Proliferative enteropathy (PE) is one of the most important infections in pigs caused by Lawsonia intracellularis, an obligate intracellular bacterium. The purpose of the present investigation was to develop monoclonal antibodies with specificity to L. intracellularis useful both for diagnostic purposes (by immunohistochemistry) and for bacterial characterization. Several antibody producing hybridomas were established by fusion of mouse myeloma with spleen cells from BALB/c mice immunized with mucosa scrapings of the intestinal mucosa from a L. intracellularis infected pig. A monoclonal antibody (mAb), Law1-DK, isotyped as IgG2b was selected by indirect immunofluorescence antibody test (IFAT). Histological sections of the intestines from pigs affected by proliferative enteropathy and in vitro grown bacteria in cell culture were tested positive for the presence of L. intracellularis with the mAb. A molecule at 21 kDa was recognized by the mAb in a Western blotting analysis when a whole-cell preparation of L. intracellularis was run on a sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). This antigen was released from L. intracellularis by mild heat treatment and was resistant to proteinase K digestion, suggesting it to be non-protein, e.g., lipopolysaccharide (LPS). This suggestion was supported by its presence in the aqueous phase of a phenol-water extract. The inhibitory effect of periodate oxidation on the antigen-antibody binding confirmed the participation of a carbohydrate epitope. The new mAb was tested highly specific for L. intracellularis by applying in situ hybridization with a L. intracellularis specific probe targeting 16S ribosomal RNA simultaneously with the IFAT. 相似文献

The objective of the study was to investigate the hypothesis that perioperative lymphocytopenia was due to apoptosis of these
cells induced by either halothane or epidural anaesthesia in dogs. The relationship between apoptosis induction and plasma
concentrations of the stress hormone cortisol and the cytokines TNF-alpha and IL-10 was examined as well. The study was performed
on 22 healthy mongrel dogs, equal numbers from both genders, weighing 18.3 ± 2.9 kg, and aged between 3–5 years. Dogs were
divided in three groups. Eight of the animals were anaesthetized with halothane, another eight received epidural anaesthesia
using lidocaine, and six served as controls. Venous blood samples were obtained immediately before (0 minute) anaesthesia,
during deep anaesthesia (120 minute), and on the next day (24 hour) in order to determine the following parameters: the total
lymphocyte counts, the percentage of apoptotic peripheral blood mononuclear cells (PBMC) by flow cytometry, plasma concentrations
of the cytokines tumor necrosis factor – alpha (TNF-alpha) and interleukin – 10 (IL-10) by enzyme-linked immunosorbent assay
(ЕLISA), and plasma cortisol levels by radioimmune assay. Both halothane and epidural anaesthesia in dogs induces apoptosis
of PBMC with slight decrease in total lymphocyte counts. These immunomodulatory effects were transient and faded till the
24th hour. Concerning the mechanism of inducing lymphocyte apoptosis by general or epidural anaesthesia, it seemed that neither
cortisol, nor the tested cytokines TNF-alpha and IL-10 were implicated in this process. Further investigations are necessary

Fifty bacterial isolates obtained from compost were tested in vitro against the causal agents of green mould in Agaricus bisporus. Isolate B-38 which induced 48.08% in vitro growth inhibition of T. harzianum T54 and 52.25% of T. aggressivum f. europaeum T77 was identified as Bacillus subtilis, based on 16S rDNA sequence and used in mushroom growing room experiments. B. subtilis B-38 did not decrease mycelial growth rate of Agaricus bisporus A15 in mushroom compost in glass tubes. After applying prochloraz-manganese, B. subtilis B-38 and B. subtilis QST 713, no significant differences in BE values among treatments were found concerning both total yield and the weight of healthy mushrooms. Statistical analyses showed that only inoculation significantly influenced the healthy mushroom yield. In plots inoculated with T. harzianum T54 disease incidence was significantly lower after treatments with prochloraz-manganese (11.81%), B. subtilis QST 713 (12.26%) and B. subtilis B-38 (14.19%) compared to the control (28.16%), as well as in plots inoculated with T. aggressivum f. europaeum T77 11.88%, 12.2% and 15.03%, respectively, in comparison with the control (23.47%). Statistically significant differences were not found among the efficacy values of tested bio-fungicides based on B. subtilis and the commercial fungicide prochloraz-manganese suggesting the use of B. subtilis B-38 and B. subtilis QST 713 as good alternatives to chemical fungicides. 相似文献

Coffee leaf rust (CLR), caused by Hemileia vastarix, is one of the most serious diseases of coffee plantations and cause great losses in coffee production. We aimed to examine coffee varieties, shade, age of coffee plants, coffee plant density and soil properties in relation to CLR infection. To do this, we established a total of 75 plots in three agroforestry coffee plantations in the central Peruvian Amazon. We gathered data there in 2011 (dry season) on the presence/absence of CLR; coffee variety; age and density of coffee plants, and also took hemispherical photographs to determine canopy openness. In 2014 (wet season), we again gathered data on the same variables. In 2012, we collected soil samples from a subset of the plots. At all plantations, coffee variety had a significant effect on CLR incidence, with the Catimor variety infected less frequently than Caturra. Coffee plant age had a significant positive effect on CLR incidence. Increasing coffee density also increased CLR incidence for some of the studied plantations/seasons. Comparing those plots from which data were collected in the dry and wet seasons, we found that CLR presence was significantly higher in the wet season. The effect of shade on CLR incidence was not clear. Catimor and Caturra varieties showed opposite trends of CLR incidence in response to shade quantity in most cases (Caturra variety CLR incidence was decreasing with shading increase and Catimor CLR incidence decreasing with decreasing shading).
