The effect of different polishing techniques on loss of mineral elements from rice grains was quantified using a panel of indica and tropical japonica genotypes, previously classified as differing in ease of polishing. Gradients in mineral elements across the bran-endosperm interface were quantified using micro-scaled precision abrasive polishing in combination with inductively coupled plasma mass spectrometry and synchrotron X-ray fluorescence microscopy. Frictional polishing, similar to that of commercial mills, i.e. 8–10% loss of grain weight, reduced the concentration of Fe, Mg, P, K and Mn by 60–80% in all genotypes. Following gentler polishing (3–5% weight loss), genotypes classified as difficult to polish showed smaller decreases in Fe, Mg, P, K and Mn compared to genotypes classified as easy to polish. The concentration of other elements, e.g. Zn, S, Ca, Cu, Mo and Cd, showed comparable reductions (<30%) irrespective of polishing technique or ease of polishing. The different patterns of polishing losses of minerals reflected their distribution within the grain. Five-fold differences in the reduction of Zn concentration during polishing were observed for different genotypes which started with similar Zn concentrations in the unpolished grain, thus showing clear potential for selecting genotypes with reduced polishing losses of Zn. 相似文献

Marine natural products are a diverse, unique collection of compounds with immense therapeutic potential. This has resulted in these molecules being evaluated for a number of different disease indications including the neglected protozoan diseases, human African trypanosomiasis and Chagas disease, for which very few drugs are currently available. This article will review the marine natural products for which activity against the kinetoplastid parasites; Trypanosoma brucei brucei, T.b. rhodesiense and T. cruzi has been reported. As it is important to know the selectivity of a compound when evaluating its trypanocidal activity, this article will only cover molecules which have simultaneously been tested for cytotoxicity against a mammalian cell line. All available published statistics on the exploitation of the Amazonian manatee Trichechus inunguis are collected and analysed to determine the major trends in exploitation and, if possible, their effects on the species. The manatee's role in the Amazonian economy has traiditionally been on the level of subsistnce hunting for local consumption; export of surplus manatee products has been limited and sporadic, and keeping of records has been even more so. Unknown quantities of manatee meat were shipped to the Guianas and West Indies in the 17th century, At least from the 1780s to about 1925, the only manatee product common in Amazonian commerce was mixira (fried meat packed in lard). From 1935 to 1954, manatee hides were exported to southern Brazil and elsewhere for the manufacture of heavy-duty leather. From 1954 to the legal banning of manatee hunting in 1973, manatees were again heavily exploited for meat. At no time did manatee products make up more than a small fraction of a percent of the Amazon region's trade. Numbers of animals killed per year cannot be determined with any precision, but early statistics on mixira reflect no more than 1000/year, while the peak years of hide and meat production probably required 4000–7000 manatees/year, exclusive of unrecorded subsistence hunting. Taking the latter into account, it is likely that actual mortality ran to several thousand animals per year throughout the past two centuries. Decline of meat production in the 1960s may reflect overexploitation. Evidence was also found of some exploitation for meat of West Indian manatees T. manatus in the states of Alagoas and Bahia, Brazil, the latter constituting the southernmost record of the species since the early 19th century. 相似文献

The effects of different tank colours (white, yellow sandy and black) on the growth, mortality and biomass production were studied for hatchling and early juvenile cuttlefish. For hatchlings, the use of different colour tanks did not promote differences in growth due to the higher variability (standard deviation) found in the white‐ and sand‐coloured tanks. Black tanks promoted the lowest and highest values for total mortality and biomass respectively. For juveniles, the use of different tank colours promoted different growth (P<0.05), but not mortality. Black tanks promoted the best results in terms of growth and biomass. The results obtained in the present study advise the use of black (or dark colour) tanks in the hatchling and early juvenile stages to reduce the standard deviations associated with growth, mortality and biomass production. The life cycle of cuttlefish fed ad libitum exclusively on live grass shrimp (Palaemonetes varians) was studied during 5 consecutive generations. Different culture temperatures promoted different (P < 0.05) exponential growth for each life cycle, being summer generations shorter than those of winter. Higher temperatures promoted higher IGR’s and mortality, while lower temperatures promoted increased life span, reproduction stages, total fecundity and total egg biomass. Increased generations also seemed to increase fertility. A “hybrid” generation promoted the best results in terms of hatchling weight, individual fecundity and fertility. Mean egg weight was related to female size and embryonic development took longer at lower temperatures. Brood stock sex ratios seemed to be temperature related. All of these culture aspects were also compared between themselves in order to establish future brood stock methodologies. Grass shrimp proved to be a good diet for the culture of cuttlefish throughout the life cycle. The use of only one species reduces costs and labor associated to cuttlefish culture. 相似文献

Individual growth rates, feeding rates (%BWd?1) and food conversions for cuttlefish (S. officinalis) hatchlings and juveniles were determined during this study. A flow‐through system was used. Water temperature reached 30 °C during the hottest part of the day, gradually decreasing to 25 °C during the night; salinity varied between 37 ± 3 ppt and lights were kept on for 14 h day?1. Hatchlings were placed in separate compartments with a water volume of 1.2 L. Juvenile cuttlefish (from 0.5 to 25 g) were placed in bigger baskets, with a water volume of 5.2 L. Water flow was 120 L h?1. The biggest cuttlefish used in these experiments (> 25 g) were gathered in groups of five and placed in circular tanks (water volume of 250–300 L). Thus, results obtained in this case are means and not individual data. During the first 10, 20, 30 and 40 days, mean growth rates (of all individuals sampled by age group) decreased consistently (11.8 ± 4.1, 9.8 ± 1.8, 8.1 ± 2.2 and 7.3 ± 0.7%BW?1 respectively); in similar fashion, mean feeding rates decreased with age group (33.7 ± 13.5, 22.0 ± 7.9, 17.3 ± 3.9 and 16.7%BWd?1 respectively). Mean food conversions varied between 3.6 and 2.5 between the age groups. When grouping results by weight class, similar patterns occur, as growth and feeding rates decrease consistently as cuttlefish grow bigger. Highest mean growth and feeding rates are obtained by hatchlings (< 0.1 g) with 12.4 ± 4.5 and 35.3 ± 15.1%BWd?1, respectively, while the lowest growth and feeding rates were recorded for the largest animals, between 15 and 25 g (3.4 ± 1.1 and 10.8 ± 4.1%BWd?1 respectively). The objective of the current study was to determine the sensitivity and specificity of real-time polymerase chain reaction (real-time PCR) for feline hemoplasmas when applied to DNA extracted from dried whole-blood smears in comparison to that for DNA extracted from liquid whole blood. Blood samples were collected into ethylenediamine tetra-acetic acid tubes from 305 cats with possible or suspected hemoplasmosis, and dried blood smears from each sample were prepared. DNA was extracted from blood smears and a 160-microl aliquot of each liquid blood sample by using a robotic extractor and was subjected to real-time PCR for feline glyceraldehyde-3-phosphate dehydrogenase (liquid blood), 18S ribosomal RNA (dried blood), and "Candidatus Mycoplasma haemominutum", Mycoplasma haemofelis, and "Candidatus Mycoplasma turicensis" DNA. When using the results for liquid whole blood as the gold standard, the sensitivity of each assay for "Ca. M. haemominutum", M. haemofelis, and "Ca. M. turicensis" was 49 of 66 (74%), 11 of 13 (85%), and 11 of 20 (55%), respectively. The specificity of each assay was 224 of 234 (96%), 287 of 287 (100%), and 280 of 280 (100%), respectively. When possible, liquid blood samples should be submitted for detection of feline hemoplasmas by using real-time PCR. The improved sensitivity of real-time PCR on blood smears for M. haemofelis compared with that of the other hemoplasma species may reflect the higher organism burdens associated with infection with this species. 相似文献

As the staple food for 35% of the world's population, wheat is one of the most important crop species. To date, sequence-based tools to accelerate wheat improvement are lacking. As part of the international effort to sequence the 17-billion-base-pair hexaploid bread wheat genome (2n = 6x = 42 chromosomes), we constructed a bacterial artificial chromosome (BAC)-based integrated physical map of the largest chromosome, 3B, that alone is 995 megabases. A chromosome-specific BAC library was used to assemble 82% of the chromosome into 1036 contigs that were anchored with 1443 molecular markers, providing a major resource for genetic and genomic studies. This physical map establishes a template for the remaining wheat chromosomes and demonstrates the feasibility of constructing physical maps in large, complex, polyploid genomes with a chromosome-based approach. 相似文献
