The overall accuracy of an enzyme-linked immunosorbent assay (ELISA) used to detect Johne's disease at herd level was explored in relation to an imperfect test (fecal culture) in 57 Israeli dairy herds. Receiver-operating characteristic (ROC) analysis indicated an area under the curve (AUC) that corresponded to a test accuracy of 82.0% (69.5% to 90.9%; 95% confidence), with optimized herd sensitivity and herd specificity of 70.4% and 83.3%, respectively; and predictive values of 79.2 (+) and 75.8% (-). The optimal ELISA cutoff was 3.16% (> 3.16% seropositive cows in a herd), which was associated with likelihood ratios (LR) of 4.22 (+LR) and 0.36 (-LR), and post-test probabilities of 0.79 (+) and 0.17 (-). For herds with < or = 200 cows (n = 19 herds), the 95% confidence interval (CI) for the AUC was 0.62-0.97 and the optimal cutoff was 3.33% (HSe = 87.5, HSp = 81.8); for herds with > 200 but < or = 270 cows (n = 19 herds), the 95% AUC CI was 0.62-0.97 and the optimal cutoff was 1.13% (HSe = 90.0, HSp = 77.78); and for herds with > 270 cows (n = 19 herds), the 95% AUC CI was 0.69-0.99 and the optimal cutoff was 0.7% (HSe = 100.0, HSp = 70.0). The AUC was not influenced by across-herd prevalence [R2 (adjusted) = 0.0, P > 0.05]. Findings may be applied to facilitate targeted sampling of herds similar to those evaluated. For instance, a test cutoff of 0.76% could be considered for "ruling disease in," while a cutoff of 3.7% could be used for "ruling disease out." Caveats that may influence this analysis are discussed.  相似文献

The present work aims to investigate if the variation of each parameter in Longissimus lumborum muscle could correspond to the same or to a similar variation of the parameter in the other muscles. The work presents results of Pearson's correlations between Longissimus lumborum samples and other muscle samples, such as Biceps femoris, Rectus femoris, Semimembranosus, Supraspinatus and Semitendinosus in horse meat. A total of 27 male IHDH (Italian Heavy Draught Horse) breed foals were employed. They were slaughtered at 11 months of age and the above‐mentioned muscles were sampled. The Longissimus lumborum muscle showed to be representative of other muscles and of the whole carcass for some chemical parameters (moisture, protein and ash) and for some fatty acids profile patterns such as C12:0, C14:0, total monounsaturated fatty acid and polyunsaturated fatty acid, but poor correlations were recorded for intramuscular fat concentration, rheological and colorimetric parameters. Although almost all the qualitative parameters in meat are affected by the anatomical site and by the muscle, the Longissimus lumborum is often not representative in horse meat with regard to modifications of this parameters.  相似文献

It has been reported for many soils that maize (Zea mays L.) has a higher soil‐P critical level than soybean (Glycine max L.) and sunflower (Helianthus annuus L). The objective of this work was to compare the rhizosphere P depletion in these three species in order to investigate if they differ in their capacity to acquire soil P. Sequential P fractionation and pH were determined in rhizosphere and nonrhizosphere soil samples from field and greenhouse experiments. Neither sunflower (the species with highest rhizosphere acidification) nor soybean or maize showed a significant relationship between P depletion and rhizosphere pH. The labile P fraction and the NaOH‐Pi fraction had lower values in the rhizosphere than in the bulk soil in 38% and 77% of the studied cases, respectively. Sunflower and especially maize presented a more intense Pi depletion than soybean. The comparison between sunflower and maize revealed that neither of them took a clear advantage over the other in terms of P depletion. Rhizosphere Pi depletion was associated with the amount of P acquired by the plants. We conclude that the accessibility to different P pools does not explain the differences in soil‐P critical levels among the three species.  相似文献

A method has been developed for the extraction of capsaicinoids from peppers by pressurized liquid extraction (PLE); these compounds are determined by reverse phase high-performance liquid chromatography (HPLC), with detection by fluorescence spectrophotometry and mass spectrometry (MS). The stability of capsaicin and dihydrocapsaicin has been studied at different temperatures (50-200 degrees C), and several extraction variables have been assayed: solvent (methanol, ethanol, and water), different percentages of water in the methanol (0-20%) and in the ethanol (0-20%), and the number of extraction cycles. The study has evaluated the repeatability (RSD < 7%) and the reproducibility (RSD < 7%) of the method. Finally, the PLE method developed has been applied to quantify the capsaicinoids present in three varieties of hot peppers cultivated in Spain, quantifying five capsaicinoids: nordihydrocapsaicin, capsaicin, dihydrocapsaicin, an isomer of dihydrocapsaicin, and homodihydrocapsaicin.  相似文献

Summary The species Lupinus hispanicus, throught its use as self-seeding forage, can contribute to solve the problem of forage scarcity during the summer in the livestock farming of the central-west Iberian Peninsula. A feature of the highest importance in achieving self-seeding is the seed-coat permeability. An evaluation of this characteristic has been carried out in a Lupinus hispanicus collection from Salamanca (Spain), its inheritance, and the influence of daily temperature fluctuations on the softening of the seeds. All the lines but one had impermeable seeds. It was established that the permeability of the seeds is controlled by the genotype of the mother-plant and the inheritance of this characteristic follows a simple Mendelian model, the permeability being recessive. It was also observed that seeds subjected to daily temperature fluctuations had an increased level of permeability.  相似文献

We analyze the impact of grazing on dung beetle diversity at the Barranca de Metztitlán Biosphere Reserve, a xeric ecosystem in central Mexico with a long history of use by humans. We compared the community structure, as well as the alpha and beta diversity between two cover conditions (open and closed vegetation) that represent the impact of grazing within a habitat, and between habitat types (submountainous and crassicaule scrublands). From 576 samples we collected 75,605 dung beetles belonging to 20 taxa. While mean species richness and diversity were different between habitat types, cumulative species richness was not. The effects of grazing on vegetation structure influenced the cumulative species richness and diversity of dung beetles in the submountainous scrubland, where grazing has created land mosaics of a grassland matrix with scrubland patches. This was not the case in the crassicaule scrubland where the impact of grazing is not as evident. Beta diversity significantly responds to the effects of grazing on habitat conditions. We discuss the ecological factors that may promote these responses by landscape diversity components. We also identify the species that could act as useful indicators to monitor the effect of land management on biodiversity. Our results indicate cattle farming maintains a diversified land mosaic, and these areas support more diverse dung beetle ensembles than homogeneous areas of closed, shrubby vegetation cover. Thus, controlled grazing activity could certainly favour the conservation of dung beetle biodiversity and improve ecosystem functioning by maintaining dung decomposition rates.  相似文献

We analyze the status of the Angel de la Guarda deer mouse (Peromyscus guardia), a species endemic to Mexico, based on our own fieldwork, bibliographic records and information from colleagues. This species, with different subspecies on three islands and a population of an undetermined subspecies on a fourth, was apparently common until the mid-1960s. Currently the species is critically endangered, if not already extinct. One of the subspecies and the undetermined population are presumed extinct. Of the two other subspecies, one is at least in critical condition, and the other at least reduced. The demise of the species can be attributed to the introduction of domestic cats. The case of P. guardia is a good example of the vulnerability of other taxa of endemic rodents on islands in the Gulf of California.  相似文献

Dehesa ecosystems are open woodlands with scattered oak trees as their main component. As a result of differing land-uses, the structure of vegetation found within dehesas varies between: (i) oak trees and intercropped cereals (cropped), (ii) oak trees and native grass vegetation (grazed), and (iii) oak trees with abundant understorey shrubs (encroached). The aim of this study is to investigate whether land-use influences the water dynamics of dehesas by measuring available soil water content (AWC) in the upper 250 cm of the soil at different distances from tree trunks (maximum 30 m) at four Quercus ilex dehesas in Central–Western Spain. The technique used was Time Domain Reflectometry and the study was undertaken between May of 2002 and December of 2005. Leaf water potential (Ψ) was also measured on trees at one site by mean of a pressure chamber. Within the upper meter of the soil, it appears that trees, grasses and shrubs extracted soil water resources in a similar way from both beneath and beyond the tree canopy. However, encroached plots in general showed lower average AWC values than cropped or grazed plots (3.7, 5.6, and 6.2% in encroached, cropped and grazed, respectively). Cereal crops do not compete more strongly than grasses with trees for available soil water resources. The similar Ψ values found at cropped and grazed plots supported these results. From our results, it could be hypothesized that ploughed dehesas could facilitate soil re-watering in the plots with pronounced slopes. The decrease of AWC values at encroached plots with respect to the cropped and grazed plots was found mostly beyond the tree trunk at deeper soil layers, indicating that shrubs use water partly not accessible to trees. The presence of an understory of shrubs seems to have slightly increased the water constraints on trees during the summer period (Ψd values of − 0.5, − 0.5, and − 0.8 MPa in cropped, grazed, and encroached plots, respectively). In cropped and grazed plots, an important amount of water seems to have remained unused for trees and grasses.  相似文献

Demographic changes introduced by migration strongly affect economic activities and may thus trigger land-use changes. Migration has been usually overlooked in land use change modelling, even though it is recognized as a dominant demographic factor that influences land use. This paper analyzes to what extent migration patterns influence land-cover and land-use change at the watershed level. A RS-GIS and statistical approach was used to quantify and analyze both land-cover change and change in population per spatial unit. It was hypothesized that migration should exert a strong effect on land-cover change. The exercise was carried out in the Basin of Lake Cuitzeo, Michoacan, in Central Mexico, an area of high emigration to the USA, albeit showing a net increase in its total population. The expansion of scrubland areas at the expense of rain-fed agricultural land is associated with the abandonment of agricultural land with poor soils. As a consequence, vegetal succession has been promoted and subtropical scrubland increased.  相似文献

The present experiments on goats were conducted with the aim of verifying (a) whether the identity of the goat kids can affect the characteristics of milk let-down during suckling (Study 1) and (b) whether the presence of the goat kids may improve the milk let-down during hand milking (Study 2). In Study 1, 16 lactating goats with twins and 12 with single kids were used. Dams with single goat kids ejected more milk when suckling their kids at Days 5 and 20 of lactation, than when suckling aliens. Dams with twins ejected more milk when suckling their own kids than aliens only at Day 5 of lactation. Taking into account all the dams, they ejected more milk with more fat, protein, and lactose during suckling their own litter than when suckling aliens. In Study 2, when dams (n = 13) were hand milked in the presence of their own litter or in its absence, they ejected more milk with more fat, protein, and lactose than when milked in the presence of alien kids. We conclude that milk let-down in goats can be influenced by the type of stimuli applied or to which they are exposed to during suckling and milking.  相似文献
