Gas transport in soils is usually assumed to be purely diffusive, although several studies have shown that non-diffusive processes can significantly enhance soil gas transport. These processes include barometric air pressure changes, wind-induced pressure pumping and static air pressure fields generated by wind interacting with obstacles. The associated pressure gradients in the soil can cause advective gas fluxes that are much larger than diffusive fluxes. However, the contributions of the respective transport processes are difficult to separate. We developed a large chamber system to simulate pressure fields and investigate their influence on soil gas transport. The chamber consists of four subspaces in which pressure is regulated by fans that blow air in or out of the chamber. With this setup, we conducted experiments with oscillating and static pressure fields. CO2 concentrations were measured along two soil profiles beneath the chamber. We found a significant relationship between static lateral pressure gradients and the change in the CO2 profiles (R2 = 0.53; p-value <2e-16). Even small pressure gradients between −1 and 1 Pa relative to ambient pressure resulted in an increase or decrease in CO2 concentrations of 8% on average in the upper soil, indicating advective flow of air in the pore space. Positive pressure gradients resulted in decreasing, negative pressure gradients in increasing CO2 concentrations. The concentration changes were probably caused by an advective flow field in the soil beneath the chamber generated by the pressure gradients. No effect of oscillating pressure fields was observed in this study. The results indicate that static lateral pressure gradients have a substantial impact on soil gas transport and therefore are an important driver of gas exchange between soil and atmosphere. Lateral pressure gradients in a comparable range can be induced under windy conditions when wind interacts with terrain features. They can also be caused by chambers used for flux measurements at high wind speed or by fans used for head-space mixing within the chambers, which yields biased flux estimates.  相似文献

Woodpastures (open, grazed woodlands with a mosaic of grassland, shrub and tree patches) are of high biological and cultural value and have become a threatened ecosystem in Europe. Spontaneous tree regeneration in the presence of large herbivores, is an essential process for management and restoration of this structurally diverse habitat. We examined the suitability of five vegetation types (grasslands, ruderal vegetations, tall sedges, rush tussocks and bramble thickets), grazed by large herbivores, for tree regeneration. We hypothesized that bramble thickets and tall herb communities operate as safe sites for palatable tree species through the mechanism of associational resistance. We set up a field experiment with tree seedlings in grazed and ungrazed conditions and recorded mortality and growth of seedlings of two palatable tree species (Quercus robur and Fraxinus excelsior) during three growing seasons. In the same experiment, we studied the effect of a two year’s initial time gap before grazing.Bramble thickets were suitable safe sites for survival and growth of seedlings of both species. Tall sedges, soft rush tussocks and ruderal vegetations with unpalatable or spiny species provided temporal protection, allowing seedlings to survive. Tree regeneration in livestock grazed grassland was highly constrained. Rabbits may undo the nursing effects of bramble thickets. The first year’s survival is of major importance for the establishment of trees. Subsequent grazing affects growth rather than survival. A two year’s initial time gap before grazing, had positive effects on survival, but did not enhance outgrowth of unprotected trees.  相似文献

In the analysis of food additives, past emphasis was put on the development of chromatographic techniques to separate target components from a complex matrix. Especially in the case of artificial food colors, direct spectrophotometric measurement was seen to lack in specificity due to a high spectral overlap between different components. Multivariate curve resolution (MCR) may be used to overcome this limitation. MCR is able to (i) extract from a complex spectral feature the number of involved components, (ii) attribute the resulting spectra to chemical compounds, and (iii) quantify the individual spectral contributions with or without a priori knowledge. We have evaluated MCR for the routine analysis of yellow and blue food colors in absinthe spirits. Using calibration standards, we were able to show that MCR equally performs as compared to partial least-squares regression but with much improved chemical information contained in the predicted spectra. MCR was then applied to an authentic collective of different absinthes. As confirmed by reference analytics, the food colors were correctly assigned with a sensitivity of 0.93 and a specificity of 0.85. Besides the artificial colors, the algorithm detected a further component in some samples that could be assigned to natural coloring from chlorophyll.  相似文献

In 2006 bluetongue (BT) emerged for the first time in North-Western Europe. Reliable diagnostic tools are essential in controlling BT but data on the diagnostic sensitivity (Se) and specificity (Sp) are often missing. This paper aims to describe and analyse the results obtained with the diagnostics used in Belgium during the 2006 BT crisis. The diagnosis was based on a combination of antibody detection (competitive ELISA, cELISA) and viral RNA detection by real-time RT-PCR (RT-qPCR). The performance of the cELISA as a diagnostic tool was assessed on field results obtained during the epidemic and previous surveillance campaigns. As the infectious status of the animals is unknown during an epidemic, a Bayesian analysis was performed. Both assays were found to be equally specific (RT-qPCR: 98.5%; cELISA: 98.2%) while the diagnostic sensitivity of the RT-qPCR (99.5%) was superior to that of the cELISA (87.8%). The assumption of RT-qPCR as standard of comparison during the bluetongue virus (BTV) epidemic proved valid based on the results of the Bayesian analysis. A ROC analysis of the cELISA, using RT-qPCR as standard of comparison, showed that the cut-off point with the highest accuracy occurred at a percentage negativity of 66, which is markedly higher than the cut-off proposed by the manufacturer. The analysis of the results was further extended to serological and molecular profiling and the possible use of profiling as a rapid epidemiological marker of the BTV in-field situation was assessed. A comparison of the serological profiles obtained before, during and at the end of the Belgian epidemic clearly showed the existence of an intermediate zone which appears soon after BTV (re)enters the population. The appearance or disappearance of this intermediate zone is correlated with virus circulation and provides valuable information, which would be entirely overlooked if only positive and negative results were considered.  相似文献

Clostridium perfringens which is a causative agent of several diseases in animals and humans is capable of producing a variety of toxins. Isolates are typed into five types on the basis of the presence of one or more of the four major toxins genes, i.e. cpa, cpb, etx, and iap. A decade ago another toxin termed beta2 (beta2) and its gene (cpb2) were identified. Two alleles of cpb2 are known and a possible link between differences in gene expression and allelic variation has been reported. A correlation between the level of expression and the origin of the isolates has also been suggested. The demonstration and typing of the cpb2 gene in the genome of isolates can be seen as a vital part of research on the role of the beta2 toxin in the pathogenesis of disease. This study describes a PCR with a single primer set which in contrast to published primer sets recognizes both alleles. Subsequent restriction enzyme analysis of the PCR product enables typing of the alleles. Applying this protocol on a total of 102 isolates, a sub-variant was found which occurred only in C. perfringens isolates from pigs and appeared to be the predominant variant found in C. perfringens isolates from this species.  相似文献

Objective To estimate the risks of anaesthetic and sedation‐related mortality in companion animals in the UK. (The Confidential Enquiry into Perioperative Small Animal Fatalities, CEPSAF). Study design A prospective cohort study with nested case–control study. Animal population All small animals anaesthetized and sedated at participating centres between June 2002 and June 2004. Methods Patient outcomes at 48 hours (alive, dead and killed) were recorded. Anaesthetic and sedation‐related death was defined as death where surgical or pre‐existing medical causes did not solely cause death. Species‐specific risks of anaesthetic‐related death and 95% confidence intervals (95% CI) were calculated. Risks were also estimated in the sub‐sets of dogs, cats and rabbits that were either healthy or sick (ASA 1–2 and 3–5, respectively). Results One hundred and seventeen veterinary practices participated in the study and 98 036 dogs, 79 178 cats and 8209 rabbits were anaesthetized and sedated. Overall risks of anaesthetic and sedation‐related death in dogs were 0.17% (1 in 601, 95% CI 0.14–0.19%), in cats 0.24% (1 in 419, 95% CI 0.20–0.27%) and in rabbits 1.39% (1 in 72, 95% CI 1.14–1.64%) within 48 hours of the procedure. In healthy dogs, cats and rabbits, the risks were estimated to be 0.05% (1 in 1849, 95% CI 0.04–0.07%), 0.11%, (1 in 895, 95% CI 0.09–0.14%) and 0.73% (1 in 137, 95% CI 0.54–0.93%), respectively. In sick dogs, cats and rabbits, the risks were 1.33%, (1 in 75, 95% CI 1.07–1.60%), 1.40% (1 in 71, 95% CI 1.12–1.68%) and 7.37% (1 in 14, 95% CI 5.20–9.54%), respectively. Postoperative deaths accounted for 47% of deaths in dogs, 61% in cats and 64% in rabbits. Most other small animal species had higher mortality risks. Conclusions and clinical relevance Small animal anaesthesia appears to be increasingly safe. Greater patient care in the postoperative period could reduce fatalities.  相似文献

During last decades, native uniqueness decreased in local livestock breeds due to the introgression of high‐yielding breeds. Recovery of native uniqueness became important because of conservation aspects regarding native genetic diversity and native traits. Thereby the expectation exists, that the relation between native uniqueness and genetic gain is contradictory. The aim of this study was to explore the influence of native uniqueness on performance traits and the total merit index in a local red cattle breed from Northern Germany. Data contained a pedigree file of 178,255 Red Dual‐Purpose cattle, 809 target genotypes and 3,581 reference genotypes from introgressed breeds. Native genetic contributions were tested for correlation with performance traits of milk yield, longevity, foundation, somatic cells, fertility and maternal calving and the total merit index. The study revealed that native uniqueness is favourably related to longevity (0.16), foundation (0.23), and somatic cells (0.08), and the total merit index (0.10). Selection on native uniqueness could probably lead to an increased longevity, udder health and genetic gain of the Red Dual‐Purpose cattle. Moreover, it was shown that the Red Dual‐Purpose cattle was not upgraded through introgression of high‐yielding breeds.  相似文献

The relative contents of long-chain fatty acids in P. multocida and P. haemolytica were investigated. A dependence on the composition of the broth was established. Accordingly, comparative quantitative studies on fatty acid contents have to be conducted using bacteria grown with the same lot of broth medium. As for P. multocida, there were significant differences between the serovars (C14 in TDHM and C16, delta 2C18 in BPL). These differences are, however, not significant to replace serotyping. Highly significant differences were also detected between P. multocida isolates from nasal swabs and pneumonic lungs (interims of C14, delta C16 on BPL and BRU). The largest differences were measured for strains grown on BRU, which is interpreted as an expression of virulence. Significant differences were found between biotypes A and T of P. haemolytica, namely for C14, C16 in TDHM, and C14, delta C16, C16, C18 in BPL medium.  相似文献
