The applicability of commercially available remote sensing instrumentation was evaluated for site-specific management of abiotic and biotic stress on cotton (Gossypium hirsutum L.) grown under a center pivot low energy precision application (LEPA) irrigation system. This study was conducted in a field where three irrigation regimes (100%, 75%, and 50% ETc) were imposed on areas of Phymatotrichum (root rot) with the specific objectives to (1) examine commercial remote sensing instrumentation for locating areas showing biotic and abiotic stress symptomology in a cotton field, (2) compare data obtained from commercial aerial infrared photography to that collected by infrared transducers (IRTs) mounted on a center pivot, (3) evaluate canopy temperature changes between irrigation regimes and their relationship to lint yield with IRTs and/or IR photography, and (4) explore the use of deficit irrigation and the use of crop coefficients for irrigation scheduling. Pivot-mounted IRTs and an IR camera were able to differentiate water stress among irrigation regimes. The IR camera distinguished between biotic (root rot) and abiotic (drought) stress with the assistance of groundtruthing. The 50% ETc regime had significantly higher canopy temperatures than the other two regimes, which was reflected in significantly lower lint yields when compared to the 75% and 100% ETc regimes. Deficit irrigation down to 75% ETc had no impact on lint yield, indicating that water savings were possible without reducing yield. 相似文献

This work studied the potential biotechnological applications of a naviculoid diatom (IMA053) and a green microalga (Tetraselmis marina IMA043) isolated from the North Adriatic Sea. Water, methanol, and dichloromethane (DCM) extracts were prepared from microalgae biomass and evaluated for total phenolic content (TPC) and in vitro antioxidant properties. Biomass was profiled for fatty acid methyl esters (FAME) composition. The DCM extracts had the highest levels of total phenolics, with values of 40.58 and 86.14 mg GAE/g dry weight (DW in IMA053 and IMA043, respectively). The DCM extracts had a higher radical scavenging activity (RSA) than the water and methanol ones, especially those from IMA043, with RSAs of 99.65% toward 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt (ABTS) at 10 mg/mL, and of 103.43% against 2,2-diphenyl-1-picrylhydrazyl (DPPH) at 5 mg/mL. The DCM extract of IMA053 displayed relevant copper chelating properties (67.48% at 10 mg/mL), while the highest iron chelating activity was observed in the water extract of the same species (92.05% at 10 mg/mL). Both strains presented a high proportion of saturated (SFA) and monounsaturated (MUFA) fatty acids. The results suggested that these microalgae could be further explored as sources of natural antioxidants for the pharmaceutical and food industry and as feedstock for biofuel production. 相似文献

Harmful algal blooms pose a challenge regarding food safety due to their erratic nature and forming circumstances which are yet to be disclosed. The best strategy to protect human consumers is through legislation and monitoring strategies. Global warming and anthropological intervention aided the migration and establishment of emerging toxin producers into Europe’s temperate waters, creating a new threat to human public health. The lack of information, standards, and reference materials delay effective solutions, being a matter of urgent resolution. In this work, the recent findings of the presence of emerging azaspiracids, spirolildes, pinnatoxins, gymnodimines, palitoxins, ciguatoxins, brevetoxins, and tetrodotoxins on European Coasts are addressed. Chlorophyll fluorescence spectral analysis permits detection, monitoring, and evaluation of abiotic stresses upon healthy plants using illumination of a light source in the UV?CVIS spectral range. This technique indirectly assesses the amount of physiological stress caused by photosynthetic damage, specifically damage to photosystem II, in plants. The objective of this study was to detect the toxicity of cadmium in maize plants via spectral analysis of chlorophyll fluorescence. The analysis is noninvasive and nondestructive and is used to follow the temporal evolution of changes in the chlorophyll content and physiological state of Zea mays L. seedlings under cadmium stress. Conventional techniques were also used to evaluate the dry matter production and Cd accumulation in plant leaves. Plants exhibited a notable reduction in dry matter production and chlorophyll levels with the administration of increasing doses of Cd in the nutrient solution. The fluorescence analysis was sensitive to changes caused by Cd in maize plants, detecting damage caused by different treatments before visual symptoms were observed. This technique has a practical application and produces rapid results that can be used in the evaluation of Cd-induced stress in plants and the detection of areas contaminated by this element. 相似文献

An inexpensive and portable surface plasmon resonance (SPR) sensor, SPReeta Evaluation Kit SPR3, has been used to develop a biosensor for the determination of fluoroquinolone antibiotics (FQs) and to demonstrate its performance analyzing FQ residues in milk samples. The SPReeta three-channel gold chips were activated with a mixed self-assembled monolayer (m-SAM) and functionalized with a FQ haptenized protein. Binding of the antibody produced a concentration-dependent increase of the SPR signal as a result of the change in the refraction index. Similarly, the presence of the FQ produced a dose-dependent decrease of the response, which allowed a good limit of detection (LOD) to be obtained (1.0 ± 0.4 μg L(-1) for enrofloxacin in buffer). The response was reproducible in all three channels, on different injections and days, and also between chips. Milk samples could be analyzed after a simple sample treatment involving fat removal by centrifugation and dilution with water. Under these conditions calibration curves were obtained showing that FQ residues can be analyzed in milk samples with an IC(50) value of 26.4 ± 7.2 μg L(-1) and a LOD of 2.0 ± 0.2 μg L(-1) (for enrofloxacin), far below the European Union regulations for this antibiotic family in this matrix. Finally, the paper also demonstrates that the biosensor is able to selectively detect the presence of FQs in milk samples, even in the presence of other antibiotics. The objective of this study was to examine the relationship between soil water potential, nitrifier community structure and nitrification activity in semiarid soils. Soils were collected after a 5-month dry period (end of summer) and subsequently rewetted to specific water potentials and incubated for 7 days prior to analysis of nitrification activity and nitrifier community structure. The approach used in this study targeted a 491bp segment of the amoA gene which encodes the active site of the ammonia monooxygenase enzyme, which is the key enzyme for all aerobic ammonia oxidisers. amoA serves as a useful target for environmental studies since it is both specific and universal for all ammonia oxidisers and reflects the phylogeny of the ammonia oxidisers. Our results suggest that in semiarid soils water potential plays a key role in determining the structure of ammonia oxidising bacteria (AOB), and that additionally AOB community structure is correlated to potential nitrification rate in these soils. 相似文献

The amino acid composition and the physicochemical and functional properties of quinoa protein isolates were evaluated. Protein isolates were prepared from quinoa seed by alkaline solubilization (at pH 9, called Q9, and at pH 11, called Q11) followed by isoelectric precipitation and spray drying. Q9 and Q11 had high levels of essential amino acids, with high levels of lysine. Both isolates showed similar patterns in native/SDS-PAGE and SEM. The pH effect on fluorescence measurements showed decreasing fluorescence intensity and a shift in the maximum of emission of both isolates. Q9 showed an endotherm with a denaturation temperature of 98.1 degrees C and a denaturation enthalpy of 12.7 J/g, while Q11 showed no endotherm. The protein solubility of Q11 was lower than that of Q9 at pH above 5.0 but similar at the pH range 3.0-4.0. The water holding capacity (WHC) was similar in both isolates and was not affected by pH. The water imbibing capacity (WIC) was double for Q11 (3.5 mL of water/g isolate). Analysis of DSC, fluorescence, and solubility data suggests that there is apparently denaturation due to pH. Some differences were found that could be attributed to the extreme pH treatments in protein isolates and the nature of quinoa proteins. Q9 and Q11 can be used as a valuable source of nutrition for infants and children. Q9 may be used as an ingredient in nutritive beverages, and Q11 may be used as an ingredient in sauces, sausages, and soups. 相似文献
