Elevations in cytosolic free calcium concentration ([Ca(2+)](cyt)) constitute a fundamental signal transduction mechanism in eukaryotic cells, but the molecular identity of Ca(2+) channels initiating this signal in plants is still under debate. Here, we show by pharmacology and loss-of-function mutants that in tobacco and Arabidopsis, glutamate receptor-like channels (GLRs) facilitate Ca(2+) influx across the plasma membrane, modulate apical [Ca(2+)](cyt) gradient, and consequently affect pollen tube growth and morphogenesis. Additionally, wild-type pollen tubes grown in pistils of knock-out mutants for serine-racemase (SR1) displayed growth defects consistent with a decrease in GLR activity. Our findings reveal a novel plant signaling mechanism between male gametophyte and pistil tissue similar to amino acid-mediated communication commonly observed in animal nervous systems.  相似文献

  1. The conservation of marine megafauna presents numerous difficulties owing to their high mobility over difficult-to-access oceanic areas that impairs the collection of basic, but essential, biological information.
  2. The Bryde's whale (Balaenoptera edeni) is one of the most elusive species of baleen whales, and although it is known to be a seasonal visitor to several archipelagos in Macaronesia (the Azores, Madeira, and Canaries), there are no studies regarding its occurrence or geographical connectivity in this area of the Atlantic.

A 14-year photographic database was used to determine short-term (intra-seasonal) and long-term (inter-annual) Bryde's whale site fidelity and to estimate individual residency times in Madeira, whereas photographic catalogues from Madeira and the Canaries were compared in order to assess large-scale movements (i.e. on the scale of hundreds of kilometres).
  4. In Madeira, 59 individuals were identified, 27 (45.8%) of which were recaptured. Of these, 10 individuals (37.0%) presented short-term site fidelity and 17 individuals (63.0%) presented long-term site fidelity, with a maximum recapture interval of 12 years. Lagged identification rates showed that five individuals (SE = 2) remained in the area for 32 days (SE = 108 days) before leaving and not returning during the same year. Seven individuals were seen both in Madeira and the Canaries (catalogue comprising 51 individuals), three of which were identified multiple times in both archipelagos, with a minimum of 43 days between consecutive sightings.

This information combined with the fact that this species is commonly sighted accompanied by calves and feeding in both archipelagos highlights the ecological importance of this area for Bryde's whales. This should be taken into consideration by policymakers when implementing conservation measures, where coordination of effort among countries is needed. This study also reinforces the value of using data from platforms of opportunity and of making photographic data open access.

Parasitic infection is recognized worldwide as a limiting factor in the production of goats, and various control methods are used to reduce economic losses, often without considering the epidemiology of the parasites. This has led to the development of highly tolerant parasite populations and the presence of chemical residues in the beef and milk. The objective of this study was to determine the level of knowledge of goat farmers about parasitic diseases and to correlate this with the epidemiology of endoparasites and parasite control practices in goat farms in the state of Minas Gerais, Brazil. The analysis was based on a questionnaire applied by trained veterinarians. The sample was homogeneous throughout the state, covering 18.4% (157/853) of municipalities. Eighty-four dairy goat farms in 81 municipalities and 200 properties with beef goats in 76 municipalities were evaluated. The herd size per goat farm ranged from 4 to 57 (average 24) for beef herds and from 2 to 308 (average 63) for dairy farms. The majority of the beef herd production was extensive and semi-extensive (98.5%), while the dairy herds were maintained under intensive farming (98.8%). The mixed production of goats and sheep was reported by 36.5% of beef goat farmers and by 20.2% of dairy goat farmers. Among the beef goats farms on which the technological level was determined, 2.0% were categorized as having high technological level, 34.5% as medium, and 63.5% as low. Of the 84 dairy farms, 30% operated at a high, 47% at a medium, and 23% at a low technological level. The adoption of practices to reduce parasitism, such as the quarantine of animals, treatment of newly arrived animals, regular cleaning of the floor, and technical assistance, was significantly higher on dairy farms than on beef farms. Although 85.7% of dairy farmers and 83% of beef farmers medicate their animals, the treatments were performed without technical criteria, and deworming intervals ranged from 30 to 120 days or more. The average interval between treatments was significantly longer in dairy goat herds (4.8 months) than in the beef herds (3.6 months). The most commonly used drugs were macrocyclic lactones (37.7% in dairy and 39.5% in beef herds) and benzimidazoles (48.9% in dairy and 31.5% in beef herds). Goat production in Minas Gerais is still in its infancy, and even though using a control program associated with other health practices, producers still rely heavily on chemicals to get satisfactory results.  相似文献

Changing climate is not the only driver for alterations in the dynamic interaction between arthropod vectors of zoonotic parasites and their hosts, including humans. A suite of other factors ranging from urbanization and deforestation to changing demographics in both developing and developed countries, the impact of the recent economic crisis, increased global movement of people and animals and follow-on effects of major catastrophes. This article reviews the most important vector-borne parasites of zoonotic concern that are changing/expanding their distribution patterns in both endemic and/or previously non-endemic areas. We include the discussion of the changing aspects of malaria, leishmaniasis, babesiosis, Chagas disease as well as of some spirurid and filarioid nematodes.  相似文献

Longitudinal studies evaluating the evolution of clinical, haematological, biochemical findings in young dogs exposed for the first time to multiple vector-borne pathogens have not been reported. With the objective of assessing the evolution of clinical, haematological and biochemical findings, these parameters were serially monitored in naturally infected dogs throughout a 1-year follow-up period. Young dogs, infected by vector-borne pathogens based on cytology or polymerase chain reaction, were examined clinically and blood samples were obtained at seven different follow-up time points. Dogs were randomized to group A (17 dogs treated with a spot-on formulation of imidacloprid 10% and permethrin 50%) or to group B (17 dogs untreated). In addition, 10 4-month-old beagles were enrolled in each group and used as sentinel dogs. At baseline, Anaplasma platys was the most frequently detected pathogen, followed by Babesia vogeli, Bartonella spp., Ehrlichia canis and Hepatozoon canis. Co-infections with A. platys and B. vogeli, followed by E. canis and B. vogeli, A. platys and H. canis and A. platys and Bartonella spp. were also diagnosed. In dogs from group B, abnormal clinical signs were recorded at different time points throughout the study. No abnormal clinical signs were recorded in group A dogs. Thrombocytopenia was the most frequent haematological alteration recorded in A. platys-infected dogs, B. vogeli-infected dogs and in dogs co-infected with A. platys and B. vogeli or A. platys and Bartonella spp. Lymphocytosis was frequently detected among dogs infected with B. vogeli or co-infected with A. platys and B. vogeli. Beagles were often infected with a single pathogen rather than with multiple canine vector-borne pathogens. There was a significant association (p<0.01) between tick infestation and A. platys or B. vogeli, as single infections, and A. platys and B. vogeli or A. platys and Bartonella spp. co-infections. This study emphasizes the clinical difficulties associated with assigning a specific clinical sign or haematological abnormality to a particular canine vector-borne disease.  相似文献

Objective To determine the effects of topical 0.5% tropicamide on anterior segment morphology (ASM) and intraocular pressure (IOP) in normal and glaucomatous cats. Animals used Normal cats and cats with inherited primary congenital glaucoma (PCG). Procedures Control IOP curves were performed in untreated normal and PCG cats. In the first experiment, tropicamide was applied OD in eight normal and nine PCG cats. IOP and pupillary diameter (PD) were measured at 0, 30, and 60 min, then hourly until 8 h post‐treatment. In a second experiment, six normal and seven PCG cats received tropicamide OD. High‐resolution ultrasound images were obtained at 0, 1, 5, and 10 h post‐treatment to measure ASM changes. IOP and PD were measured OD at 0, 1, 2, 3, 5, 7, and 9 h. Results In untreated normal cats IOP OU decreased throughout the day. In PCG cats IOP OU had wide fluctuations over time. In normal cats IOP response varied in the treated eye but did not change significantly in untreated eyes. IOP significantly increased from baseline in both eyes of all treated PCG cats. Increases in IOP were associated with some ASM changes. Cats with PCG had a significantly smaller angle recess areas, diminished ciliary clefts and decreased iris‐lens contact. ASM changes were not strongly correlated with IOP in all cats. Conclusions The ASM of PCG cats is markedly different from normal cats, and clinically significant increases in IOP OU occur in cats with PCG after tropicamide treatment. The mechanism for this increase remains unclear.  相似文献

The aim of this study was to validate an A/T single nucleotide polymorphism (SNP) corresponding to a LINE2 sequence located ~1.1kb downstream of the IL-6 gene (SNP BIEC2-911738) and to determine if this variant is correlated with interleukin 6 (IL-6) modulation or with different plasma concentrations of Zn, Cu, Se and Fe. The frequency of the newly described variant T ranged from 0 to 23.1% among different breeds of horses. SBIEC2-911738 was not associated with changes in IL-6 plasma levels. Increased Cu:Zn ratios were observed in horses carrying the AT genotype independently of breed when stabled for 24h after physical exercise.  相似文献
