The feasibility of global polio eradication is being questioned as a result of continued transmission in a few localities that act as sources for outbreaks elsewhere. Perhaps the greatest challenge is in India, where transmission has persisted in Uttar Pradesh and Bihar despite high coverage with multiple doses of vaccine. We estimate key parameters governing the seasonal epidemics in these areas and show that high population density and poor sanitation cause persistence by not only facilitating transmission of poliovirus but also severely compromising the efficacy of the trivalent vaccine. We analyze strategies to counteract this and show that switching to monovalent vaccine may finally interrupt virus transmission. 相似文献

Low-mass stars, approximately 1 to 2 solar masses, near the Main Sequence are efficient at producing the helium isotope 3He, which they mix into the convective envelope on the giant branch and should distribute into the Galaxy by way of envelope loss. This process is so efficient that it is difficult to reconcile the low observed cosmic abundance of 3He with the predictions of both stellar and Big Bang nucleosynthesis. Here we find, by modeling a red giant with a fully three-dimensional hydrodynamic code and a full nucleosynthetic network, that mixing arises in the supposedly stable and radiative zone between the hydrogen-burning shell and the base of the convective envelope. This mixing is due to Rayleigh-Taylor instability within a zone just above the hydrogen-burning shell, where a nuclear reaction lowers the mean molecular weight slightly. Thus, we are able to remove the threat that 3He production in low-mass stars poses to the Big Bang nucleosynthesis of 3He. 相似文献

The ataxia telangiectasia mutated (ATM) protein kinase is a critical component of a DNA-damage response network configured to maintain genomic integrity. The abundance of an essential downstream effecter of this pathway, the tumor suppressor protein p53, is tightly regulated by controlled degradation through COP1 and other E3 ubiquitin ligases, such as MDM2 and Pirh2; however, the signal transduction pathway that regulates the COP1-p53 axis following DNA damage remains enigmatic. We observed that in response to DNA damage, ATM phosphorylated COP1 on Ser(387) and stimulated a rapid autodegradation mechanism. Ionizing radiation triggered an ATM-dependent movement of COP1 from the nucleus to the cytoplasm, and ATM-dependent phosphorylation of COP1 on Ser(387) was both necessary and sufficient to disrupt the COP1-p53 complex and subsequently to abrogate the ubiquitination and degradation of p53. Furthermore, phosphorylation of COP1 on Ser(387) was required to permit p53 to become stabilized and to exert its tumor suppressor properties in response to DNA damage. 相似文献

The Hawaiian-Emperor bend has played a prominent yet controversial role in deciphering past Pacific plate motions and the tempo of plate motion change. New ages for volcanoes of the central and southern Emperor chain define large changes in volcanic migration rate with little associated change in the chain's trend, which suggests that the bend did not form by slowing of the Hawaiian hot spot. Initiation of the bend near Kimmei seamount about 50 million years ago (MA) was coincident with realignment of Pacific spreading centers and early magmatism in western Pacific arcs, consistent with formation of the bend by changed Pacific plate motion. 相似文献

In developing embryos of some extant spiralian animals, polar lobe formation is one of the symmetry-breaking mechanisms for segregation of maternal cytoplasmic substances to certain blastomeres and not others. Polar lobe formation leads to unique early cleavage morphologies that include trilobed, J-shaped, and five-lobed structures. Fossil embryos similar to modern lobeforming embryos are recognized from the Precambrian Doushantuo Formation phosphates, Weng'an, Guizhou Province, China. These embryos are abundant and form a developmental sequence comparable to different developing stages observed in lobe-forming embryos of extant spiralians. These data imply that lobe formation is an evolutionarily ancient process of embryonic specification. 相似文献

The orb web is a spectacular evolutionary innovation that enables spiders to catch flying prey. This elegant, geometric structure is woven with silk fibers that are renowned for their superior mechanical properties. We used silk gland expression libraries to address a long-standing controversy concerning the evolution of the orb-web architecture. Contrary to the view that the orb-web design evolved multiple times, we found that the distribution and phylogeny of silk proteins support a single, ancient origin of the orb web at least 136 million years ago. Furthermore, we substantially expanded the repository of silk sequences that can be used for the synthesis of high-performance biomaterials. 相似文献

- 1. SECORE (SExual COral REproduction) Project is an initiative of public aquariums and research institutions to produce and exchange sexual coral recruits for the sustainable management of ex situ populations. Here we present the results of the initial three years (2002–2004).
- 2. Primary polyps (n=501) of corals (Acropora tenuis, Agaricia humilis, Favia fragum) were transported from Rotterdam Zoo to Cologne, Burgers', Hagenbeck and London Zoos, where development of juveniles was monitored for 10 months. All polyps were produced at Rotterdam Zoo from laboratory colonies (A. humilis, F. fragum), and from larvae generated from field collected gametes at Akajima, Okinawa, Japan (A. tenuis). Additionally, planulae of A. tenuis (n=1440) were transported from Rotterdam Zoo to Burgers' Zoo and to London Zoo to obtain primary polyps.
- 3. Larval settlement (A. tenuis) was observed to be 3.00 ± 2.57% (mean ± SD; n=1480) in 2002 and 17.36 ± 6.01% (mean ± SD; n=1480) in 2003, significantly lower compared to settlement at Rotterdam Zoo (57.84 ± 11.01% in 2003; mean ± SD, n=1480). High post‐transport survival rates of 95.18 ± 4.86% (mean ± SD; n=501) were observed in primary polyps of all species.
- 4. Juvenile survival (t=10 months; A. tenuis: 18.4–86.2%; A. humilis: 0–19.7%; F. fragum: 13.3–72.7%) differed significantly between institutions. Mean colony sizes (measured 10 months after transportation) were, in all cases, similar or higher to those reported from literature.
- 5. The results demonstrate the potential of this method to serve as an economical and sustainable alternative to existing mostly exploitative techniques for aquarium stocking. The use of sexual recruits provides an effective and low cost alternative, which is, in principle, applicable to all coral species.
- 6. The project was extended from 9 to 28 institutions across Europe, the USA and Japan in 2004.

OBJECTIVE: To determine an optimal dose of carbon 13 ((13)C)-labeled aminopyrine for use in a (13)C-aminopyrine demethylation blood test in healthy dogs. ANIMALS: 9 adult dogs. PROCEDURES: Food was withheld from each dog for 12 hours. A 2-mL baseline blood sample was obtained from each dog and placed into an evacuated tube containing sodium heparin. Carbon 13-labeled aminopyrine was administered IV at doses of 1, 2, 5, or 10 mg/kg. Additional blood samples (2 mL) were obtained and placed into evacuated tubes containing sodium heparin 30, 45, 60, and 75 minutes after (13)C-aminopyrine administration. Hydrochloric acid was used to extract CO(2) from blood samples. The extracted gas was analyzed by fractional mass spectrometry to determine the percentage dose of (13)C administered as (13)C-aminopyrine and recovered in extracted gas (PCD). RESULTS: Gross evidence of clinical adverse effects was not detected in any dog after administration of (13)C-aminopyrine. The mean coefficient of variation (CV) for PCD was significantly lower than the mean CV for the summation of PCD values up to a given sampling time (CUMPCD). Mean PCD values among the 4 doses for each sample time were not significantly different. Administration of (13)C-aminopyrine at a dose of 2 mg/kg resulted in the lowest interindividual variability. CONCLUSIONS AND CLINICAL RELEVANCE: The PCD is superior to CUMPCD for the quantification of aminopyrine demethylation. Administration of (13)C-(13)C-aminopyrine at a dose of 2 mg/kg is appropriate for use in the (13)C-aminopyrine demethylation blood test in healthy dogs. 相似文献

OBJECTIVE: To evaluate the efficacy and safety of intra-articular administration of ethyl alcohol for arthrodesis of tarsometatarsal joints in horses. ANIMALS: 8 healthy female horses without lameness or radiographic evidence of tarsal joint osteoarthritis. PROCEDURE: In each horse, 1 tarsometatarsal joint was treated with 4 mL of 70% ethyl alcohol and the opposite joint was treated with 4 mL of 95% ethyl alcohol. Lameness examinations were performed daily for 2 weeks, followed by monthly evaluations for the duration of the 12-month study. Radiographic evaluations of both tarsi were performed 1 month after injection and every 3 months thereafter. Gross and histologic examinations of the tarsi were undertaken at completion of the study. RESULTS: Horses had minimal to no lameness associated with the treatments. Radiography revealed that 8 of 16 joints were fused by 4 months after treatment, with significantly more joints fused in the 70% ethyl alcohol group. Fifteen of 16 joints were considered fused at postmortem examination at 12 months. Gross and histologic examinations revealed foci of dense mature osteonal bone spanning the joint spaces. Bony fusion appeared to be concentrated on the dorsolateral, centrolateral, and plantarolateral aspects of the joints. Significant differences were not detected between treatment groups for lameness or pathologic findings. CONCLUSIONS AND CLINICAL RELEVANCE: Administration of ethyl alcohol into the tarsometatarsal joint of healthy horses appeared to facilitate arthrodesis of the joint in a pain-free manner. Results warrant further investigation into the potential use of ethyl alcohol in horses clinically affected with osteoarthritis of the tarsometatarsal and distal intertarsal joints. 相似文献
