Hot-iron branding uses thermal injury to permanently identify cattle causing painful tissue damage. The primary objective was to examine the physiological and behavioral effects of oral meloxicam (MEL), compared to a control, administered at the time of hot-iron branding in Angus and Hereford steers and heifers. The secondary objectives were to investigate breed and sex effects on pain biomarkers. A total of 70 yearlings, consisting of 35 heifers and 35 steers (Angus, Hereford, or Angus × Hereford), were enrolled in the study. Animals were blocked by sex, randomized across weight, and assigned to receive MEL (1 mg/kg) or a placebo (CON). Biomarkers were assessed for 48 h after branding and included infrared thermography (IRT), mechanical nociceptive threshold (MNT), accelerometry and a visual analog scale (VAS), and serum cortisol and prostaglandin E2 metabolites (PGEM). Wound healing was assessed for 12 wk. Hair samples to quantify cortisol levels were taken prior to and 30 d post-branding. Responses were analyzed using repeated measures with calf nested in treatment as a random effect, and treatment, time, treatment by time interaction, breed, and sex as fixed effects. There was a treatment by time interaction for PGEM (P < 0.01) with MEL having lower values than CON at 6, 24, and 48 h (MEL: 18.34 ± 3.52, 19.61 ± 3.48, and 22.24 ± 3.48 pg/mL, respectively; CON: 32.57 ± 3.58, 37.00 ± 3.52, and 33.07 ± 3.48 pg/mL; P < 0.01). MEL showed less of a difference in maximum IRT values between the branded (2.27 ± 0.29 °C) and control site (3.15 ± 0.29 °C; P < 0.01). MEL took fewer lying bouts at 0–12 h (4.91 bouts ± 0.56) compared with CON (6.87 bouts ± 0.55; P < 0.01). Compared with Hereford calves, Angus calves exhibited greater serum but lower hair cortisol, greater PGEM, more lying bouts, and less healed wound scores at 3, 4, and 5 wk. Compared with heifers, steers exhibited lower PGEM, lower branding site and ocular IRT, higher MNT, and lower plasma meloxicam levels. Steers spent more time lying, took more lying bouts and had greater VAS pain, and more healed wound scores at 5 wk than heifers. Meloxicam administration at branding reduced branding and control site temperature differences and reduced lying bouts for the first 12 h. Breed and sex effects were observed across many biomarkers. Changes from baseline values for IRT, MNT, lying time, step count, VAS pain, and wound scoring all support that branding cattle is painful.  相似文献

In Israel, black spot caused by Alternaria alternata is the main postharvest factor that impairs the quality and reduces the storability of persimmon fruit (Diospyros kaki cv. Triumph). The fungus infects the fruit in the orchard and remains quiescent until harvest. After harvest, the pathogen slowly colonizes the fruit during storage at 0 °C, which elicits black spot symptom development 2–3 months after storage entry. A commercial postharvest dip treatment in chlorine at 500 mg L?1, released from sodium troclosene tablets, effectively controlled black spot in fruit stored for up to 2 months. However, decay incidence increased as the length of storage was extended beyond 2.5 months. The long incubation period that precedes black spot symptom development after harvest enabled the development of a series of integrative approaches for application at the pre- and postharvest stages, in combination with the commercial chlorine dip treatment, to improve the control of black spot disease. Preharvest treatments included treatment with the cytokinin-like N1-(2-chloro-4-pyridyl)-N3-phenylurea (CPPU) 30 d after fruit set, or a single spray with the curative fungicide polyoxin B 14 d before harvest, and when one of these was applied in combination with the postharvest chlorine dip treatment, the black spot infected area was reduced by 3 and 60%, respectively, compared with the chlorine dip alone. At the postharvest stage, fogging during storage, or post-storage on-line spraying with sodium troclosene, when applied in combination with the postharvest chlorine dip, improved the percentage of marketable fruit by 2 or 10%, respectively, compared with the chlorine dip alone. The results indicate that postharvest pathogens that show a slow colonization pattern might enable the integration of pre- and postharvest disease control methods to improve quality and reduce postharvest disease development.  相似文献

This study describes the isolation of equine herpesvirus-2 (EHV-2) from the lung of an aborted equine fetus in Argentina. The isolated virus was confirmed as EHV-2 by indirect immunofluorescence using a rabbit anti-EHV-2 polyclonal antiserum and by virus-neutralization test using an equine polyclonal antibody against EHV-2. Restriction endonuclease DNA fingerprinting with BamHI also confirmed the identity of the virus as EHV-2. Furthermore, viral nucleic acid was detected by polymerase chain reaction from the original lung sample and from the DNA obtained from cells infected with the virus isolate. This work constitutes the first reported isolation of EHV-2 from an aborted equine fetus. The presence of EHV-2 in the lung of the aborted fetus would indicate that this virus is capable of crossing the placental barrier. However, no cause-effect relationship was established between the EHV-2 isolate and the abortion.  相似文献

The efficacy of oxolinic acid (at 200 and 300 μg a.i./l) and of several antibiotic compounds (streptomycin sulfate at 100 μg a.i./l, glycocide B at 700 μg a.i./l, kasugamycin at 80 μg a.i./l and gentamicin sulfate at 30 and 60 μg a.i./l) againstErwinia amylovora, the causal agent of fire blight in pears, was evaluated in 43 orchard experiments in 1997–2000 in Israel. In addition to the above orchard experiments, the efficacy of the bactericides was tested in live experiments with artificial inoculation. Natural fire blight symptoms were observed in 16 of the 43 experiments; in 13 of them, disease intensity and its distribution among the experimental plots provided a basis for data analysis, leading to reliable conclusions concerning the efficacy of the tested bactericides. Oxolinic acid at 300 μg a.i./l was highly effective againstE. amylovora and reduced disease severity significantly in all experiments, as compared with the untreated plots; however, a concentration of 200 μg a.i./l was not effective in some cases. Among the tested antibiotics, only gentamicin sulfate was as effective as oxolinic acid. Results of the artificial inoculation experiments corroborated those obtained in the naturally infected orchards. The pre-infection activity of oxolinic acid was determined on blossom clusters that were sprayed with the bactericide before inoculation. Control efficacy on blossom clusters sprayed 1–4 days before inoculation ranged from 68% to 80%, a level which did not differ significantly from that observed on blossom clusters sprayed on the day of inoculation (80% control). The postinfection activity of oxolinic acid was determined on blossom clusters that were sprayed with the compound after inoculation. Oxolinic acid was as effective when applied 1 or 2 days after inoculation as when it was applied on the day of inoculation; however, application of the bactericide 3 days after inoculation no longer resulted in significant disease suppression. Growth responses and nitrogen constituents were tested in young citrus trees grown on gradually decreasing amounts of phosphorus in greenhouse solution cultures. The phosphorus concentration in dry matter of minus-P leaves dropped to a third of that in control leaves and then remained steady throughout the experiment, while the total nitrogen concentration was low at the beginning and, rising gradually, reached an enormous amount. Nitrogen estimations in minus-P leaves showed the specific dynamics of the soluble nitrogen fraction: its content was low at the onset of the deficiency, and rose rapidly due to intensive accumulation of arginine in the tissues. Under conditions of acute phosphorus deficiency, arginine constituted more than 60% of the free amino acids and arginine nitrogen above 40% of the total, while in normal leaves arginine did not exceed 8% of the free amino acids, and about 1–% of the total.Similar patterns of increased nitrogen level due to arginine accumulation were found in leaves of phosphorus-deficient grapefruit and orange trees under field conditions.  相似文献

Crude extracts and fractions of Bertholletia excelsa stem barks were tested for trypanocidal activity. Acetone and methanol extracts showed significant in vitro trypanocidal activity against trypomastigote form of Trypanosoma cruzi since in the concentration of 500 microg/ml, the parasites were reduced in 100% and 90.3% respectively, whereas the triterpene betulinic acid pure isolated from hexane extract presented 75.4%.  相似文献

A young adult Labrador retriever dog was presented for surgical debulking of hepatic alveolar echinococcosis. Computed tomography detected hepatomegaly with multiple large cavitary masses with extension of tissue from a lesion wall into the caudal vena cava and numerous nodules in all lung lobes. Following euthanasia, histology confirmed parasitic vesicles with granulomatous reaction in all lesions, and polymerase chain reaction (PCR) established the causative agent to be Echinococcus multilocularis. This report is the first to present imaging features of pulmonary E. multilocularis granulomata in a dog.  相似文献

The role of phospholipase A2 in the induction of drip loss from pig muscle has been investigated. In samples from porcine M. longissimus dorsi, total PLA2 activity as well as mRNA and protein levels of the group VIA iPLA2 (iPLA2-VIA) increased during the initial 4 h post-mortem period. Morphological studies of porcine muscle showed that at 4 h post-mortem, gaps had formed between muscle fibers and that the sarcolemma membrane borders appeared blurred. At the same time iPLA2-VIA protein levels were increased inside muscle fibers and at the sarcolemma. iPLA2-VIA mRNA abundance in samples from different breeds of pigs with variations in drip loss revealed no clear correlation between drip loss level and iPLA2-VIA expression. Together, these data indicate that during the post-mortem period, iPLA2-VIA expression and activity is increased at the muscle fiber membranes. PLA2 activity may affect membrane permeability and consequently the progression of drip formation in porcine muscle.  相似文献

Long pentraxin 3 (PTX3) is a conserved pattern-recognition secreted protein and a host-defence-related component of the humoral innate immune system. The aim of the present study was to characterize swine PTX3 (SwPTX3) protein expression in influenza virus infected pigs. First, we performed in silico studies to evaluate the cross-reactivity of PTX3 human antibodies against SwPTX3. Secondly, we used in vitro analysis to detect SwPTX3 presence in swine bone marrow dendritic cells (SwBMDC) upon stimulation with different agents by Western blot and immunofluorescence. Finally, the levels of SwPTX3 were assessed in experimental infection of pigs with different strains of influenza virus. This is a novel study where the expression of SwPTX3 was evaluated in the context of a pathogen infection. The initial characterization of SwPTX3 in influenza virus infected pigs contributes to understand the role of PTX proteins in the immune response.  相似文献
