Background

Heifers not used as breeding stock are often implanted with steroids to increase growth efficiency thereby altering hormone profiles and potentially changing the environment in which ovarian follicles develop. Because bovine granulosa cell culture is a commonly used technique and often bovine ovaries are collected from abattoirs with no record of implant status, the objective of this study was to determine if the presence of an implant during bovine granulosa cell development impacts follicle stimulating hormone-regulated steroidogenic enzyme expression. Paired ovaries were collected from 16 feedlot heifers subjected to 1 of 3 treatments: non-implanted (n = 5), Revalor 200 for 28 d (n = 5), or Revalor 200 for 84 d (n = 6). Small follicle (1 to 5 mm) granulosa cells were isolated from each pair and incubated with phosphate buffered saline (n = 16) or 100 ng/mL follicle stimulating hormone (n = 16) for 24 h.

Results

Granulosa cells of implanted heifers treated with follicle stimulating hormone produced medium concentrations of progesterone similar (P = 0.22) to non-implanted heifers, while medium estradiol concentrations were increased (P < 0.10) at 28 and 84 d compared to non-implanted heifers indicating efficacy of treatment. Additionally, real-time PCR analysis in response to follicle stimulating hormone treatment demonstrated a decrease in steroidogenic acute regulatory protein (P = 0.05) mRNA expression in heifers implanted for 84 d and an increase in P450 side chain cleavage mRNA in granulosa cells of heifers implanted for 28 (P < 0.10) or 84 d (P < 0.05) compared to non-implanted females. However, no difference in expression of 3-beta-hydroxysteroid dehydrogenase (P = 0.57) and aromatase (P = 0.23) were demonstrated in implanted or non-implanted heifers.

Conclusions

These results indicate follicles which develop in the presence of high concentrations of androgenic and estrogenic steroids via an implant tend to demonstrate an altered capacity to respond to follicle stimulating hormone stimulation. Thus, efforts should be made to avoid the use of implanted heifers to study steroidogenesis in small follicle granulosa cell culture systems.  相似文献

Caloric restriction (CR) has been shown to retard immunosenescence and to extend median and maximum life span in rodent species. Longitudinal effects of CR on the canine immune system are presented in this report. A group of 48 Labrador Retrievers, divided at weaning into weight- and sex-matched pairs, were maintained on a diet restriction protocol from age 8 weeks until death. Each restricted dog received 75% of the total food consumed by its control-fed pair mate. Immune parameters were monitored from 4 to 13 years. CR retarded age-related declines in both lymphoproliferative responses and absolute numbers of lymphocytes and the T, CD4, and CD8-cell subsets. In females, CR attenuated the age-related increase in T-cell percentages and marginally retarded the age-related increase in memory cell percentages. Age-related changes in B-cell percentages and numbers were augmented by CR. No direct effect of CR on phagocytic activity of PMN, antibody production or NK cell activity, was observed. Lower lymphoproliferative responses, lower numbers of lymphocytes, T, CD4 and CD8 cells, lower CD8 percentages and higher B-cell percentages were all found to be significantly associated with a decreased likelihood of survival in these dogs.  相似文献

This study investigated how urban cultural and economic tolerance affects urban innovative capacities based on China's prefecture‐level cities. Several tolerance indices, including ratios of migrants, rental housing, gay people, and private economies were introduced and the cities’ tolerance scores were measured using factor analysis. The results show that cities with higher cultural and economical tolerant scores were agglomerated in three metropolitan areas in China's southeastern coastal region. The spatial regression model demonstrates that urban cultural and economic tolerance increases urban innovation output and promotes innovation capacities. We also introduced the crime rate as an instrumental variable and found that the effect of tolerance on innovation remains robust. Our study suggests it is important for the Chinese government to establish an open and tolerant environment to attract migrants, creative artists, and entrepreneurs to foster urban vitality and improve urban innovative capacities.  相似文献

Genome-wide association studies (GWAS) are useful to facilitate crop improvement via enhanced knowledge of marker-trait associations (MTA). A GWAS for grain yield (GY), yield components, and agronomic traits was conducted using a diverse panel of 239 soft red winter wheat (Triticum aestivum) genotypes evaluated across two growing seasons and eight site-years. Analysis of variance showed significant environment, genotype, and genotype-by-environment effects for GY and yield components. Narrow sense heritability of GY (h 2  = 0.48) was moderate compared to other traits including plant height (h 2  = 0.81) and kernel weight (h 2  = 0.77). There were 112 significant MTA (p < 0.0005) detected for eight measured traits using compressed mixed linear models and 5715 single nucleotide polymorphism markers. MTA for GY and agronomic traits coincided with previously reported QTL for winter and spring wheat. Highly significant MTA for GY showed an overall negative allelic effect for the minor allele, indicating selection against these alleles by breeders. Markers associated with multiple traits observed on chromosomes 1A, 2D, 3B, and 4B with positive minor effects serve as potential targets for marker assisted breeding to select for improvement of GY and related traits. Following marker validation, these multi-trait loci have the potential to be utilized for MAS to improve GY and adaptation of soft red winter wheat.  相似文献

China has taken a foreign direct investment‐based approach toward increasing its capital and knowledge base, and developing into an innovative economy. However, little quantitative evidence exists about the factors that drive innovations of foreign‐invested enterprises (FIEs) there. This paper uses survey data from high‐technology firms in Shanghai to discuss factors affecting their innovativeness. It takes the concepts of absorptive capacity, export orientation, and innovation‐related cooperation as a starting point. It highlights how the interplay of strategies and resources affects innovativeness and heterogeneity of FIEs. The most innovative FIEs are endowed with a strong human capital base and R&D activities, which at the same time target export markets and whose cooperative partners involve firms other than their parent company. The results underline the necessity to differentiate between the different types of FIEs when examining their innovativeness.  相似文献

Bitter melon (Momordica Charantia L.) is an important vegetable crop with nutritional and medicinal qualities. As a member of cucurbitaceae it is monoecious with varying proportions of staminate and pistillate flowers. This study was undertaken to determine the effects of various applications of ethrel and gibberellic acid (GA3) on sex modification in M. charantia. In the first set of experiments, various concentrations of hormones were added to the seed germination medium, in the second, adult plants growing in the field were sprayed with aqueous solutions of ethrel or GA3 three times at three-day intervals. The number and sex of open flowers was recorded daily for 60 days after the first flower opened and total number of staminate and pistillate flowers was calculated at the end of the experiment. The highest frequency (29.5%) of pistillate flowers was observed in plants treated with 500 ppm ethrel at germination. Similarly, spraying of adult plants with 100 ppm GA3 increased the proportion of pistillate flowers to 26% relative to 15% in untreated controls. Both ethrel and GA3 induced significantly higher number of pistillate flowers than control. In vitro hormone application during seed germination was much more successful than spraying of field grown plants.  相似文献

Farmers in the Broadview Water District in central Californiahave been improving irrigation practices in response to risingirrigation water prices and reductions in water supply since1989, when incentive policies were first implemented to reducethe volume of subsurface drain water generated in theDistrict. The average salinity of water deliveries hasincreased, over time, as the District has recycled largeamounts of drainage water to achieve regional restrictions ondrainage water discharge. We review irrigation and drainageactivities in Broadview since 1986 with an emphasis on thesustainability of crop production when drainage discharge islimited. Average cotton yields in Broadview have declined inrecent years, both nominally and in comparison with averageyields reported for the large county in which Broadview islocated. Average tomato yields in Broadview have increased inrecent years, but county-wide yields have exceeded Broadviewyields with greater frequency than in the late 1980s. Theseobservations suggest that average crop yields in Broadview maybe starting to reflect the increasing salinity of soil andwater resources, which may be due in part to persistentrestrictions on drainage water discharge.  相似文献

A method for determining ethyl coumarate and ethyl ferulate in wine using GC-MS with deuterium-labeled analogues has been developed and used to measure the evolution of these two esters during the production of two commercial monovarietal red wines, cv. Grenache and Shiraz. During fermentation, the concentration of ethyl coumarate rose from low levels to 0.4 mg/L in Grenache and 1.6 mg/L in Shiraz wines. These concentrations then increased further during barrel aging to 1.4 and 3.6 mg/L, respectively. The concentration of ethyl ferulate was much lower, reaching a maximum of only 0.09 mg/L. Conversion of ethyl coumarate and ethyl ferulate to their corresponding ethylphenols was observed during fermentations of a synthetic medium with two strains of Dekkera bruxellensis (AWRI 1499 and AWRI 1608), while a third (strain AWRI 1613) produced no ethylphenols at all from these precursors. Strains AWRI 1499 and 1608 produced 4-ethylphenol from ethyl coumarate in 68% and 57% yields, respectively. The corresponding yields of 4-ethylguaiacol from ethyl ferulate were much lower, 7% and 3%. Monitoring of ethyl coumarate and ethyl ferulate concentration during the Dekkera fermentations showed that the selectivity for ethylphenol production according to yeast strain and the precursor was principally a result of variation in esterase activity. Consequently, ethyl coumarate can be considered to be a significant precursor to 4-ethylphenol in wines affected by these two strains of Brettanomyces/Dekkera yeast, while ethyl ferulate is not an important precursor to 4-ethylguaiacol.  相似文献
