Modulating sphingolipid biosynthetic pathway rescues photoreceptor degeneration

Mutations in proteins of the Drosophila phototransduction cascade, a prototypic guanine nucleotide-binding protein-coupled receptor signaling system, lead to retinal degeneration and have been used as models to understand human degenerative disorders. Here, modulating the sphingolipid biosynthetic pathway rescued retinal degeneration in Drosophila mutants. Targeted expression of Drosophila neutral ceramidase rescued retinal degeneration in arrestin and phospholipase C mutants. Decreasing flux through the de novo sphingolipid biosynthetic pathway also suppressed degeneration in these mutants. Both genetic backgrounds modulated the endocytic machinery because they suppressed defects in a dynamin mutant. Suppression of degeneration in arrestin mutant flies expressing ceramidase correlated with a decrease in ceramide levels. Thus, enzymes of sphingolipid metabolism may be suitable targets in the therapeutic management of retinal degeneration.     

How the Orthopedic Foundation for Animals (OFA) is tackling inherited disorders in the USA: using hip and elbow dysplasia as examples

The Orthopedic Foundation for Animals (OFA) maintains an on-line health pedigree database for inherited disorders of animals. With the American Kennel Club Canine Health Foundation, the OFA maintains the Canine Health Information Center (CHIC) for parent breed clubs to identify breed-specific required health tests. Analysis of the results of OFA evaluations in the hip and elbow registries show that selection based on phenotype improves conformation. Disorders with complex inheritance respond best to selection based on depth (ancestors) and breadth (siblings) of pedigree health test results. This information can be derived from vertical pedigrees generated on the OFA website.     

Collaborative study comparing the spiral plate and aerobic plate count methods.

The spiral plate count method is a semiautomated plating technique that greatly reduces manpower and material costs normally associated with the pour plating technique. In this collaborative study, 8 laboratories compared the spiral and pour plating techniques, using 4 samples each of 3 products: frozen pumpkin pie, frozen chicken pot pie, and shampoo. The results show that 10 of the 12 comparisons of means of the pour and spiral methods were not significantly different; 2 values were significant at alpha = 0.01. Overall, the components of variance were less than that of the current milk standard, and the replicate per cent coefficient of variation was satisfactory. This study indicates that the spiral plate method is an acceptable alternative to the pour plate method; the spiral plate method has been adopted as official first action.
