To determine whether phenolase activity is related to susceptibility or resistance to blackspot, tissue from bud and stem ends of potato tubers was assayed for differences in monophenolase and polyphenolase activities, changes in activity due to bruising, and different forms of phenolase. No relationship between blackspot and phenolase activity was found when activities of selected tissue were determined with p-cresol, tyrosine or chlorogenic acid as substrates. A significant positive relationship was found between polyphenolase activity and blackspot susceptibility when total or soluble or particulate fractions of phenolase activity were determined with catechol. Stem tissue had a significantly higher rate of phenolase activity than did bud-end tissue, where activity was determined with p-cresol, tyrosine or catechol; no significant differences in activity between the ends were found when chlorogenic acid was used.  相似文献

Three experiments were conducted to evaluate spray-dried blood cells (SDBC) and crystalline isoleucine in nursery pigs. In Exp. 1, 120 pigs were used to evaluate 0, 2, 4, and 6% SDBC (as-fed basis) in a sorghum-based diet. There were six replicates of each treatment and five pigs per pen, with treatments imposed at an initial BW of 9.3 kg and continued for 16 d. Increasing SDBC from 0 to 4% had no effect on ADG, ADFI, and G:F. Pigs fed the 6% SDBC diet had decreased ADG (P < 0.01) and G:F (P = 0.06) compared with pigs fed diets containing 0, 2, or 4% SDBC. In Exp. 2, 936 pigs were used to test diets containing 2.5 or 5% SDBC (as-fed basis) vs. two control diets. There were six replicates of each treatment at industry (20 pigs per pen) and university (six pigs per pen) locations. Treatments were imposed at an initial BW of 5.9 and 8.1 kg at the industry and the university locations, respectively, and continued for 16 d. Little effect on pig performance was noted by supplementing 2.5% SDBC, with or without crystalline Ile, in nursery diets. Pigs fed the 5% SDBC diet without crystalline Ile had decreased ADG (P < 0.01), ADFI (P < or = 0.10), and G:F (P < 0.05) compared with pigs fed the control diets. Supplementation of Ile restored ADG, ADFI, and G:F to levels that were not different from that of pigs fed the control diets. In Exp. 3, 1,050 pigs were used to test diets containing 5, 7.5, or 9% SDBC (as-fed basis) vs. a control diet. There were six replicates of each treatment at the industry (20 pigs per pen) location and five replicates at the university (six pigs per pen) locations. Treatments were imposed at an initial BW of 6.3 and 7.0 kg at the industry and university locations, respectively, and continued for 16 d. Supplementation of 5% SDBC without crystalline Ile decreased ADG and G:F (P < 0.01) compared with pigs fed the control diet, but addition of Ile increased ADG (P < 0.01) to a level not different from that of pigs fed the control diet. The decreased ADG, ADFI, and G:F noted in pigs fed the 7.5% SDBC diet was improved by addition of Ile (P < 0.01), such that ADG and ADFI did not differ from those of pigs fed the control diet. Pigs fed diets containing 9.5% SDBC exhibited decreased ADG, ADFI, and G:F (P < 0.01), all of which were improved by Ile addition (P < 0.01); however, ADG (P < 0.05) and G:F (P = 0.09) remained lower than for pigs fed the control diet. These data indicate that SDBC can be supplemented at relatively high levels to nursery diets, provided that Ile requirements are met.  相似文献

A peripheral vasodilatory agent, isoxsuprine hydro-chloride, was evaluated in a controlled study for its efficacy in the treatment of acute equine laminitis. Eight healthy, adulthorses of variable age and sex were used in the trial. Acute laminitis was induced in 5 of the horses by oral carbohydrate overload. Intravenous isoxsuprine therapy (1.8 mg/kg) was initiated in 3 of the horses receiving carbohydrate overload at first sign of clinical lameness and repeated at 12-hour intervals. Intravenous saline placebos were administered on a similar schedule to 2 control horses which also received a carbohydrate overload. The remaining 3 horses served as further controls. Local and systemic responses to induction of laminitis and isoxsuprine administration were assessed by subjective evaluation of clinical lameness in a double blind trial; nuclear scintigraphy and radiography of the distal forelimbs; and assessment of physical, hematological and biochemical parameters.

Pronounced tachycardia, hypotension and sweating accompanied the intravenous infusion of isoxsuprine. The 3 horses treated with isoxsuprine following the induction of laminitis showed a more rapid improvement in soundness than horses receiving saline placebos. No horse developed rotation of the third phalanx in response to the diet Nuclear scintigraphy indicated that blood perfusion patterns within the hoof of laminitic horses altered with isoxsuprine therapy, but an overall increase or decrease in perfusion was not apparent. Alterations in serum enzyme and electrolyte profiles with the onset of laminitis generally concurred with findings previously reported for this model of the disease. No change in coagulation profiles accompanied the onset of laminitis or isoxsuprine administration. Blood gas analysis indicated an increase in median palmar vein oxygen partial pressure (PO2) levels with onset of laminitis. A concurrent decrease in the median palmar arteriovenous oxygen partial pressure difference (AVO2) was significant at the P<0.01 level. There was no difference in median palmar vein PO2 values between thoseanimals receiving isoxsuprine and those receiving saline placebo therapy. Results of the trialindicated that isoxsuprine may be beneficial in the treatment of acute laminitis. Further controlled studies are appropriate.  相似文献

Historically many towns in inland Australia disposed of their treated sewage by pumping into local rivers. This is no longer a feasible proposition. Alternatives to river pumping include irrigation and/or aquaculture. As treated sewage effluent may contain large amounts of nitrogen, phosphorus and sodium salts, if not managed carefully, soil salinity, sodicity and nutrient accumulation could increase. The objective of this study was to evaluate if gypsum application had any effect on soil‐quality changes in a Vertisol due to irrigating a cotton–wheat rotation with tertiary treated sewage effluent. The treatments were application of 2·5 t ha−1 of gypsum in June 2000 before commencing irrigation and an untreated control. Annually, between June 2000 and April 2004, irrigation water quality and soil changes in nitrate‐N, EC1:5, pH, organic carbon, Cl, dispersion index, and exchangeable cations to a depth of 1·8 m were measured and deep drainage inferred with the chloride mass balance method. Cotton lint yield and fibre characteristics were also evaluated. Irrigation with treated sewage effluent increased exchangeable Na in all depths, and exchangeable Ca and K in the clayey‐textured surface 0·6 m, but decreased exchangeable Ca and K, and SOC in the coarser clay‐loam‐textured depths > 0·6 m. Nitrate‐N leaching, associated with deep drainage had occurred, as the crops had not used all the N in irrigation water. Gypsum application decreased exchangeable Ca, increased dispersion and during the 2003–2004 season deep drainage, but had no effect on salinity, sodicity or pH. Application of commercial gypsum at sub‐optimal rates with sodium‐rich irrigation water is, therefore, unlikely to improve soil properties. Stubble incorporation before sowing cotton in 2003 appears to have mobilized gypsum applied during 2000. Gypsum application reduced cotton lint yield and fibre quality during 2003–2004. Transgenic glyphosate-resistant (GR) soybean [Glycine max (L.) Merr.] has enabled highly effective and economical weed control. The concomitant increased application of glyphosate could lead to shifts in the soil microbial community. The objective of these experiments was to evaluate the effects of glyphosate on soil microbial community structure, function and activity. Field assessments on soil microbial communities were conducted on a silt loam soil near Stoneville, MS, USA. Surface soil was collected at time of planting, before initial glyphosate application and 14 days after two post-emergence glyphosate applications. Microbial community fatty acid methyl esters (FAMEs) were analyzed from these soil samples and soybean rhizospheres. Principal component analysis of the total FAME profile revealed no differentiation between field treatments, although the relative abundance of several individual fatty acids differed significantly. There was no significant herbicide effect in bulk soil or rhizosphere soils. Collectively, these findings indicate that glyphosate caused no meaningful whole microbial community shifts in this time period, even when applied at greater than label rates. Laboratory experiments, including up to threefold label rates of glyphosate, resulted in up to a 19% reduction in soil hydrolytic activity and small, brief (<7 days) changes in the soil microbial community. After incubation for 42 days, 32-37% of the applied glyphosate was mineralized when applied at threefold field rates, with about 9% forming bound residues. These results indicate that glyphosate has only small and transient effects on the soil microbial community, even when applied at greater than field rates.  相似文献
