The effects of elevated concentrations of atmospheric CO2 (e[CO2]) on the nutritive value of wheat vegetative matter and grain as a feedstock for ruminants were investigated in a study undertaken at the Australian grains free‐air CO2 enrichment (AGFACE) facility. The study included two commercial wheat cultivars (Janz and Yitpi) and two genetic selections from a Seri/Babex population (SB003 and SB062) which had previously been characterised for low and high water‐soluble carbohydrate accumulation efficiency. The trial was grown under ambient (~390 µmol/mol) and elevated (~550 µmol/mol) CO2 conditions, and plants harvested at tillering, anthesis and physiological maturity. Composition analyses to determine the nutritive value for ruminant feed were undertaken on stems, leaves and grain. Plant and grain nitrogen were reduced in the e[CO2] treatments, and as expected, the water‐soluble carbohydrates increased. All genotypes responded to e[CO2] with the effects of altered composition evident within 60 days of sowing. Determinants of ruminant feed quality such as neutral and acid detergent fibre and estimated in vitro metabolisable energy were not significantly affected. The reduced plant and grain N will impact on the nutritive value and supplementation may be required. The impact of e[CO2] on chemical composition of wheat plants may be greater if the predicted climate change is associated with concomitant abiotic stress such as high ambient temperature or low soil moisture.  相似文献

Summary The soluble proteins from two near-isogenic lines of Pisum sativum. cv. William Massey have been compared electrophoretically. These lines differ in their physiological response to wilt (Fusarium oxysporum f. sp. pisi), one being susceptible and the other resistant. The total protein profiles derived from the two lines appear to be identical. The resistant line differs electrophoretically from the susceptible line in carrying an esterase component which has been derived from its resistant parent, cultivar Delwiche Commando.  相似文献

Virally encoded microRNAs (miRNAs) have recently been discovered in herpesviruses. However, their biological roles are mostly unknown. We developed an algorithm for the prediction of miRNA targets and applied it to human cytomegalovirus miRNAs, resulting in the identification of the major histocompatibility complex class I-related chain B (MICB) gene as a top candidate target of hcmv-miR-UL112. MICB is a stress-induced ligand of the natural killer (NK) cell activating receptor NKG2D and is critical for the NK cell killing of virus-infected cells and tumor cells. We show that hcmv-miR-UL112 specifically down-regulates MICB expression during viral infection, leading to decreased binding of NKG2D and reduced killing by NK cells. Our results reveal a miRNA-based immunoevasion mechanism that appears to be exploited by human cytomegalovirus.  相似文献

Crop coefficients are a widely used and universally accepted method for estimating the crop evapotranspiration (ETc) component in irrigation scheduling programs. However, uncertainties of generalized basal crop coefficient (Kcb) curves can contribute to ETc estimates that are substantially different from actual ETc. Limited research with corn has shown improvements to irrigation scheduling due to better water-use estimation and more appropriate timing of irrigations when Kcb estimates derived from remotely sensed multispectral vegetation indices (VIs) were incorporated into irrigation-scheduling algorithms. The purpose of this article was to develop and evaluate a Kcb estimation model based on observations of the normalized difference vegetation index (NDVI) for a full-season cotton grown in the desert southwestern USA. The Kcb data used in developing the relationship with NDVI were derived from back-calculations of the FAO-56 dual crop coefficient procedures using field data obtained during two cotton experiments conducted during 1990 and 1991 at a site in central Arizona. The estimation model consisted of two regression relations: a linear function of Kcb versus NDVI (r2=0.97, n=68) used to estimate Kcb from early vegetative growth to effective full cover, and a multiple regression of Kcb as a function of NDVI and cumulative growing-degree-days (GDD) (r2=0.82, n=64) used to estimate Kcb after effective full cover was attained. The NDVI for cotton at effective full cover was ~0.80; this value was used to mark the point at which the model transferred from the linear to the multiple regression function. An initial evaluation of the performance of the model was made by incorporating Kcb estimates, based on NDVI measurements and the developed regression functions, within the FAO-56 dual procedures and comparing the estimated ETc with field observations from two cotton plots collected during an experiment in central Arizona in 1998. Preliminary results indicate that the ETc based on the NDVI-Kcb model provided close estimates of actual ETc.Communicated by R. Evans  相似文献

ABSTRACT Puccinia spp. are widespread pathogens of cereals and grasses that annually cause significant yield losses worldwide, especially in barley, oat, and wheat. Urediniospore morphology and early symptom development have limited usefulness for distinguishing Puccinia spp. Therefore, we developed real-time polymerase chain reaction assays for rapid detection of the four rust pathogen species, Puccinia graminis (Pers.:Pers.), P. striiformis (Westend.), P. triticina (Eriks.), and P. recondita (Roberge ex Desmaz.). Duplex assays were constructed for the nuclear rDNA gene, using the variable internal transcribed spacer 1 (ITS1) region to distinguish between species, and the conserved 28S region as an internal control. Species-specific ITS1 primer/probe sets were highly specific and could detect <1 pg of DNA. The species-specific primer/probe sets showed positive results over a linear range of DNA five orders of magnitude or greater. Specificity of the assays was tested using multiple collections representing a range of races and formae speciales within a species. Additionally, assay specificity was evaluated by testing a range of other grass rust pathogens, as well as other fungi. The 28S primer/probe combination was successful in detecting all Puccinia spp. tested within the duplex assays, validating the integrity of each assay. Finally, the assays were used to identify unknown rust fungi infecting pasture grasses.  相似文献

The speed and sensitivity of PCR-based assays allow shorter turnaround times for the detection of pathogens for which culture and serological methods are difficult or unavailable. PCR was performed with primer sets Cms50 and Cms72, designed previously by Millset al. (1997) through subtractive hybridization to detectClavibacter michiganensis subspeciessepedonicus (Cms). In bacterial suspensions, fewer than three cells/10 ul reaction were detected after PCR amplicons were hybridized with specific DIG-labeled DNA probes in an enzyme-linked oligonucleosorbent assay (ELOSA). In naturally infected tuber samples representing three cultivars of potato, the diagnostic sensitivity of PCR/ ELOSA was 96%, while the specificity exceeded 99%. PCR/ELOSA detectedCms in infected tuber samples with equal sensitivity regardless of colony morphology, potato cultivar, or primer sets.  相似文献

To answer the question of whether horse height, cannon bone circumference, and loin width can be used as indicators of weight-carrying ability in light horses, eight mature horses underwent a submaximal mounted standard exercise test under four conditions: carrying 15, 20, 25, or 30% of their body weight. Heart rate was monitored, plasma lactate concentration was determined in jugular blood samples pre-exercise, immediately post-exercise, and 10 minutes post-exercise, with serum creatine kinase activity determined at the same times as plasma lactate concentrations, with additional samples collected at 24 hours and 48 hours post-exercise. Muscle soreness and muscle tightness scores were determined using a subjective scoring system 24 hours before and 24 hours after exercise. Heart rates remained significantly higher when the horses carried 25 and 30% of their body weight. Plasma lactate concentrations immediately and 10 minutes after exercise differed when horses carried 30% of their body weight compared with 15, 20, and 25% weight carriage. Horses tended to have a greater change in muscle soreness and muscle tightness when carrying 25% of their body weight, and a significant change in soreness and tightness scores was found in horses carrying 30% of their body weight. Loin width and cannon bone circumference were found to be negatively correlated to the changes in muscle soreness and tightness scores. In conclusion, the data suggest that horses with wider loin and thicker cannon bone circumference became less sore when carrying heavier weight loads.  相似文献

A miniature pinscher-cross was evaluated for chronic coughing. Computed tomography and bronchoscopy revealed severe, diffuse, cylindrical bronchiectasis secondary to eosinophilic bronchopneumopathy. Computed tomography is the gold standard for diagnosis of bronchiectasis in humans, and should be further investigated in dogs as a means of characterizing severity and pattern of disease.  相似文献
