (Acipenser baerii) larvae. Three different stocking densities were tested: low (LD, 30 larvae/l), mid (MD, 80 larvae/l) and high (HD, 150 larvae/l) in a recirculating aquaculture system. Larvae were sampled at hatching (T0), schooling (T1) and complete yolk‐sac absorption (T2) stage and were weighed and processed for muscle tissue histometrical analyses and for qualitative morphological study analyses; fatty acid profile was also determined by Gas Chromatography—Flame Ionization Detector analysis. Low‐density larvae presented a higher weight than MD or HD at T2 (p < 0.05). Histometrical analysis revealed that total muscle area was similar at T1 and T2, but higher than T0, while it was lower at HD at schooling (p < 0.05). The fatty acid profile revealed no differences between densities while, during development, there was a selective consumption: sparing or increasing of essential fatty acids to the detriment of their precursors. Our study suggests that lower densities appear to be more suitable to rear Siberian sturgeon in this particular stage of development. Indeed, larvae reared at the lower density were heavier and longer while larvae reared at the higher density showed lower muscle proliferation rate. As a consequence, LD larvae may exert an increase of potential growth at a mid‐long term. 相似文献
