Purpose

Salt marsh plants are colonising wastes from a steel plant deposited on the Coina River Banks posing a potential contamination risk to the Tagus estuary ecosystem. The objectives of this study were to assess the uptake, accumulation and translocation of hazardous elements/nutrients in three spontaneous halophytic species, to evaluate the capacity of Tamarix africana to stabilise a contaminated salt marsh soil, and to evaluate the ecotoxicity of the pore water and elutriates from phytostabilised soils.

Materials and methods

The work comprises the following: fieldwork collection of soil samples from Coina River (an affluent of Tagus River) bank landfill, estuarine water and spontaneous plants (Aster tripolium, Halimione portulacoides and Sarcocornia sp.), and greenhouse studies (microcosm assay) with T. africana growing in one landfill salt marsh soil, for 97 days, and watered with estuarine water. Soils were analysed for pH, EC, Corganic, NPK, iron and manganese oxides. Soils total (acid digestion) elemental concentrations were determined by ICP/INAA. Estuarine waters, plants roots and shoots (acid digestion), soils available fraction (diluted organic acids extraction-RHIZO or pore water), and salts collected from the T. africana leaves surface were analysed for metals/metalloids (ICP-MS). Ecotoxicity assays were performed in T. africana soil elutriates and pore waters using Artemia franciscana and Brachionus plicatillis.

Results and discussion

Soils were contaminated, containing high total concentrations of arsenic, cadmium, chromium, copper, lead and zinc. However, their concentrations in the available fraction were <4 % of the total. The estuarine waters were contaminated with cadmium, but negligible ecotoxicological effect was observed. The spontaneous plants had significant uptake of the above elements, being mostly stored in the roots. Elemental concentrations in the shoots were within the normal range for plants. These species are not hazardous elements accumulators. Tamarix africana was well adapted to the contaminated saline soils, stored the contaminants in the roots, and had small concentrations of hazardous elements in the shoots. Excretion of hazardous elements by the salt glands was also observed. Elutriates from soils with and without plant did not show ecotoxicity.

Conclusions

The salt marsh species play an important role in the stabilisation of the soils in natural conditions. Tamarix africana showed potential for phytostabilisation of saline-contaminated soils. The low translocation of the elements from roots to shoots and/or active excretion of the elements by the salt glands was a tolerance mechanism in T. africana.

Background

In women with breast cancer submitted to neoadjuvant chemotherapy based in doxorubicin, tumor expression of groups of three genes (PRSS11, MTSS1, CLPTM1 and PRSS11, MTSS1, SMYD2) have classified them as responsive or resistant. We have investigated whether expression of these trios of genes could predict mammary carcinoma response in dogs and whether tumor slices, which maintain epithelial-mesenchymal interactions, could be used to evaluate drug response in vitro.

Methods

Tumors from 38 dogs were sliced and cultured with or without doxorubicin 1 μM for 24 h. Tumor cells were counted by two observers to establish a percentage variation in cell number, between slices. Based on these results, a reduction in cell number between treated and control samples ≥ 21.7%, arbitrarily classified samples, as drug responsive. Tumor expression of PRSS11, MTSS1, CLPTM1 and SMYD2, was evaluated by real time PCR. Relative expression results were then transformed to their natural logarithm values, which were spatially disposed according to the expression of trios of genes, comprising PRSS11, MTSS1, CLPTM1 and PRSS11, MTSS1, SMYD2. Fisher linear discrimination test was used to generate a separation plane between responsive and non-responsive tumors.

Results

Culture of tumor slices for 24 h was feasible. Nine samples were considered responsive and 29 non-responsive to doxorubicin, considering the pre-established cut-off value of cell number reduction ≥ 21.7%, between doxorubicin treated and control samples. Relative gene expression was evaluated and tumor samples were then spatially distributed according to the expression of the trios of genes: PRSS11, MTSS1, CLPTM1 and PRSS11, MTSS1, SMYD2. A separation plane was generated. However, no clear separation between responsive and non-responsive samples could be observed.

Conclusion

Three-dimensional distribution of samples according to the expression of the trios of genes PRSS11, MTSS1, CLPTM1 and PRSS11, MTSS1, SMYD2 could not predict doxorubicin in vitro responsiveness. Short term culture of mammary gland cancer slices may be an interesting model to evaluate chemotherapy activity.  相似文献

A 12-year-old Rottweiler dog was presented with a history of prostration, weight loss and hyporexia for six months. Based on complete blood tests (hematological and biochemical analyses), bone marrow examination and imaging analysis, a diagnosis of chronic myeloid leukemia was made. Treatment with hydroxyurea at a dosage of 18?mg/kg twice daily was not effective in controlling the high count of white blood cells. Furthermore, after 35 days of hydroxyurea treatment, the animal developed onycholysis, with sloughing of the claws of the left pelvic and left thoracic limbs and exposure of the distal phalanx. Interruption of the medication was implemented, with clinical healing of the ungual lesions observed three months after initiation of the drug. White blood cells returned to normal after using cyclophosphamide. Currently, the animal is in complete remission, having a disease-free interval of 575 days without chemotherapy. To the authors' knowledge, this is the first report of hydroxyurea-induced onycholysis within a short-term period in a dog diagnosed with chronic myeloid leukemia.  相似文献

A microsatellite DNA-based method for Vitis vinifera grape must authentication is presented. Five of the most important port wine producing grape cultivars (Tinta Roriz, Tinto C?o, Touriga Francesa, Touriga Nacional, and Tinta Barroca) were typed at four microsatellite loci described by Bowers et al. (Genome 1996, 39, 628-633) and Thomas and Scott (Theor. Appl. Genet. 1993, 86, 985-990). The corresponding 5 varietal musts and 26 must mixtures that result from the combination of the five varieties were also typed at the four loci. There were no differences between the corresponding leaf and varietal must profiles. All must combinations showed the expected band profiles corresponding to the sum of the varietal band profile components. The Passiflora L. (Passifloraceae) genus consists of about 500 species widely distributed throughout tropical and subtropical regions, commonly known as the passion flower. The passion flowers are considered ornamental plants because of the exotic beauty of their flowers and variant foliage. Since their introduction to the Old World, around 1625, seventeenth century, they have been used to decorate European glasshouses and gardens. Interspecific hybridization is used to produce single ornamental plants. However in some countries, such as Brazil, being centre of origin of many species, the potential value of these species has hardly been exploited. The beauty of these wild species and their hybrids is little known in Brazil. The Eucalyptus pathogen Quambalaria eucalypti has been reported from several subtropical and tropical countries of the Southern Hemisphere. During the course of a nationwide monitoring programme aimed at the detection of pathogens in commercial eucalypt plantations in Portugal, Q. eucalypti was found affecting Eucalyptus globulus. The identity of the pathogen was confirmed by sequence analyses of the internal transcribed spacer (ITS) region of the rDNA operon, as well as morphological characteristics. Quambalaria eucalypti is widespread throughout the eucalypt‐producing areas of Portugal. Pathogenicity studies revealed that different host genotypes showed various degrees of susceptibility to the pathogen. To our knowledge, this is the first report of Q. eucalypti on Eucalyptus spp. in Portugal and in the Northern Hemisphere.  相似文献

This paper characterizes wood and charcoal made from Hymenaea aurea, Mimosa scabrella, Tabebuia capitata and Eucalyptus alba by scanning electron microscopy and near infrared for the purpose of developing a technique to identify the source of charcoal samples. Ten test samples were prepared for each species, oriented in transverse, radial and tangential section. Samples were carbonized in a muffle furnace with a final temperature of 450 °C and a heating rate of 1.66 °C/min. The microscopic structures of the wood and charcoal samples were verified by scanning electron microscopy, and spectra were obtained with a spectrophotometer, equipped with an integrating sphere and operating in transmittance mode. The spectra of the wood samples present characteristic bands of lignocellulosic material, while the spectra of the charcoal samples show small or nil absorption in the near-infrared region. The near-infrared spectroscopy technique was efficient in discriminating the four species studied.  相似文献
