Rice blast (Pyricularia oryzae) has become a serious disease on commercial rice (Oryza sativa) crops in northern Australia and is present there on wild rice (O. australiensis). Characterisation of the host range of P. oryzae is fundamental to both reducing disease spread and to preventing development of epidemics via better management of Poaceae inoculum reservoirs in Australia. Studies on response of three different wild O. australiensis sources toward four isolates of P. oryzae showed all genotypes very susceptible to three isolates [WAC13466 (race IA-1), BRIP53376 (race IB-3), NT2014a (race unknown)], but resistant to isolate BRIP39772 (race IA-3). Studies to investigate levels of blast disease development following inoculation on a range of Poaceae hosts showed both P. oryzae isolates (WAC13466, BRIP53376) were highly virulent on barley (disease index, DI?=?100%), and on Phalaris and O. australiensis (DI?=?70%). However, isolate BRIP53376 showed a significantly higher level of aggressiveness (DI ~80%) on ryegrass, wild oat and rice. Of the two wheat cultivars tested, only one cultivar showed disease and only from WAC13466 (DI ~30%). Sweet corn and goosegrass were also susceptible to both blast isolates, but DI was <50%. That P. oryzae was virulent across these diverse Poaceae hosts, highlights, for Australia, the possibility for these species in, first, harbouring P. oryzae isolates highly virulent to commercial rice, second, fostering spread of rice-attacking P. oryzae strains into regions currently free of rice blast, and third, potential for these alternative host species to encourage development of more virulent host-specific strains of P. oryzae. The current study is an important step towards facilitating improved crop protection in the medium and long term from reducing P. oryzae disease epidemics via a better understanding and management of inoculum reservoirs in Australia.

The clinical utility of various specimens was examined for the early diagnosis of canine distemper (CD). Seven healthy dogs at 17 weeks of age were experimentally infected with a field isolate of canine distemper virus. The RT-PCR was carried out to detect CDV NP gene. Dogs showed mild fever and leukopenia, however, typical clinical signs of CD were not seen through the experimental period. CDV amplicons were detected more, earlier and for longer period in the conjunctival swabs than in the other samples employed. These results suggested that conjunctival swab samples, which are easy to obtain and non-invasive, would be the most suitable and practical specimen for the early antemortem diagnosis of CDV infection.  相似文献

Microvascularization of domestic fowl kidneys was studied using scanning electron microscopy (SEM) of vascular corrosion casts (VCCs). Two types of nephrons, mammalian-type (MT) and reptilian-type (RT) nephrons and their glomerular structure were analysed quantitatively by 3D morphometry. A significant difference in shape and size between the MT and RT glomeruli was found. The mean diameter of the RT glomeruli was about 56 µm, while that of MT glomeruli was significantly larger, namely about 80 µm. The afferent arterioles in mammalian-type glomeruli usually bifurcated into two lobular branches and formed a complex glomerular capillary network with numerous loops. Reptilian-type glomeruli consisted of a single capillary forming few loops and leaving the glomerulus as efferent arteriole. Diameters of afferent and efferent arteriolar replicas were similar in all three kidney divisions of MT and RT nephrons. The absence of the interconnecting branches between the MT nephron capillaries at the gross inspection suggests that the mammalian-type nephron glomeruli, although more complex than the reptilian type, are not equivalent to those in mammalian kidneys.  相似文献

The type virus of the family Hypoviridae, Cryphonectria hypovirus 1 strain EP713 (CHV1-EP713), infects Cryphonectria parasitica, the filamentous causal fungus of chestnut blight, and reduces its virulence. This pathosystem serves as a model to study fungus-mycovirus interactions. We previously developed a genetic screening protocol for host factors associated with symptom induction by CHV1-EP713 and its mutants. In the procedure the standard field fungal isolate EP155 was transformed by cDNA from a mild hypovirus mutant Cys(72), launching virus infection, and mutagenized by random plasmid insertion with pHygR conferring hygromycin resistance. We now report an extension of the study to characterize different mutant strains, with different phenotypes than their parental strain TCys(72)-1. TCys(72)-1 is moderately reduced in pigmentation and sporulation compared to the uninfected wild-type strain EP155. Mutants sfb1, sfb2 and k202 were characterized biologically and molecularly in comparison to the previously isolated mutant (namA) and the parental strain. These mutants harbored one (sfb1) or more copies (sfb2 and k202) of the mutagenic plasmid, pHygR. The three mutants had similar biological attributes; that is, vegetative growth rate, conidiation and virulence (assay on apples) was reduced on potato dextrose agar media, relative to the parental strain and pigmentation was the same or slightly increased. Interestingly, viral dsRNA accumulation levels were apparently unaltered in these mutants. The screening method was efficient for mining fungal mutants with unusual hypovirus symptoms. Further, characterization of the mutants provides interesting insights into symptom induction by the hypovirus.  相似文献

The experimental results of orthogonal cutting of maple and the modeling of the cutting mechanics are presented. The tool cutting forces were measured for different feed rates. A set of equations relating the tangential and feed forces to the tool edge width and feed rate (chip thickness) to calculate the chip and edge cutting force coefficients was developed. Then the chip force and edge force coefficients were calculated from experimentally obtained cutting forces and were plotted in a polar-coordinate system with respect to the fiber orientation of the maple disk. The polar-coordinate presentation of the cutting force results and the calculated cutting force coefficients provides an excellent visual appreciation of the relation between the cutting forces and the wood fiber orientation. Chips were also collected from various sectors of the wood disk. This analysis further identified the effects of fiber orientation and cutting forces on the types of chip formed and hence the cutting mechanics involved. By applying the calculated cutting coefficients for each tool orientation (in respect to the grain) it is possible to predict the feed and tangential forces for any feed rates. There is good agreement between the predicted and measured cutting forces.  相似文献

Summary Two collecting missions were carried out in the Democratic People's Republic of Korea in 1986 within the frame of the agreement between the Academies of Sciences of the DPRK and the German Democratic Republic. The first mission was concentrated on genetic resources of the genusAllium. 23 samples have been collected. The second mission included cereals, grain legumes, vegetables and medicinal plants and resulted in 145 accessions. The areas covered are rich in land-races of cultivated plants and their wild relatives. Promising material for research and breeding purposes was collected.

Zusammenfassung Zwei Sammelreisen wurden 1986 in der Koreanischen Demokratischen Volksrepublik innerhalb des Abkommens zwischen den Akademien der Wissenschaften der KDVR und der Deutschen Demokratischen Republik durchgeführt. Die erste Reise konzentrierte sich auf die genetischen Ressourcen der GattungAllium. 23 Proben wurden gesammelt. Die zweite Reise schloß Getreide, Körnerleguminosen, Gemüse und Arzneipflanzen ein und erbrachte 145 Sammelnummern. Die untersuchten Gebiete sind reich an Landsorten von Kulturpflanzen und ihren wildwachsenden Verwandten. Es wurde aussichtsreiches Material für Forschungs-und Züchtungszwecke gesammelt.

This study was undertaken to determine the effects of dietary lipid levels on growth and nutrient utilization, and carbon (C), nitrogen (N) and phosphorus (P) balances in juvenile turbot (Psetta maxima) in brackish water. Four isonitrogenous diets (50 % protein) differing in dietary lipid levels from 10 to 19 by 3 % increments (named 10L, 13L, 16L and 19L, respectively) were fed to four replicate groups of fish with an initial weight of 54.4 ± 0.2 g for 9 weeks. Significantly better growth and feed conversion ratio were observed in fish fed 13L than those fed 16L and 19L. The whole body, fillet, viscera and liver dry matter and lipid levels were increased as dietary lipid was increased. Liver protein levels were lower in 16L and 19L than 10L. There was an ascending pattern in the whole-body C levels and C/N ratios with the increase in dietary lipids. No significant effect of dietary lipids on either protein efficiency ratio or N and P retentions was detected. Dietary lipids led to an increase in C losses to the environment. Overall, high dietary lipids when introduced at an adequate protein level did reduce growth and feed utilization without a protein-sparing effect and resulted in higher C losses in juvenile turbot reared, and a dietary lipid level of about 13 % seemed to be optimum.  相似文献
