A total of 251 Dura cross Pisifera (DxP) hybrid palms from six populations descending from six parental African Pisifera origins and involving 12 progenies were analyzed with 19 selected Simple Sequence Repeats (SSR) markers. A total of 110 alleles were produced, ranging from three to eight per SSR, with a mean of 5.8 alleles per SSR locus. Of these, 68.5% were considered shared alleles by more than one population and the remaining 31.5% were population specific alleles. They generated between six and 21 haplotypes in all populations, and depending on the SSR marker, between one and 10 haplotypes within populations. Various parameters for analyzing genetic variability, differentiation and genetic structure were computed using GenAlEx, Structure and Darwin software. The obtained results confirmed microsatellites as a robust, feasible and trustful method for obtaining DNA fingerprints, tracing the source of oil palm samples. With respect to the authenticity of materials or for solving legitimacy issues, accession belonging to each population by SSR markers could be distinguished, but additional SSR should be screened for improving this process. 相似文献

Schistosomiasis is a parasitic disease that affects more than 250 million people. The treatment is limited to praziquantel and the control of the intermediate host with the highly toxic molluscicidal niclosamide. Marine algae are a poorly explored and promising alternative that can provide lead compounds, and the use of multivariate analysis could contribute to quicker discovery. As part of our search for new natural compounds with which to control schistosomiasis, we screened 45 crude extracts obtained from 37 Brazilian seaweed species for their molluscicidal activity against Biomphalaria glabrata embryos and schistosomicidal activities against Schistosoma mansoni. Two sets of extracts were taxonomically grouped for metabolomic analysis. The extracts were analyzed by GC–MS, and the data were subjected to Pattern Hunter and Pearson correlation tests. Overall, 22 species (60%) showed activity in at least one of the two models. Multivariate analysis pointed towards 3 hits against B. glabrata veliger embryos in the Laurencia/Laurenciella set, 5 hits against B. glabrata blastula embryos, and 31 against S. mansoni in the Ochrophyta set. Preliminary annotations suggested some compounds such as triquinane alcohols, prenylated guaianes, dichotomanes, and xenianes. OBJECTIVE: To determine the amount of time required for surface temperatures of thoracic and pelvic limbs in horses to return to pre-exercise temperatures after high-speed treadmill exercise, as detected via infrared thermographic imaging. DESIGN: Prospective study. ANIMALS: 6 Thoroughbreds. PROCEDURES: All horses had been trained on and conditioned to use of a high-speed treadmill. Baseline thermographic images were obtained 3 days prior to exercise (baseline). Horses were exercised on a treadmill at a walk for 5 minutes, a slow trot (3 m/s) for 5 minutes, a trot (5 to 6 m/s) for 5 minutes, and a slow gallop (6 to 8 m/s) for 5 minutes, then back to a trot for 3 minutes, a slow trot for 3 minutes, and a walk for 3 minutes prior to stopping. Thermal images were obtained immediately after stopping exercise (0 minutes) and 5, 15, 45, and 60 minutes and 6 hours after stopping exercise. Ambient temperature surrounding each horse was recorded. RESULTS: In all regions, significant differences in surface temperatures were detected between thermograms obtained before exercise and those obtained immediately after, 5 minutes after, and 15 minutes after exercise was stopped. There were no significant differences in surface temperatures between thermograms obtained before exercise and those obtained > or = 45 minutes after exercise was stopped. CONCLUSIONS AND CLINICAL RELEVANCE: In horses, images generated via infrared thermography are not influenced by exercise-generated heat > or = 45 minutes after exercise is stopped. 相似文献

In this study, fecal Escherichia coli isolates (n=188) from 34 dog-owner pairs and 26 healthy control humans (2 isolates per individual) were tested for susceptibility to 6 antimicrobials and screened for virulence genes. Genetic diversity between canine and owner isolates was evaluated by pulsed-field gel electrophoresis (PFGE). Canine isolates exhibited significantly different rates of resistance to four and two antimicrobials, compared to control and owner isolates, respectively. Of the genes examined, the prevalence of sfa, hly, and cnf genes in canine isolates were higher than in control isolates, but not than in owner isolates. These results suggest that characteristics of owner isolates are somewhat similar to canine isolates, compared to isolates from non-dog owners. In addition, PFGE analysis revealed that transfer of E. coli between owners and their dogs had occurred within 3/34 (8.8%) households. Considering the effects of dog ownership on the population of E. coli isolates from owners, further epidemiological studies are required. 相似文献

In Uganda, a low-income country in east Africa, African swine fever (ASF) is endemic with yearly outbreaks. In the prevailing smallholder subsistence farming systems, farm biosecurity is largely non-existent. Outbreaks of ASF, particularly in smallholder farms, often go unreported, creating significant epidemiological knowledge gaps. The continuous circulation of ASF in smallholder settings also creates biosecurity challenges for larger farms. In this study, an on-going outbreak of ASF in an endemic area was investigated on farm level, including analyses of on-farm environmental virus contamination. The study was carried out on a medium-sized pig farm with 35 adult pigs and 103 piglets or growers at the onset of the outbreak. Within 3 months, all pigs had died or were slaughtered. The study included interviews with farm representatives as well as biological and environmental sampling. ASF was confirmed by the presence of ASF virus (ASFV) genomic material in biological (blood, serum) and environmental (soil, water, feed, manure) samples by real-time PCR. The ASFV-positive biological samples confirmed the clinical assessment and were consistent with known virus characteristics. Most environmental samples were found to be positive. Assessment of farm biosecurity, interviews, and the results from the biological and environmental samples revealed that breaches and non-compliance with biosecurity protocols most likely led to the introduction and within-farm spread of the virus. The information derived from this study provides valuable insight regarding the implementation of biosecurity measures, particularly in endemic areas. 相似文献

Genetic transformation systems require protocols that allow regenerating transgenic plants from transformed tissues. This study aimed to establish a protocol for indirect organogenesis in leaf explants of a Eucalyptus grandis × E. urophylla AEC 224 clone. During callogenesis stage, several concentrations of NAA and then NAA or 2,4-D combined with TDZ were tested in JADS culture medium for 30 days, followed by subculture of the explants in the regeneration medium, containing 5.0 µM BA and 0.5 µM NAA for another 30 days. In these media, the explant oxidation rate was high (95 %). Thus, in order to reduce oxidation, different culture media were compared: WPM, MS, JADS and modified QL, followed by explant transfer onto regeneration medium. The highest percentage of regeneration and the lowest oxidation rate were achieved on WPM medium. Then, NAA and 2,4-D were tested in combination with TDZ and also TDZ and BA combined with NAA in WPM medium. The most efficient culture media in terms of shoot regeneration were WPM supplemented with 0.25 µM TDZ and 0.1 µM NAA during 30 days for callus induction and then with 5.0 µM BA and 0.5 µM NAA for another 30 days. This protocol yielded a regeneration rate of 43 %, with a low oxidation of tissues. A rooting experiment was conducted using half strength MS medium and comparing three concentrations of IBA (2.46, 4.90 and 7.35 µM). The highest rooting percentage (35 %) was obtained on medium containing 2.46 µM IBA. Once the shoots were rooted, acclimatization in a greenhouse was not challenging and plant survival reached 100 %. 相似文献

Cereal production is chronically deficit in the Timbuktu region of Mali, sufficient for only 4.5 months of annual household
consumption. Small-scale, village-based irrigation schemes, usually 30–35 ha in size, irrigated by a diesel motor pump, have
become important to improve food security in this arid region. The NGO Africare has worked during the past 12 years with farmers
in Goundam and Dire circles to establish irrigation schemes and provide them with technical assistance. In 2007, Africare
undertook a first test of the System of Rice Intensification (SRI) in Goundam circle. After farmers observed a yield of 9 t ha−1 of paddy compared to 6.7 t ha−1 in the control plot there was interest in larger scale testing of the SRI system. In 2008, Africare, in collaboration with
the local Government Agriculture Service and with support from the Better U Foundation, implemented a community-based evaluation
of SRI with 60 farmers in 12 villages. Farmers in each village selected five volunteers, who each installed both SRI and control
plots, side by side, starting the nurseries on the same day and using the same seed. For SRI plots, seedlings were transplanted
one plant hill−1 at the two-leaf stage (on average, 11.6 days old), with spacing of 25 cm × 25 cm between hills and aligned in both directions.
This allowed farmers to cross-weed with a cono-weeder, on average 2.4 times during the season. In the control plots, farmers
planted 3 plants hill−1 with seedlings 29.4 days old and spaced on average 23.7 cm, not planted in lines. Weeding was done by hand. 13 t ha−1 of organic matter was applied under SRI management, and 3 t ha−1 in the control plots. Fertilizer use was reduced by 30% with SRI compared to the control. Although alternate wetting and
drying irrigation is recommended for SRI, this was not optimally implemented due to constraints on irrigation management within
the scheme; thus water savings were only 10% compared to the control. Average SRI yield for all farmers reached 9.1 t ha−1, with the lowest being 5.4 t ha−1 and highest being 12.4 t ha−1. SRI yields were on average 66% higher than the control plots at 5.5 t ha−1, and 87% higher than the yields in surrounding rice fields at 4.9 t ha−1. Number of tillers and panicles hill−1, number of tillers and panicles m−2, and panicle length and number of grains panicle−1 were clearly superior with SRI compared to control plants. Farmers tested five varieties, all of which produced better under
SRI. The SRI system allowed for a seed reduction of 85–90%: from 40–60 kg ha−1 for the control plots to 6.1 kg ha−1 under SRI. Although production costs per hectare were 15% higher for SRI, revenue was 2.1 times higher than under the control.
Farmers were very satisfied with these results. In 2009/2010, Africare and the Government’s agriculture service worked with
over 270 farmers in 28 villages to scale up SRI practices and to test innovations, including composting techniques, optimization
of irrigation, and techniques to reduce labor requirements and production costs. The good crop performance along with other
advantages was confirmed in this third year with SRI yields of 7.7 t ha−1 (n = 130 farmers) compared to 4.5 t ha−1 in farmers’ fields. 相似文献

Root-knot nematodes (RKN) are obligate parasite species of the genus Meloidogyne that cause great losses in Arabica coffee (Coffea arabica L.) plantations. Identification of resistant genotypes would facilitate the improvement of coffee varieties aiming at an
environmental friendly and costless nematode control. In this work, the C. arabica genotype ‘UFV 408-28’ was found to be resistant to the most destructive RKN species M. incognita. Pathogenicity assays indicated that the highly aggressive populations of M. incognita races 1, 2 and 3 were not able to successfully reproduce on ‘UFV 408-28’ roots and displayed a low gall index (GI = 2). An
average reduction of 87% reduction of the M. incognita population was observed on ‘UFV 408-28’ when compared to the susceptible cultivar ‘IAC 15’. By contrast, ‘UFV 408-28’ was
susceptible to the related species M. exigua and M. paranaensis (GI = 5 and 4, respectively). Histological observations performed on sections of UFV408-28 roots infected with M. incognita race 1 showed that nematode infection could be blocked right after penetration or during migration and establishment stages,
at 6 days, 7 days and 8 days after infection (DAI). Fluorescence and bright field microscopy observations showed that root
cells surrounding the nematodes exhibited HR-like features such as accumulation of phenolic compounds and a necrotic cell
aspect. In the susceptible ‘IAC 15’ roots, 6 DAI, feeding sites contained giant cells with a dense cytoplasm. Necrotic cells
were never observed throughout the entire infection cycle. The HR-like phenotype observed in the ‘UFV 408-28’—M. incognita interaction suggests that the coffee resistance may be mediated by a R-gene based immunity system and may therefore provide new insights for understanding the molecular basis of RKN resistance
in perennial crops. 相似文献
