Changes in structural and chemical components of wood delignified by fungi   总被引:3,自引:0,他引:3  
Summary Cerrena unicolor, Ganoderma applanatum, Ischnoderma resinosum and Poria medulla-panis were associated with birch wood that had been selectively delignified in the forest. Preferential lignin degradation was not uniformly distributed throughout the decayed wood. A typical white rot causing a simultaneous removal of all cell wall components was also present. In the delignified wood, 95 to 98% of the lignin was removed as well as substantial amounts of hemicelluloses. Scanning and transmission electron microscopy were used to identify the micromorphological and ultrastructural changes that occurred in the cells during degradation. In delignified areas the compound middle lamella was extensively degraded causing a defibration of cells. The secondary wall, especially the S2 layer, remained relatively unaltered. In simultaneously white-rotted wood all cell wall layers were progressively removed from the lumen toward the middle lamella causing erosion troughs or holes to form. Large voids filled with fungal mycelia resulted from a coalition of degraded areas. Birch wood decayed in laboratory soil-block tests was also intermittently delignified. Selective delignification, sparsely distributed throughout the wood, and a simultaneous rot resulting in the removal of all cell wall components were evident. Scanning electron microscopy appears to be an efficient technique for examining decayed wood for fungi with the capacity to selectively delignify wood.The authors would like to thank Kathy Zuzek for technical assistance and Dr. M. Brassicaceous plants rich in glucosinolates have been used as biofumigants for the management of soilborne pathogens. Efficacy of Brassica plant tissue has mainly been attributed to toxic isothiocyanates released upon the hydrolysis of glucosinolates. Management of Phytophthora cinnamomi, the causal agent of oak root rot in rangeland ecosystems using biofumigation, is promising, but requires further validation. The biofumigation activity of 14 brassicaceous plants was evaluated under experimental conditions. All evaluated plants rich in sinigrin suppressed (100%) the mycelial growth of P. cinnamomi, while plants rich in aromatic or other aliphatic glucosinolates had little or no suppressive effect. Simulating soil amendment in field conditions, the effects on natural soil artificially infested with P. cinnamomi chlamydospores were examined with Brassica juncea, Eruca vesicaria and Lepidium sativum, three species with different glucosinolate profiles. Only B. juncea decreased the viability of chlamydospores significantly in comparison with untreated soil only 1 day after biofumigation, whereas E. vesicaria needed 8 days to reach significance and L. sativum had no effect at all. Despite the decreases in soil inoculum, biofumigation with B. juncea did not prevent the root infections in a highly susceptible host (Lupinus luteus). However, biofumigation with plants rich in sinigrin, such as B. juncea, decreased P. cinnamomi soil inoculum under the experimental minimum threshold for oak disease expression. Although biofumigation should be considered as an effective measure to be incorporated in integrated control of the oak disease, biofumigation by itself would not be effective enough for the substantial suppression of P. cinnamomi inoculum.  相似文献

Effects of 10 microM cadmium (supplied as Cd nitrate) on the utilization and allocation of iron (Fe) were investigated in poplar (Populus alba L.) plants grown in nutrient solution with Fe(III)-EDTA or Fe(III)-citrate as the Fe source. The effects of Cd were also compared with those of Fe deprivation. The accumulation of Fe in roots was 10-fold higher in plants grown with Fe-citrate than with Fe-EDTA. Cadmium decreased leaf chlorophyll concentrations and photosynthetic rates, and these decreases were more marked in plants grown with Fe-citrate than with Fe-EDTA. In both Fe treatments, addition of Cd caused large increases in root and shoot apoplasmic and non-apoplasmic Cd contents and increases in root Fe content; however, Cd decreased shoot Fe content, especially in plants grown with Fe-citrate. New leaves of plants grown with Fe-citrate had small cellular (non-apoplasmic) Fe pools, whereas these pools were large in new leaves of plants grown with Fe-EDTA. Non-apoplasmic Cd pools in new leaves were smaller in plants grown with Fe-citrate than with Fe-EDTA, indicating that inactivation of non-apoplasmic Cd pools is facilitated more by Fe-EDTA than by Fe-citrate. In the presence of Cd, Fe-EDTA was also superior to Fe-citrate in maintaining an adequate Fe supply to poplar shoots. Differences in plant responses to Fe-EDTA and Fe-citrate may reflect differences in long-distance transport of Fe rather than in acquisition of Fe by roots.  相似文献

Within a research project on quality of plants for forestation of agricultural lands, we studied the aetiology of a late seedling rot affecting holm oak (Quercus ilex ssp. ballota) in two forest nurseries in southern Spain. Major disease symptoms were foliage wilting and necrosis of feeder roots. Phytophthora cinnamomi, Phytophthora cryptogea and Phytophthora drechsleri were isolated from necrotic roots of holm oaks. Selected isolates of the three Phytophthora species were pathogenic to Quercus ilex ssp. ballota and Quercus suber seedlings in artificial inoculations. Soil flooding conditions were essential for infection and root rot development. There was no host specificity among the species, the isolates of P. cinnamomi being the most virulent in all inoculated plants. In these inoculations, Q. ilex ssp. ballota plants were more susceptible than those of Q. suber. This work is the first report of P. cinnamomi, P. drechsleri and P. cryptogea affecting Q. ilex ssp. ballota in forest nurseries.  相似文献

The use of overhead trellis systems for the production of dry-on-vine (DOV) raisins and table grapes in California is expanding. Studies were conducted from 2006 to 2009 using Thompson Seedless grapevines grown in a weighing lysimeter trained to an overhead arbor trellis and farmed as DOV raisins for the first two years and for use as table grapes thereafter. Maximum canopy coverage for the two lysimeter vines across years was in excess of 80 %. Seasonal (15 March–31 October) evapotranspiration for the lysimeter vines (ETLys) was 952 mm in 2007 (farmed as DOV raisins) and 943 and 952 mm (when farmed as table grapes). The maximum crop coefficient (K cLys) across all 4 years ranged from 1.3 to 1.4. These maximum values were similar to those estimated using the relationship where K c is a function of the amount of shaded area measured beneath the canopy at solar noon (K c = 0.017 × percent shaded area). Covering the lysimeter’s soil surface with plastic (and then removing it) numerous times during the 2009 growing season (1 June–14 September) reduced ETLys from an average of 6.4 to 5.6 mm day?1 and the K c from 1.07 to 0.93. A seasonal basal K c (K cb) was calculated for grapevines using an overhead trellis system with a 13 % reduction in the K cLys across the growing season.  相似文献

Micro-irrigation technologies are promoted for various reasons in India. Despite the reported significant economic advantages, and the concerted support of the government and NGOs, the current micro-irrigation area in India remains an insignificant proportion of its potential. This paper analyzes: (1) the economics of alternative micro-irrigation technologies, (2) the determinants of adoption, (3) the poverty outreach of the different micro-irrigation systems, and (4) the sustainability implications of micro-irrigation adoption. In line with the findings of other studies, this study indicates that micro-irrigation technologies result in a significant productivity and economic gains. The most important determinants of micro-irrigation adoption include access to groundwater, cropping pattern, availability of cash, and level of education, the social status and poverty status of the farmer. Contrary to the expectations, the majority of the current adopters of low-cost micro-irrigation systems are the better-off farmers. The study indicates that the impact of micro-irrigation systems on the sustainability of groundwater resources depends upon the magnitude of the overall productivity gain following the shift from traditional irrigation method to micro-irrigation system, the pattern of use of the saved water, and the type and potential number of adopters. BACKGROUND: Greenhouse and nursery producers use entomopathogens (nematodes and fungi) to control soil pests. Although it is known that the physical and chemical properties of mineral soil significantly impact upon soil pathogens, the influence of soilless media used for plant production on entomopathogen performance is poorly understood. RESULTS: Survival and foraging distance were differently affected by sand:peat, bark and sawdust media for entomopathogenic nematodes, but not for the immobile fungus Metarhizium anisopliae. Redwood sawdust medium consistently had a negative impact upon entomopathogenic nematodes. Dividing media into individual components supported the hypothesis that redwood sawdust reduced foraging and infection abilities of S. riobrave and H. bacteriophora. Physically altering the components by adding sand significantly improved foraging and infection success for S. riobrave in media not optimum for foraging. CONCLUSION: This study is the first to highlight the importance of selecting the appropriate soilless media and pathogen species combinations to increase efficacy of biological control. H. bacteriophora was able to find hosts in a wider diversity of medium components than S. riobrave, although both nematode species performed well in peat moss and recycled plant material. These results suggest that peat moss, recycled plant material and hardwood bark are components amenable to EPN biological control programs. Copyright © 2012 Society of Chemical Industry  相似文献

This report describes an outbreak of Mycoplasma bovis mastitis affecting 45 cows in a herd of 122 dairy cattle in Northern Italy. Clinically, the outbreak was characterized by agalactia, multiple swollen and painless quarters, high milk somatic cell count and unresponsiveness to conventional antibiotic therapy. M. bovis was isolated from the milk samples of all the 32 affected cows tested and from the mammary tissue of three affected cows that underwent necropsy. No other pathogens were isolated from these samples. Lesions in two of the necropsied cows were characterized by mild chronic suppurative mastitis and galactophoritis. The other necropsied cow showed a chronic necrosuppurative and pyogranulamaous galactophoritis, a condition not previously associated with M. bovis. M. bovis was detected immunohistochemically in the lumen of the affected mammary ducts suggesting that ascending infection via the teat canal was the likely route of transmission. No other intralesional pathogens were demonstrated microscopically.  相似文献

Background

Canine influenza virus (CIV) H3N8 infection is thought to be highly contagious and potentially deadly. Low seroprevalence in dogs participating in the sport of flyball in 2009 was reported. It remains unknown if prevalence is changing in dogs at risk.

Objective

To determine seroprevalence of antibodies to CIV H3N8 in a population of pet dogs that participated in an indoor flyball tournament in Pennsylvania in November 2010.

Methods

Medical, travel, and activity histories for previous 10.5 months were obtained from consenting owners and serum samples were collected from dogs. Antibodies to CIV H3N8 were measured using hemagglutination inhibition.

Results

Of 251 competing dogs, 103 dogs were tested. Overall, seroprevalence of CIV H3N8 in dogs participating in flyball was 1.9% (95% CI, 0.05–6.8%). Thirty‐five dogs were tested in both 2009 (all titers <8) and 2010 (all titers <8). Ten of 103 dogs had been vaccinated for CIV H3N8. Two dogs had positive titers (>8) and both had been vaccinated twice in the previous month. Titers in remaining 8 vaccinated dogs were undetectable. Seroprevalence in vaccinated dogs was 20% (95% CI, 5.7–50.1%), whereas in unvaccinated dogs it was 0% (95% CI, 0–4.0%). Although 7 dogs had history of respiratory signs, only 1, which was vaccinated, was serologically positive for CIV H3N8.

Conclusions

Consistent with low prevalence of CIV H3N8 exposure previously reported in flyball dogs, unvaccinated dogs had 0% prevalence, suggesting a stable exposure rate. However, low seroprevalence may result in unprotected flyball dogs at risk for developing CIV H3N8.  相似文献
