Nineteen multiparous barren Holstein cows were subjected to an induction of lactation protocol for 21 d administering estradiol cypionate (2 mg kg−1 of body weight (BW) d−1, on day 1 to 14), progesterone (0.10 mg kg−1 of BW, on day 1 to 7), flumethasone (0.03 mg kg−1 of BW, on day 18 to 20) and recombinant bovine somatotropin (rbST; 500 mg per cow, on day 1, 6, 16 and 21). At the end of lactation and with a minimum of a 2-mon dry period, the same cows were again hormonally induced into lactation. Cows in both lactations were not artificially inseminated, they were milked 3 times daily and received rbST throughout lactation. Mean accumulated milk yield at 305 d in milk (DIM) did not differ between the first and second induced lactations ((9 710 ±1 728) vs. (9 309±2 150) kg; mean±SD). Total milk yield ((12 707±3 406) vs. (12 306±4 218) kg; mean±SD) and lactation length ((405±100) vs. (410±91) d; mean±SD) were not different between the first and second induced lactations. In a second study, 15 empirical models including exponential, power law, yield-density, sigmoidal and miscellaneous models were compared for their suitability by modeling 12-mon (n=334), 18-mon (n=164) and 29-mon (n=22) lactation cycles of Holsteins cows induced into lactation and treated with rbST throughout the lactation. Hoerl (Y= ab1/xxc), Wood (Y=axb exp(cx)) and Dhanoa (Y=ax(bc)exp(cx)) models were equally suitable to describe 12-mon lactations. An exponential model with five parameters (Y=exp(a+bx+cd2+e/x)) showed the best fit for milk yield for 18-mon lactations. The rational model (Y=a+bx/1+cx+dx2) was found to produce the closest fit for 29-mon lactations. It was concluded that, with the protocol used in the present study, multiparous cows respond favorably to a second cycle of induced lactation, with milk yield similar to that experienced during the first cycle. Thus, dairy producers might be able to lengthen the productive life of infertile high producing cows with a renewal of artificial lactation, which would imply an overall reduction in voluntary culling of cows. Also, various equations used to describe the lactation curves demonstrated the potential for fitting monthly milk records of Holstein cows with prolonged lactations and induced hormonally into lactation.  相似文献

A long‐term field experiment on a Haplic Phaeozem, established 1949 with four levels of potassium (K) supply (5, 69, 133, and 261 kg K ha?1), was analyzed for the interaction between K supply and yield loss of five crop species by water shortage. The crop species were cultivated simultaneously side‐by‐side in the following rotation: potato (Solanum tuberosum L.), silage maize (Zea mays L.), spring wheat (Triticum aestivum L.), beet (Beta vulgaris L.), and spring barley (Hordeum vulgare L.). The treatment with 133 kg K ha?1 supply had a nearly balanced K budget. In the treatments with lower supply, the soil delivered K from its mineral constituents. On the low‐K plots (especially on those with only 5 kg K ha?1), crops suffered yield depressions of nearly all main harvest products (cereal grains, potato tubers, beet storage roots, silage maize) and by‐products (straw, beet leaves) by up to 40.7% of dry matter. Only wheat grains were an exception. Potassium concentrations in the harvested plant parts decreased nearly in parallel to the reduction of their dry matter yields, with the exception of cereal grains, which kept stable concentrations even in the treatment with only 5 kg K ha?1. A comparison of four year‐pairs with differing levels of precipitation in yield‐relevant periods showed an average water shortage‐induced depression of dry matter yields by 19.7% in the main harvest products. The severity of this yield depression was not mitigated by elevated K supply, with the exception of beet leaves, where the dry matter production was stabilized by high K supply. In this crop, the reduction of storage‐root yield was associated with a decrease in harvest index and was therefore obviously caused by an inhibition of assimilate translocation from the leaves into these organs, in contrast to cereals, where water shortage primarily affected dry matter production in vegetative organs. It is concluded that the physiological causes of yield reduction by drought stress and the possibility of its amelioration by K supply differ between plant species and organs.  相似文献

Many tropical soils include sesquioxides, which influence the stability of soil organic matter (OM) and aggregation to an extent that is not fully characterized. The present study was carried out on a range of 18 topsoil samples (0–10 cm) from low-activity clay (LAC) soils from sub-Saharan Africa and Brazil, and aimed: (i) at characterizing the size distributions of water-stable aggregates and organic constituents, (ii) at studying how these distributions were affected by texture and sesquioxides, and (iii) how they interacted.The distributions of stable aggregates were generally dominated by macroaggregates (> 200 μm), and those of organic constituents by fine OM (< 20 μm). Aggregation was not clearly affected by soil texture, while total soil carbon (Ct) and the amount of carbon (C) as fine OM increased with soil content in clay plus fine silts (< 20 μm). Stable macroaggregation correlated with Ct and with C amount as fine OM, but each of them correlated more closely with citrate-bicarbonate-dithionite-extractable aluminium (Al), which was not expected. Stable macroaggregation also correlated with C amounts as coarse- and medium-sized OM (> 200 and 20–200 μm, respectively), but each of them correlated more closely with oxalate-extracted Al. These results suggested that for the LAC tropical soils under study OM and aggregate stability depended closely on Al-containing sesquioxides, on Al-substituted crystalline hematite and goethite especially. These sesquioxides also seemed to play a dominant role in the relations between aggregation and OM. As far as soils rich in sesquioxides are concerned, this confirmed that OM is not the main aggregating agent, and suggested that physical protection within aggregates is not necessarily the main mechanism for OM stabilization. However, as soil sesquioxide content cannot be managed easily, the effect of land use on soil OM and aggregation was determinant at the local scale: indeed, for a given location, stable macroaggregation, Ct and C amount as fine OM generally decreased with land use intensification (i.e. cultivation, tillage, reduced surface cover).  相似文献

Ecological restoration has become an important technique for mitigating the human impacts on natural vegetation. Planting seedlings is the most common approach to regain lost forest cover. However, these activities require a large economic investment. Direct seeding is considered a cheaper and easier alternative technique, in which tree seeds are introduced directly on the site rather than transplanting seedlings from nurseries. To evaluate the effectiveness of direct seeding, we conducted a comprehensive search of the literature using ‘restoration’, ‘direct seeding’ and ‘sowing’ as keywords, and we performed a meta‐analysis using 30 papers and 89 species. We used two different measures of restoration success: seed germination probability and success probability (the chance that a seed germinates and survives until the end of the experiment). In general, restoration attempts using direct‐seeding techniques were relatively unsuccessful. On average, seed germination and success probability were 0·239 and 0·114, respectively, and were not affected by climate, species successional group or the application of pre‐germinative treatments. Germination and success probability increased with seed size, and the use of physical protections resulted in a nearly twofold increase in germination probability, but this effect faded by the end of the experiments. Because of the low rate of seedling success, we suggest the use of direct seeding as a complementary technique to reduce restoration costs, particularly for species with large seeds and known high germination rates, but our results do not support direct seeding as a substitute for seedling planting. Purification of the lectin from Phaseolus acutifolius var. escumite was achieved by affinity chromatography on a column containing glutaraldehyzed membranes from blood group O erythrocytes. The lectin is a tetrameric glycoprotein of 121 kDa with 10% of sugar by weight composed by four subunits of 30 kDa as determined by SDS-PAGE. The lectin is composed of four isolectins as determined by ion-exchange chromatography on a mono-S column. The lectin and its isolectins showed identical NH2 terminal residues (ANDLSFNFQR FNETN) with homology to the PHA leucoagglutinin-precursor. Peptide mass fingerprint from each lectin isoform determined from tryptic peptides by MALDI-TOF (matrix assisted laser desorption ionization-time-of-flight) showed differences among subunits, thus suggesting microheterogeneity in their amino acid sequences or different glycosylation patterns. The lectin and its four isolectins agglutinated erythrocytes without serological specificity and showed mitogenic activity on human leukocytes; moreover, the main effect was rather toward CD8+ than to CD4+ human peripheral lymphocytes. The lectin from escumite was not inhibitable by simple sugars; however, the specificity of the lectin and its isoforms was mainly addressed toward galactose residues present in bi- or triantennary N-acetyllactosamine-type glycans.  相似文献

Various methods have been used previously to impart hydrophobicity to cotton. In this study, thin co-polymer films of trifluoroethyl acrylate (TFEA) and octafluoro pentyl methacrylate (OFPM) were formed on cotton using individual fluorosurfactants and surfactant mixtures through the process of admicellar polymerization. Efficacy of 6 different fluorosurfactant systems and process conditions including surfactant type, concentration, adsorption/adsolubilization time, polymerization time, and monomer concentration have been investigated in order to obtain hydrophobic coatings on woven cotton fabric. Adsolubilization times as short as 2 h and polymerization times as short as 1 h yielded satisfactory performance. Characterization of the hydrophobic polymer films obtained included static contact angles, SEM imaging, elemental analysis and XPS analysis. Treated fabrics exhibited good water repellency for several surfactants and surfactant mixtures with initial supernatant monomer concentrations of 4 mM. Surfactant mixtures were able to outperform the separate individual species in admicellar polymerization.  相似文献

为提供信息改良油料种子脂肪酸,研究脂肪酸碳链延长酶1(FAE1)的底物特异性,将甘蓝型油菜A、C基因组的BnaA.FAE1和BnaC.FAE1分别在亚麻荠种子中表达。其转基因种子中山嵛酸与芥酸含量的比值(C22:0/C22:1)分别为0.18和0.88,表明BnaA.FAE1对单不饱和脂肪酸亲和力较高,而BnaC.FAE1则对饱和脂肪酸亲和力较高,这种差异在各转基因世代表现稳定。通过分析T3转基因家系种子中酰基辅酶A(Acyl-CoAs)的组成,进一步证明了上述结论。但是在拟南芥中分别异源表达上述基因,并没有观察到类似的底物特异性差异。在甘蓝型油菜祖先种,白菜型油菜和甘蓝种子中调查其C22:0/C22:1比值,同样未发现其FAE1底物特异性存在差异。综上认为BnaA.FAE1和BnaC.FAE1在亚麻荠中存在底物特异性。  相似文献

Sulphated polysaccharides (SP) extracted from seaweeds have antiviral properties and are much less cytotoxic than conventional drugs, but little is known about their mode of action. Combination antiviral chemotherapy may offer advantages over single agent therapy, increasing efficiency, potency and delaying the emergence of resistant virus. The paramyxoviridae family includes pathogens causing morbidity and mortality worldwide in humans and animals, such as the Newcastle Disease Virus (NDV) in poultry. This study aims at determining the antiviral activity and mechanism of action in vitro of an ulvan (SP from the green seaweed Ulva clathrata), and of its mixture with a fucoidan (SP from Cladosiphon okamuranus), against La Sota NDV strain. The ulvan antiviral activity was tested using syncytia formation, exhibiting an IC50 of 0.1 μg/mL; ulvan had a better anti cell-cell spread effect than that previously shown for fucoidan, and inhibited cell-cell fusion via a direct effect on the F0 protein, but did not show any virucidal effect. The mixture of ulvan and fucoidan showed a greater anti-spread effect than SPs alone, but ulvan antagonizes the effect of fucoidan on the viral attachment/entry. Both SPs may be promising antivirals against paramyxovirus infection but their mixture has no clear synergistic advantage.  相似文献
